Abstract

1317034 Substituted 1-phenyl ethers of 3-[(4- phenyl alkyl) piperazin-1-yl] propan - 2 - ol and derivatives thereof PFIZER Ltd 5 Nov 1971 [6 Nov 1970] 52855/70 Heading C2C Novel compounds of the Formula I wherein R is acylamino, R<SP>1</SP> is H, halogen or C 1-6 alkyl, R<SP>2</SP> is H or C 1-6 alkyl; R<SP>3</SP> is H, halogen, C 1-6 alkyl or alkoxy; Alk is C 2-4 saturated aliphatic hydrocarbon radical, the two free valencies being separated by a chain of at least 2 carbon atoms; and X is oxygen or sulphur, the esters of compounds in which R<SP>2</SP> is hydrogen, and N-oxides and pharmaceutically acceptable salts thereof may be prepared (a) where R<SP>2</SP> is H, by reacting an epoxide with an N-substituted piperazine III (b) reacting a chlorohydrin or ether thereof IV with a compound III, (c) reacting a compound II or IV with piperazine to yield V and reacting the product with an appropriately substituted phenyl alkyl halide (or compounds wherein halide is replaced by a suitable leaving group, (d) acylation of compounds I in which R is amino, (e) alkylation or esterification of compounds I in which R<SP>2</SP> is hydrogen, (f) N- oxide formation. Pharmaceutical compositions of the compounds I show antihypertensive activity when administered orally or parenterally with the usual excipients.
