Abstract
Description
一実施形態では、本発明は、魚にその食餌サイクルにわたって水産養殖試料組成物を与えることにより、水産養殖魚肉製品を持続的に生産する方法であって、組成物中の魚油のすべてまたは一部をエイコサペンタエン酸(「EPA」)およびドコサヘキサエン酸(「DHA」)の単一の微生物源と置き換えることにより、水産養殖試料組成物を配合するステップを含む方法に関する。
本開示では、いくつかの用語および略語が使用される。次の定義が提供される:
「多価不飽和脂肪酸」は、「PUFA」と省略される。
「トリアシルグリセロール」は、「TAG」と省略される。
「総脂肪酸」は、「TFA」と省略される。
「脂肪酸メチルエステル」は、「FAME」と省略される。
「乾燥細胞重量」は、「DCW」と省略される。
− 少なくとも40%w/wDHAおよびEPA、好ましくは約50%w/wDHAおよびEPA、
− 約0.2:1〜1:1、好ましくは0.4:1〜0.8:1のEPA:DHA比、および
− 安定性を提供するために添加される少なくとも1種の酸化防止剤
を含有する、シゾキトリウム(Schizochytrium)由来の油である
ATCC寄託番号PTA−10212で寄託されている単離された微生物を、下に記載する通りに個々の発酵運転における成長特性について試験した。典型的な培地および培養条件を表3に示す。
pH6.5〜9.5、約6.5〜約8.0、または約6.8〜約7.8;
温度:摂氏15〜30度、摂氏約18〜約28度、または摂氏約21〜約23度;
溶存酸素:0.1〜約100%飽和、約5〜約50%飽和、または約10〜約30%飽和;および/または
以下に制御されるグリセロール:5〜約50g/L、約10〜約40g/L、または約15〜約35g/L。
最初の体重およそ200gのタイセイヨウサケを、1タンクあたり50匹の魚で1.5mタンクにランダムに分配した。水の温度は10℃の範囲である。魚は、実験食餌を与えるのを開始する前に2週間、制御食餌に適応させる。
Claims (26)
- 魚にその食餌サイクルにわたって水産養殖試料組成物を与えることにより、水産養殖魚肉製品を持続的に生産する方法であって、前記組成物中の魚油のすべてまたは一部をエイコサペンタエン酸(「EPA」)およびドコサヘキサエン酸(「DHA」)の単一の微生物源と置き換えることにより、水産養殖試料組成物を配合するステップを含む方法。
- 前記水産養殖試料組成物が、前記水産養殖試料組成物に対する重量パーセントとして測定されて少なくとも約0.8%である総量のEPAおよびDHAを含む、請求項1に記載の方法。
- EPAの濃度対DHAの濃度の比が、前記微生物源中または前記水産養殖試料組成物中のEPAおよびDHAの個々の濃度に基づいて少なくとも2:1である、請求項1または2に記載の方法。
- EPAの濃度対DHAの濃度の比が、前記微生物源中または前記水産養殖試料組成物中のEPAおよびDHAの個々の濃度に基づいて1:1以下である、請求項1または2に記載の方法。
- 前記微生物源が微生物油であり、前記微生物油が、いずれも前記微生物源から得られるバイオマス、処理されたバイオマス、部分的に精製された油、および精製された油からなる群から選択される形態で提供される、請求項1〜4のいずれか一項に記載の方法。
- 前記微生物源が得られる微生物が藻類、菌類、または酵母である、請求項1〜5のいずれか一項に記載の方法。
- 前記微生物がシゾキトリウム(Schizochytrium)またはトラウストキトリウム(Thraustochytrium)属のメンバーである、請求項6に記載の方法。
- 前記微生物が、ATCC寄託番号PTA−10208、またはPTA−10209、またはPTA−10210、またはPTA−10211、またはPTA−10212、またはPTA−10213、またはPTA−10214、またはPTA−10215で寄託されている種の特性を有する、請求項7に記載の方法。
- 前記微生物が突然変異株である、請求項6〜8のいずれか一項に記載の方法。
- 前記微生物が、EPAおよびDHAを含む微生物油を含有する多価不飽和脂肪酸の生成のために遺伝子操作されたトランスジェニック微細生物である、請求項6〜8のいずれか一項に記載の方法。
- 前記水産養殖魚肉製品が、EPAの濃度対DHAの濃度の比であって、前記水産養殖魚肉製品中のそれぞれの前記濃度に基づいて2:1以上であるEPAの濃度対DHAの濃度の比を有する、請求項1〜10のいずれか一項に記載の方法。
- 前記水産養殖魚肉製品が、EPAの濃度対DHAの濃度の比であって、前記水産養殖魚肉製品中のそれぞれの前記濃度に基づいて1:1以下であるEPAの濃度対DHAの濃度の比を有する、請求項1〜10のいずれか一項に記載の方法。
- 単一の微生物源から得られるエイコサペンタエン酸(「EPA」)およびドコサヘキサエン酸(「DHA」)を含む、水産養殖試料のための試料添加組成物。
- いずれも前記微生物源から得られるバイオマス、処理されたバイオマス、部分的に精製された油、および精製された油からなる群から選択される形態で提供される、請求項13に記載の添加組成物。
- 前記微生物源が得られる微生物が藻類、菌類、または酵母である、請求項13または14に記載の添加組成物。
- 前記微生物がシゾキトリウム(Schizochytrium)またはトラウストキトリウム(Thraustochytrium)属のメンバーである、請求項15に記載の添加組成物。
- 前記微生物が、ATCC寄託番号PTA−10208、またはPTA−10209、またはPTA−10210、またはPTA−10211、またはPTA−10212、またはPTA−10213、またはPTA−10214、またはPTA−10215で寄託されている種の特性を有する、請求項16に記載の添加組成物。
- 少なくとも40%w/wのDHAおよびEPA、好ましくは約50%w/wのDHAおよびEPAを含有する精製された微生物油形態である、請求項13〜17のいずれか一項に記載の添加組成物。
