Abstract
Description
本出願は、2014年1月31日に出願された米国特許仮出願第61/934,619号及び2014年3月27日に出願された米国特許仮出願第61/971,009号の利益を主張する。これらの出願のそれぞれの内容を、それらの全体として参照により本明細書中に援用する。
本発明は概して、マトリプターゼ及びu‐プラスミノーゲン活性化因子(uPA)から選択される少なくとも1つのプロテアーゼのための基質である切断可能部分を含んでいるポリペプチド、マトリプターゼ及びuPAから選択される少なくとも1つのプロテアーゼのための基質である切断可能部分を含んでいる活性化可能抗体及び他の大型分子、並びにマトリプターゼ及びuPAから選択される少なくとも1つのプロテアーゼのための基質である切断可能部分を含んでいるこれらのポリペプチドの作製方法、及びさまざまな治療適応、診断適応及び予防適応における使用方法に関する。

医薬組成物
試薬と菌株:ストレプトアビジン複合化フィコエリトリン(SA‐PE)(Invitrogen, Life Technologies)を修飾せずに使用した。ヒトマトリプターゼ‐1(Research & Diagnostics Systems, Inc.)を修飾せずに使用した。ヒトプラスミン(Haematologic Technologies Inc.)を修飾せずに使用した。ヒトtPA(Molecular Innovations)を修飾せずに使用した。Mona(単球のアダプター)のSH3ドメインに融合させたYPetを修飾せずに使用した。TBST(50mMのTris HCl、150mMのNaCl、0.05%のTween20、pH7.4)を使用した。E.コリ(E. coli)MC1061(Casadaban et al., JMB 138(2):179-207 (1980))を使用した。すべての細菌増殖を、別の抗生物質を指定しない限り34μg/mLのクロラムフェニコールを補ったLB培地(LB)中で激しく振盪しながら、37℃にて実施した。
細胞全体におけるマルチコピー基質ディスプレイの使用はマトリプターゼ‐1によって切断される基質集団の選択を可能にした。組み換えヒトマトリプターゼ‐1を使用した選択を、2010年2月23日に交付された米国特許第7,666,817B号に記載のとおり実施した。(プラットフォームeCLiPS3.0‐NSUB_SPを使用した)基質部位に隣接する領域のバックグラウンド加水分解を、各反応条件下で計測して、指定された基質領域において起こる加水分解を保証した。最終的なプールを、マトリプターゼ‐1、プラスミン、及びtPAに対して試験した。該プールは、マトリプターゼ‐1によって切断されたが、tPA又はプラスミンによって切断されなかった。図1は、TBST中でのマトリプターゼ‐1によるプールSMP30の切断を示し、図2は、共にTBST中、プールSMP17に関する50nMのマトリプターゼ‐1による切断、及びtPAに対する抵抗性を示す。同様の技術を使用して、別のライブラリをスクリーニングして、マトリプターゼ‐1とu‐プラスミノーゲン活性化因子の両方で切断されるが、tPA又はプラスミンによって切断されない基質を選択した。
細胞全体におけるマルチコピー基質ディスプレイの使用は切断反応速度の簡単で直接的な定量的特徴づけを可能にした。その結果、フローサイトメトリーを、基質変換に基づいて個々の単離クローンを格付けするのに使用し、そしてクローンをDNA配列決定によって同定した。このようにして、各クローンに関する変換の程度を、いくつかの異なったプロテアーゼ濃度にて測定し、そして、ミカエリス‐メンテンモデル(反応速度データ分析の項)に当てはめることができる。観察した二次反応速度定数(kcat/KM)を各基質対マトリプターゼ‐1に関して決定した。(プラットフォームeCLiPS3.0‐NSUB_SPを使用した)基質に隣接する領域のバックグラウンド加水分解を各反応条件下で計測して、指定された基質領域において起こる加水分解を保証した。例えば、図3は、TBST中、マトリプターゼ‐1によるアミノ酸配列VAGRSMRPを含む基質の切断を示す。
この実施例は、本開示の基質に関して、それらが活性化可能抗体内に組み込まれた場合のインビトロにおける活性を実証する。
マトリプターゼによる切断のために選択される基質を含んでいる得られた活性化可能抗体は、マトリプターゼに関して約6,500〜100,000M−1s−1の範囲のkcat/KM値を有した(試験した5つの基質)。
この実施例は、本開示の基質に関して、それらが活性化可能抗体に組み込まれて、マウスに注入された場合のインビボにおける安定性を実証する。
試薬と菌株:ヒトuPA(カタログ番号1310‐SE、Research & Diagnostics Systems, Inc.)を修飾せずに使用した。ヒトマトリプターゼ‐1(カタログ番号3946‐SE、Research & Diagnostics Systems, Inc.)を修飾せずに使用した。ヒトtPA(カタログ番号HTPA‐TC、Molecular Innovations)を修飾せずに使用した。ヒトプラスミン(カタログ番号HCPM‐0140、Haematologic Technologies Inc.)を修飾せずに使用した。抗EEモノクローナル抗体(Covance, Princeton, NJ)をAlexa647(Life Sciences)で標識し、他に修飾せずに使用した(EE647と名付けた)。
E.コリMC1061又はMC1061誘導菌株(DH10β)をすべての実験に使用した(Casadaban et al., JMB 138(2):179-207 (1980))。すべての細菌増殖を、別の抗生物質を指定しない限り、34μg/mLのクロラムフェニコール(cm)を補ったLB培地(LB)中で激しく振盪しながら37℃にて実施した。
