发明详述
本发明是基于编码位于奈瑟菌属的细胞表面的肽的基因的发现,因此其对于治疗感染的治疗试剂的制备非常有用。应当理解治疗也包括预防性治疗,例如接种。另外,尽管本发明的产物主要用于人类病人的感染,但认为兽医上的应用也在本发明的范围之内。
参考脑膜炎奈瑟球菌描述本发明。但是所有的奈瑟球菌菌株以及许多其它革兰氏阴性细菌菌株也可能包括与此处确定的蛋白质或肽具有同一或相似的氨基酸序列的蛋白质或肽。可能含有所述肽的生物体包括但不限于沙门氏菌属(Salmonella)、肠杆菌属(Enterobacter)、克雷白菌属(Klebsiella)、志贺菌属(Shigella)和耶尔森菌属(Yersinia)。
优选地,可能在本发明的各方面有用的肽与此处所确定的肽具有40%以上的相似性。更优选地,所述肽具有60%以上的序列相似性。最优选地,所述肽具有80%以上的序列相似性,例如95%的相似性。关于此处确定的多核苷酸序列,在本发明的各方面有用的有关多核苷酸与此处确定的序列具有40%以上的同一性。更优选地,所述多核苷酸序列具有60%以上的序列同一性。最优选地,所述多核苷酸序列具有80%以上的序列同一性,例如95%的同一性。
术语“相似性”和“同一性”本领域已知。使用术语“同一性”指基于在被比较的序列的相应的相同位置之间的同一性匹配的序列比对。术语“相似性”指氨基酸序列之间的比对,不仅考虑相应位置的相同的氨基酸,并考虑相应位置功能相似的氨基酸。因此,多肽序列之间的相似性指序列上相似外,还指功能上相似。
可以利用已知的方法计算基因序列之间同一性水平和氨基酸序列之间同一性或相似性水平。关于本发明,用于确定同一性和相似性的公开可用的基于计算机的方法包括BLASTP、BALSTN和FASTA(Atschul等人,J.Molec.Biol.,1990;215:403-410),BLASTX程序可获自NCBI,Gap程序获自Genetics Computer Group,Madison WI。利用Gap程序获得此处提供的相似性和同一性水平,利用Gap罚分为12和Gap长度罚分为4来确定氨基酸序列比对,用Gap罚分为50和Gap长度罚分为3来确定多核苷酸序列比对。
对本发明的基因性质鉴定以后,就可能利用该基因序列在其它微生物中检索相关的基因。这可能通过检索现有的数据库(例如EMBL或GenBank)来进行。
此处所述的技术为本领域熟知。但是,特别涉及Sambrook等人,Molecular Cloning,A Laboratory Manual(1989)以及Ausubel等人,current Protocols in Molecular Biology(1995),JohnWiley&Sons,Inc.
可以利用本领域已知的方法纯化和分离本发明的肽或蛋白质。具体地,确定了基因序列以后,就可能利用重组技术在合适的宿主中表达该基因。可以确定活性片段和有关的分子,并可能用于治疗。例如,肽或它们的活性片段可用作疫苗中的抗原性决定簇,以激发免疫应答。它们也可用于抗体制备,用于被动免疫或诊断应用。合适的抗体包括单克隆抗体或其片段,包括单链Fv片段。人源化的抗体也在本发明的范围内。抗体的制备方法对本领域技术人员是显而易见的。
肽的活性片段是指保持肽的生物学功能的片段。例如,用于激发免疫应答时,片段应具有足够的大小,使该片段产生的抗体能区别该肽和细菌微生物上的其它肽。通常,片段大小为至少30个核苷酸(10个氨基酸),优选60个核苷酸(20个氨基酸),最优选大于90个核苷酸(30个氨基酸)。
应当理解,除了其它微生物的相关分子外,本发明包括对此处确定的肽和多核苷酸所作的不明显改变生物学功能的修饰。遗传密码子的简并性可以由此处所述的多核苷酸产生具有小的碱基改变、但编码相同的肽的多核苷酸,这对本领域技术人员是显而易见的。本发明也包括互补的多核苷酸。也包括氨基酸水平的保守性置换,即可以置换不同的酸性或碱性氨基酸而基本不丧失功能。
本领域技术人员已知基于确定的肽制备疫苗。可以用合适的载体或佐剂配制疫苗组合物(例如明矾,必须或希望的),以提供针对感染的有效的免疫。疫苗制剂的制备对技术人员来说是显而易见的。
比较普遍地,并且本领域技术人员熟知,为了治疗应用,可以选择合适量的本发明活性组分以及合适的载体或赋型剂和给药途径。可以根据已知的标准(例如欲治疗的疾病的性质/严重程度、受试者的类型和/或健康等)选择或决定这些因素。
在单独的实施方案中,本发明的产物可能用于确定潜在抗微生物药物或检测毒力的筛选分析中。本领域技术人员知道常规的筛选分析,并可以利用本发明的产物进行适当方式的改动。例如,本发明的产物可能用作潜在药物的靶标,药物失活或结合靶标的能力表明其潜在的抗微生物活性。
本发明的基因也可能与微生物的毒力有关,因此缺失或失活该基因可能有效产生减毒的(无毒的)微生物。
可制备减毒的微生物具有破坏此处确定的任何基因的表达的突变。技术人员应知道破坏特定基因表达的方法。可能使用的技术包括插入失活或基因缺失技术。本发明减毒的微生物也可能包括其它在基因上的另外的突变,例如,在此处鉴定的另一种基因上,或在微生物生长所需的单独的基因上,例如,aro突变,或对于沙门菌属来说,在位于WO-A-96/17951确定的SP12区的基因上。
减毒的微生物也可能用作载体系统,用于传递异源抗原、治疗性蛋白质或核酸(DNA或RNA)。在该实施方案中,用减毒的微生物来传递异源抗原、蛋白质或核酸到体内的特定位点。可以通过常规技术(包括使用重组构建体,例如包含表达异源抗原或治疗性蛋白质的多核苷酸以及合适的启动子序列的载体)将异源抗原、肽或核酸引入到减毒的微生物中。另外,可将编码异源抗原或蛋白质的基因引入到生物体的基因组中,用内源启动子控制表达。
本发明的各种产物也可能用于兽医上的应用。

肽的确定
利用质粒载体pFW-phoA1、pFW-phoA2和pFW-phoA3(Podbielski,A.等人,Gene 1996;177:137-147)制备脑膜炎奈瑟球菌(菌株C311+)染色体DNA部分基因文库。这些载体具有组成性壮观霉素腺苷酰转移酶抗生素抗性标志,其产生高水平的壮观霉素抗性,因此容易选择。另外,这些载体含有截短的(缺乏前导序列的)大肠杆菌(Escherichia coli)碱性磷酸酶pho A基因。比较上游读框内的BamHI限制性酶切位点时,这三个载体只是缺乏前导序列的pho A基因存在的阅读框架不同。因为该截短的大肠杆菌pho A基因缺乏合适的前导序列使该酶输出过细菌膜,因此这些质粒在大肠杆菌pho A突变体(例如菌株DH5α)中增殖时没有细胞外碱性磷酸酶活性。显色碱性磷酸酶底物XP(5-溴-4-氯-3-吲哚-磷酸)不进入完整的细菌细胞,因此只能检测到输出的或表面结合的碱性磷酸酶活性。当存在输出的或表面结合的碱性磷酸酶活性时,显色底物XP被切割产生蓝色色素,通过其蓝色可以确定相应的细菌克隆。
质粒DNA用BamHI完全消化,用虾碱性磷酸酶去磷酸化。奈瑟球菌属基因组DNA用Sau3AI部分消化,使大部分的片段在用溴化乙锭染色的1%的琼脂糖凝胶上观察时呈现0.5-1.0kb大小。这些Sau3AI片段连接至制备好的pFW-phoA载体中。选择大肠杆菌菌株DH5α作为克隆宿主,因为其缺乏功能性phoA基因。在含有100μg/ml的壮观霉素和40μg/ml的XP显色底物的Luria琼脂上选择重组质粒。通过克隆的蓝色确定带有含有脑膜炎奈瑟球菌插入DNA的质粒的大肠杆菌转化子,脑膜炎奈瑟球菌插DNA补偿了缺乏前导序列的phoA基因的输出信号序列。
测序补偿了缺乏前导序列的phoA基因的输出信号序列的脑膜炎奈瑟球菌插入DNA,得到的序列检索GenBank数据库的已知蛋白质。结果示于表1。

候选蛋白质疫苗的保护性性质
筛选中确定的基因被估定为潜在的蛋白质疫苗候选者,其基于克隆的、表达的蛋白质在兔中产生免疫应答的能力,产生的抗体具有刺激补体介导的脑膜炎奈瑟球菌的溶菌作用的能力。通过活菌攻击用重组蛋白质免疫的小鼠来确定保护应答。
总之,PCR扩增、克隆候选者基因,编码的蛋白质表达并纯化。纯化的蛋白质用于产生用于酶联免疫吸附测定(ELISA)的抗体。也PCR扩增、克隆、表达和纯化PorA基因。PorA的单克隆抗体在幼兔感染模型中显示被动保护动物(Saukkonen等人,Microb.Pathog.,1987;3(4):261-267)。因此,该蛋白质用作一些试验中的阳性对照。已经表明PorA不能保护动物抵抗不同菌株的脑膜炎奈瑟球菌的攻击(Poolman,Infect.Dis.,1995;4:13),因此,任何能针对不同种类的脑膜炎奈瑟球菌菌株产生保护性免疫应答的候选者都比PorA有优势。
PCR扩增DNA

克隆疫苗候选者
将从候选者PCR扩增的DNA直接克隆到InVitrogen pCRT7/CT-TOPO载体中。该载体提供了T7启动子、核糖体结合位点和C-末端6×His标签融合体以促进表达和利用金属亲和层析的重组蛋白质的纯化。
为了克隆,将连接反应物转化进T0P10F’细胞(InVitrogen)。筛选来自转化子DNA克隆的DNA样品来检查插入DNA的方向。测序具有正确方向的插入DNA的候选者克隆以证实构建体5’区域的整合。表达和纯化
克隆的候选者转化到HMS174(DE3)pLysS感受态细胞(InVitrogen)后检测候选者基因的表达。用IPTG诱导候选者克隆的表达,并在诱导4小时后利用抗His抗体通过SDS PAGE和Western印迹分析表达。通过Talon树酯(利用在蛋白质的羧基末端克隆的6×His-标签的金属亲和层析柱(Clontech)),利用咪唑(imidozole)缓冲液从树酯上梯度洗脱大蛋白质(10-100mM),进行候选者蛋白质的纯化。
抗体纯化
抗体纯化前,预筛选在ELISA分析中对全细胞脑膜炎奈瑟球菌低反应原性的动物血清。利用100μg的蛋白质与弗氏佐剂进行起始接种,并与弗氏不完全佐剂间隔28天进行随后三次加强免疫,在兔子中产生针对每一克隆的和纯化的候选者的抗体。每次加强免疫后收集血清产生血清样本。

进行针对热杀死的脑膜炎奈瑟球菌的ELISA以证实针对纯化的蛋白质的抗体识别脑膜炎奈瑟球菌细胞。这些分析在菌株MC58以及下列菌株上进行:

热杀死的脑膜炎奈瑟球菌的制备
脑膜炎奈瑟球菌在巧克力的马血Columbia琼脂(Oxoid)上37℃、5%CO2生长14小时。从琼脂平板上刮下细胞,重悬细胞于50ml试管中的20mlPBS中。该细胞悬液在56℃加热30分钟以杀死细菌。
50μl热杀死的脑膜炎奈瑟球菌样品铺到巧克力的马血Columbia琼脂(Oxoid)上37℃、5%CO2温育18小时。这证实所有的脑膜炎奈瑟球菌都已经被杀死。然后用PBS洗涤热杀死的细胞。用PBS将悬液的OD620调整到0.1OD单位。
热杀死的脑膜炎奈瑟球菌的ELISA
用热杀死的全细胞脑膜炎奈瑟球菌进行ELISA分析。用热杀死的细胞过夜包被ELISA平板(96孔板的每一孔中加入50μl杀死的细菌PBS,4℃温育)。然后进行标准的EILSA程序,全部温育都在37℃、1小时。使用PBS/3%BSA封闭溶液、PBS/Tween 0.1%洗涤溶液、抗兔AP缀合第二抗体(Sigma)和Sigma Fast P硝基苯磷酸检测剂(Sigma)。利用合适的微量滴定板读数仪读取405nm处的数据。利用首次加强接种后7天(首次接种后的35天)获得的血清产生该数据。
ELISA数据
结果表明针对每一候选蛋白质产生的抗血清激发针对脑膜炎奈瑟球菌不同菌株的强烈应答。
体内筛选
为评价疫苗候选者的保护效能,用重组蛋白质免疫成年小鼠,通过活细菌攻击确定保护性应答。
对于每一种疫苗候选者,用25μg抗原间隔3周两次单独接种15只6周龄balb/C小鼠(皮下)。免疫程序结束后一周,用同源菌株MC58攻击该实验组。腹膜内接种500μl含细菌的Brain HeartInfusion/0.5%铁葡聚糖介质,剂量为1×106cfu。以前的结果已经表明在这些动物中起始菌血性(bacteraemic)疾病需要铁。该模型以前曾经用来证明接种疫苗的保护性效能(Lissolo等,Infect.Immun.,1995;63:884-890)。
对照组包括只用佐剂接种的动物(阴性对照)、用佐剂和纯化的PorA联合接种的动物(阳性对照)或减毒的同源菌株接种的动物。攻击后监测存活。
用候选者pho2-5接种的动物与未接种的对照相比显示80%(12/15)的存活,未接种的对照13%(2/15)存活。候选者pho2-5显示的保护水平与pro蛋白质相当(13/15)。
用候选者pho2-10、pho1-94、pho2-66接种的动物与未接种的对照相比分别显示40%(6/15)、47%(7/15)或27%(4/15)的存活,未接种的对照13%(2/15)存活。

Claims (16)
1.一种肽,由包括脑膜炎奈瑟球菌的、SEQ ID NOS.1、3、5、7、9、11、13、15、17、19、21、23、25、27、29、31、和33此处确定的任一种核苷酸序列的基因或其革兰氏阴性细菌同系物编码,或其功能性片段,用于治疗或诊断用途。
2.权利要求1的肽,其中所述的同系物在肽和核苷酸水平上有至少40%的序列相似性和同一性。
3.权利要求1的或权利要求2的肽,其中所述的同系物在肽和核苷酸水平上有至少60%的序列相似性和同一性。
4.任一前述权利要求的肽,其中所述的同系物在肽和核苷酸水平上有至少90%的序列相似性和同一性。
5.权利要求1的肽,包含SEQ ID NOS.2、4、6、8、10、12、14、16、18、20、22、24、26、28、30、32和34此处定义的任一氨基酸序列。
6.编码任一前述权利要求的肽的多核苷酸,用于治疗或诊断用途。
7.表达权利要求1-5任一项中的肽的转化宿主。
8.包含突变的微生物,该突变破坏权利要求1定义的任一核苷酸序列的表达。
9.权利要求8的微生物,其中所述的突变是插入失活或基因缺失。
10.权利要求8或权利要求9的微生物,其中所述的微生物是脑膜炎奈瑟球菌。
11.权利要求8-10任一项中的微生物,包含在第二核苷酸序列中的突变。
12.权利要求8-11任一项中的微生物,用于治疗或诊断用途。
13.一种疫苗,含有权利要求1-5任一项中的肽或其表达方法。
14.针对权利要求1-5任一项中的肽的抗体。
15.权利要求1-7任一项的产物在筛选分析中的用途。
16.权利要求1-12任一项的产物的用途,用于制备用于治疗或预防奈瑟球菌属或革兰氏阴性细菌感染相关的疾病的药物。
