Abstract
Description
本出願は、2015年5月1日に出願された米国仮出願第62/155,967号及び2015年11月13日に出願された米国仮出願第62/254,992号の利益を主張する。これらの各々の内容は、その全体が参照により本明細書に組み込まれる。
本発明のVPBPが細菌によって誘導される赤血球細胞(RBC)の溶血を阻止する能力は、本分野で知られている多数のプロトコルによって評価することができる。例えばヒトRBCをPBSの中で洗浄し、DMEMの中に2%(v/v)で再懸濁させた。Pseudomonas aeruginosa菌と系列希釈した抗体を96ウエル丸底プレートの中のRBCに添加した。プレートを37℃で2時間インキュベートした後、4℃で2時間インキュベートした。次にプレートの回転によって完全なRBCをペレットにした後、上清を平底96ウエルプレートに移して放出されたヘモグロビンを分光測光器で観察した。
本発明のVPBPが細菌によって誘導される哺乳動物細胞の細胞毒性を阻止する能力は、本分野で知られている多数のプロトコルによって評価することができる。例えばA549(肺上皮)細胞の集密的単層を96ウエルプレートの中で増殖させた。細胞をCalcein AMに装填した後、洗浄して過剰なCalceinを除去した。Pseudomonas aeruginosaとさまざまな濃度の抗体をA549細胞に添加し、37℃で2時間インキュベートした。次に単層を洗浄した後、残った細胞を蛍光によって定量した。
本発明のVPBPが細菌感染から保護する能力は、Pseudomonas aeruginosa感染のマウスモデルを用いて評価することができる。マウスにPseudomonas aeruginosaを感染させる24時間前にPcrV抗体であらかじめ処理した。t=0の時点でマウスにPseudomonas aeruginosaを気管内感染させ、生存を4日間監視した。重要なことだが、多重特異性多価VPBP含有融合タンパク質は、単一特異性多価VPBP含有融合タンパク質と比べて実質的により高い保護を与えることが見いだされた(図4A〜図4C)。本発明の多重特異性多価VPBP含有融合タンパク質は、抗PCRV抗体であるV2L2よりも実質的に強力でもある。なおV2L2は、本分野において、Pseudomonas aeruginosaが誘導する溶血の強力な阻止剤として知られている(例えばWO 2013/0170565として公開されたPCT/US2012/063639を参照のこと)。
OprI標的抗体がPseudomonas株に結合する能力は、細胞全体細菌ELISAによって評価することができる。細菌培養物を標準的な細菌培地の中で半対数相まで増殖させた後、PBSの中で洗浄して再懸濁させた。等しい体積の細菌懸濁液を96ウエルプレートに入れて37℃で24時間インキュベートした。プレートをBSAでブロックした後、抗体の系列希釈液を添加した。その後洗浄し、HRPが共役した抗ヒトFc特異性二次抗体を添加した。インキュベーションと洗浄の後、TMB基質を添加し、吸光度を600 nmで測定して細菌に対する抗体の結合を検出した。図5に示したように、OprI抗体は、調べたすべてのPseudomonas aeruginosa株、並びにPseudomonas putidaに結合することが見いだされた。
本明細書に提示する研究により、本発明のVPBPが細菌感染から保護する能力をPseudomonas aeruginosa感染のマウスモデルを用いて評価できることを示す。これらの研究では、PcrVと外側膜タンパク質I(「OprI」)の両方に結合するVPBPを特に利用する。それを本明細書では、「PcrV×OprI二重特異性融合タンパク質」、及び/又は「PcrV×OprI融合タンパク質」、及び/又は「PcrV×OprI融合体」と呼ぶ。PcrVとOprIを二重に標的とすることで、その融合ポリペプチドが細菌細胞表面に係留又は付着及び/又は結合することが可能になる。そして本明細書に提示する研究により、この二重標的が生体内でも保護を増大させることを示す。

- 前記グラム陰性細菌のIII型分泌装置の前記少なくとも1つのVチップタンパク質は、Pseudomonas aeruginosaに由来するPcrVであり、かつ前記グラム陰性細菌のIII型分泌装置の前記少なくとも1つの非Vチップタンパク質は、Pseudomonas aeruginosaに由来するOprIである、請求項35に記載の単離されたポリペプチド。
- 前記単離されたポリペプチドは、配列番号10〜44、71〜74、及び81〜84からなる群から選択されるアミノ酸配列を含む、請求項1〜36のいずれか1項に記載の単離されたポリペプチド。
