Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.改善された複合骨移植片の調製方法であって、以下の工程: (a)骨髄吸引物懸濁液を提供する工程;および (b)該骨髄吸引物懸濁液を、多孔性で生体適合性の移植可能な基体を通して 通過させる工程を包含する、方法。 2.前記骨髄吸引物懸濁液が抗凝固剤を含む、請求項1に記載の方法。 3.前記基体を通った前記骨髄吸引物懸濁液の溶出液を再利用する工程をさらに 包含する、請求項2に記載の方法。 4.前記骨髄吸引物懸濁液が前記基体を通って通過した後に該基体を洗浄する工 程をさらに包含する、請求項2に記載の方法。 5.前記溶出液が前記基体を通して再利用された後に該基体を洗浄する工程をさ らに包含する、請求項3に記載の方法。 6.前記基体が、リン酸カルシウムを含む合成セラミック、鉱化骨、鉱物質除去 骨、およびコラーゲンからなる群から選択される移植片材料を含む、請求項1に 記載の方法。 7.前記基体が外部寸法を有し、そして同じ外部寸法を有する固体物体よりも少 なくとも5倍大きい総表面積を有する、請求項1に記載の方法。 8.増殖因子を含む溶液を前記基体を通して通過させる工程をさらに包含する、 請求項1に記載の方法。 9.前記基体が、その表面に結合した細胞接着分子を含む、請求項7に記載の方 法。 10.工程(a)の前に等張液を前記骨髄吸引物に添加して、約1000万の有核細 胞/ml〜約3億有核細胞/mlの範囲の濃度を有する骨髄吸引物液懸濁液を提供す る、請求項1に記載の方法。 11.前記基体を通しての前記骨髄吸引物懸濁液の直線的溶出速度が、約2mm/ 分〜約500mm/分である、請求項1に記載の方法。 12.複合骨髄移植片であって、以下: (a)多孔性で生体適合性の移植可能な基体; (b)有核骨髄細胞の不均一集団;および (c)結合組織前駆細胞の富化集団、 を含む、複合骨髄移植片。 13.骨髄吸引物懸濁液から複合骨髄移植片を調製するためのキットであって、 以下: (a)多孔性で生体適合性の移植可能な基体;および (b)該基体を保持するための容器であって、該容器は該基体を保持し、そし てそこを通しての骨髄吸引物懸濁液の流れを可能にするように形成され、該容器 が2つの端を有し、該端の各々が開口部を規定する、容器、 を含む、キット。 14.前記基体を通しての前記骨髄吸引物懸濁液の流速を調節するために、前記 容器の1端に取り付け可能な液流調節器をさらに含む、請求項12に記載のキッ ト。 15.さらに、 (a)前記骨髄吸引物懸濁液を保持するためのレザーバー;および (b)該リザーバーから前記容器への該骨髄吸引物懸濁液の流れを調節するた めに、該リザーバーに取り付け可能な液流調節器、 を含む、請求項12に記載のキット。 16.前記容器からの前記骨髄吸引物懸濁液の溶出液を受けるための溶出液レシ ーバーをさらに含む、請求項12に記載のキット。 17.前記基体が、外部寸法を有し、そして同じ外部寸法を有する固体物体表面 積よりも少なくとも5倍大きい総表面積を有する、請求項12に記載のキット。 18.前記基体が、リン酸カルシウムを含むセラミック、コラーゲン、鉱化骨、 鉱物質除去骨からなる群から選択される材料から形成される、請求項12に記載 のキット。 19.前記基体が、その表面に化学結合した細胞接着分子を含む、請求項15に 記載のキット。 20.骨髄細胞の単離集団中の結合組織前駆細胞の濃度を増加させるための方法 であって、以下: (a)骨髄吸引物懸濁液を、多孔性で生体適合性の移植可能な基体を通して通 過させ、そこに化学結合した有核骨髄細胞を有するマトリックスを提供する工程 ; (b)該有核骨髄細胞を該マトリックスから解離させる工程;および (c)該解離した骨髄細胞を収集する工程、 を包含する、方法。
