事象特異的Taqmanアッセイは、トウモロコシ事象DAS−40278−9の存在を検出するため、及び育種集団における植物の接合生殖性状態を決定するために開発した。事象特異的アッセイを開発するために、特異的Taqmanプライマー及びプローブは、5’挿入−植物接合部に位置するDNA配列に従って設計した。DAS−40278−9の特異的検出のために、この5’−統合接合部に渡る73bpのDNAフラグメントは、2つの特異的プライマーを使用して増幅させた。このPCR産物の増幅は、その5’末端でFAMレポーターを含有するApplied Biosystems社によって合成された標的特異的MGBプローブによって測定した。AAD−1コーン事象DAS−40278−9に対するこのTaqman検出法の特異性は、16種の相違するAAD−1コーン事象及び非トランスジェニックコーン品種に対してコーン特異的内因性参照遺伝子であるインベルターゼを用いた二重フォーマットにおいて試験した。
この試験では16種の相違するAAD−1コーン事象及び非トランスジェニックコーン品種のgDNAサンプルについて試験した。gDNAは、2つのアプローチ、Qiagenキット又はCTABを用いて抽出した。Qiagenキットを用いて抽出したgDNAサンプルに対しては、修正Qiagen DNeasy 96植物キットプロトコールに従ってgDNA抽出のために8枚のコーン新鮮葉片を使用した。CTAB手順を使用することによって抽出したgDNAサンプルに対しては、約0.3gの凍結乾燥葉組織をPermingeat et al., 1998からのプロトコールに従って使用した。gDNAは、販売業者の取扱説明書(Molecular Probes社、オレゴン州ユージーン)に従ってPico Green法を用いて定量した。gDNAサンプルは、この試験のためにDNase無含有水で希釈して10ng/μLの濃度を生じさせた。
特異的Taqmanプライマー及びプローブは、DAS−40278−9事象特異的Taqmanアッセイのために設計した。これらの試薬は、AAD−1コーン事象DAS−40278−9を検出するために下記に列挙した条件を用いて使用できる。表1は、事象DAS−40278−9を検出するために特異的に開発されたプライマー及びプローブ配列を列挙している。
