Claims (1)
- 【特許請求の範囲】 1.ヒト間葉前駆体細胞の試験管内軟骨形成およびそれからのヒト軟骨細胞の試 験管内形成のための一つの組成物であって、この組成物は三次元形態での分離さ れたヒト間葉幹細胞およびそれと接触する少くとも1個の軟骨誘導剤を含む組成 物。 2.請求の範囲第1項記載の組成物であって、ここで間葉幹細胞が分離され培養 拡張されたヒト間葉幹細胞である組成物。 3.請求の範囲第1項記載の組成物であって、ここで間葉幹細胞が合成無血清環 境にある組成物。 4.請求の範囲第1項記載の組成物であって、ここで間葉幹細胞がごく近接して 濃縮される組成物。 5.請求の範囲第4項記載の組成物であって、ここで間葉幹細胞が充填細胞ある いは遠心分離細胞ペレットとして存在する組成物。 6.請求の範囲第1項記載の組成物であって、ここで軟骨誘導剤が(i)グルコ コルチコイド;(ii)トランスフォーミング成長因子−β上科の一員;(iii) コラーゲン性細胞外基質の成分;および(iv)ビタミンA類似体よりなるグルー プから選択される組成物。 7.請求の範囲第6項記載の軟骨誘導剤であって、ここでグルココルチコイドが デキサメタゾンである軟骨誘導剤。 8.請求の範囲第6項記載の軟骨誘導剤であって、ここでトランスフォーミング 成長因子−β上科の一員が骨形態発生タンパク質(望ましくはBMP−4)、T GF−β1、インヒビンAあるいは軟骨形成刺激活性因子などである軟骨誘導剤 。 9.請求の範囲第8項記載の軟骨誘導剤であって、ここで骨形態発生タンパク質 がBMP−4である軟骨誘導剤。 10.請求の範囲第6項記載の軟骨誘導剤であって、ここでコラーゲン性細胞外 基質の成分がコラーゲンIである軟骨誘導剤。 11.請求の範囲第10項記載の軟骨誘導剤であって、ここでコラーゲンIがゲ ルの形態にある軟骨誘導剤。 12.請求の範囲第6項記載の軟骨誘導剤であって、ここでビタミンA類似体が レチノイン酸である軟骨誘導剤。 13.請求の範囲第1項記載の組成物であって、ここで軟骨誘導剤がデキサメタ ゾンおよびTGF−β1の組合せである組成物。 14.間葉幹細胞を試験管内で軟骨誘導剤と接触させることにより間葉幹細胞か ら軟骨細胞を生産する一つの製法であって、ここで幹細胞が三次元形態に結合さ れている製法。 15.請求の範囲第14項記載の製法であって、ここで間葉幹細胞が分離され培 養拡張されたヒト間葉幹細胞である製法。 16.請求の範囲第14項記載の製法であって、ここで間葉幹細胞が合成無血清 環境にある製法。 17.請求の範囲第14項記載の製法であって、ここで間葉幹細胞がごく近接し て濃縮される製法。 18.請求の範囲第17項記載の製法であって、ここで間葉幹細胞が充填細胞あ るいは遠心分離細胞ペレットとして存在する製法。 19.請求の範囲第14項記載の製法であって、ここで軟骨誘 導剤が(i)グルココルチコイド;(ii)トランスフォーミング成長因子−β上 科の一員;(iii)コラーゲン性細胞外基質の成分;および(iv)ビタミンA類 似体よりなるグループから選択される製法。 20.請求の範囲第14項記載の製法であって、ここで軟骨誘導剤がデキサメタ ゾンおよびTGF−β1の組合せである製法。 21.請求の範囲第14項記載の製法であって、ここで接触の段階がヒト間葉幹 細胞のペレットを合成無血清培地で培養することを含む製法。 22.請求の範囲第21項記載の製法であって、ここで合成無血清培地が(1) 合成最少必須培地;(2)アスコルビン酸塩あるいはその類似体;(3)鉄源; (4)インスリンあるいはインスリン状成長因子;および(5)少くとも1個の 軟骨誘導剤あるいは因子を含む製法。 23.請求の範囲第14項記載の方法であって、ここで細胞が軟骨誘導組成物と 共に培養されその後硬質多孔性容器に置かれる方法。 24.請求の範囲第23項記載の方法であって、ここで硬質多孔性容器がセラミ ックキューブである方法。 25.間葉幹細胞を軟骨誘導剤に接触させることにより間葉幹細胞に軟骨形成を 誘導する一つの製法であって、ここで幹細胞が三次元形態に連結している方法。 26.請求の範囲第25項記載の製法であって、ここで間葉幹細胞が分離され培 養拡張されるヒト間葉幹細胞である製法。 27.請求の範囲第25項記載の製法であって、ここで間葉幹細胞が合成無血清 環境にある製法。 28.請求の範囲第25項記載の製法であって、ここで間葉幹細胞がごく近接し て濃縮される製法。 29.請求の範囲第25項記載の製法であって、ここで間葉幹細胞が充填細胞あ るいは遠心分離細胞ペレットとして存在する製法。 30.請求の範囲第25項記載の製法であって、ここで軟骨誘導剤が(i)グル ココルチコイド;(ii)トランスフォーミング成長因子−β上科の一員;(iii )コラーゲン性細胞外基質の成分;および(iv)ビタミンA類似体よりなるグル ープから選択される製法。 31.請求の範囲第25項記載の製法であって、ここで軟骨誘導剤がデキサメタ ゾンおよびTGF−β1の組合せである製法。 32.請求の範囲第25項記載の製法であって、ここで接触の段階がヒト間葉幹 細胞のペレットを合成無血清培地で培養することを含む製法。 33.請求の範囲第32項記載の製法であって、ここで合成無血清培地が(1) 合成最少必須培地;(2)アスコルビン酸塩あるいはその類似体;(3)鉄源; (4)インスリンあるいはインスリン状成長因子;および(5)少くとも1個の 軟骨誘導剤あるいは因子を含む製法。 34.請求の範囲第25項記載の方法であって、ここで細胞が軟骨誘導組成物で 培養されその後硬質多孔性容器に置かれる方 法。 35.請求の範囲第34項記載の方法であって、ここで硬質多孔性容器がセラミ ックキューブである方法。
