Abstract
本发明公开了一种具有天然生物活性的初乳纳米肽及其制备方法和应用。本发明以健康奶畜产犊后72小时内的初乳为原料,经过离心、超滤、pH调节、酶解、纳滤等步骤获得了富集初乳纳米肽的产品,其主要成分为富脯氨酸多肽、胰岛素样生长因子‑1、表皮细胞生长因子、转化生长因子。所述产品还可进一步制成多种食品或者日化用品,例如,制成无菌液体口服液、饮料、粉末状产品,起到调节免疫力,促进骨骼生长,改善胃肠道,促进认知发育等作用,或是加入到化妆品基质中制成护肤品,起到改善表皮营养,延缓皮肤衰老,防皱增白等作用;或加入到牙膏基质中制成牙膏,起到快速促进细胞增殖,修复口腔微损伤等作用;或加入到洗发基质中制成防脱、固发的洗发水。
Description
技术领域
本发明涉及一种具有天然生物活性的初乳纳米肽及其制备方法和应用。本发明属于生物技术领域。
背景技术
初乳是大自然为新生命创造的第一口天然的完美实物。初乳本身已被证明是百种“原生营养”的天然聚合体,比如其含有高蛋白质、丰富维生素、低乳糖等物质。人类利用牛初乳的方向,并不是为了获取其中的基础营养,而是着眼于如何提升初乳中的免疫功能因子的功效,其中一些人们所熟知的免疫球蛋白IgG、乳铁蛋白等功能因子,发挥着提高人体免疫力、改善胃肠道等保健作用。
目前初乳产品还处在粗加工阶段,食品工业只是将初乳经过脱脂,灭菌后,加工成粉,方便人们食用,但目前此工艺还处在粗加工阶段,但初乳的功效物质远不止这些,有很多更重要的小分子功效物质,比如富脯氨酸多肽(PRP)、胰岛素样生长因子-1(IGF-1)、表皮细胞生长因子(EGF)、转化生长因子(TGF)等,由于现有技术、工艺的限制,食品工业一直没有开发出富集此类物质的产品,并发挥出这些组分应有的功效。
此外,初乳中的小分子功效物质,比如胰岛素样生长因子-1(IGF-1)、表皮细胞生长因子(EGF)、转化生长因子(TGF)、转移因子(TF)等,具有分子量小、易吸收、可以多种方式与受体结合而增强受体的特异性、降低毒性作用并增强皮肤的渗透性、较好稳定性和可溶性等特点,也为其在皮肤美容、治疗方面的广泛应用提供了很好的前提。但由于现有技术、工艺的限制,现代工业一直没有开发出富集此类物质的初乳提取液,并制备成能发挥出这些组分应有功效的日化产品。
发明内容
本发明为解决现有技术中的上述问题,提出了一种适合工业化生产的,能够富集具有天然生物活性的初乳纳米肽的生产方法及其产品和应用。
为了达到上述目的,本发明采用了以下技术手段:
本发明的一种具有天然生物活性的初乳纳米肽的制备方法,其包括以下步骤:
(1)收集健康奶畜产犊后48小时内的初乳,冷藏贮运;
(2)采用连续碟片式离心机,离心除去脂肪,获得脱脂初乳;
(3)采用酸化法或凝乳酶法凝乳,破乳、离心去除酪蛋白沉淀,获得初乳液;或者采用膜孔径为50KD~100KD的滤膜进行超滤,获得初乳液;
(4)用乳酸调节初乳液pH值至5.4~5.6,静置15分钟,再调节初乳液pH至3.9,40℃水浴150分钟;然后离心去除沉淀,获得液体-Ⅰ;
(5)再将液体-Ⅰ用40%w/w的NaOH溶液调节pH至4.5,静置15分钟后用高速离心机15000rpm离心30分钟,去除沉淀,得到液体-Ⅱ;
(6)向液体-Ⅱ中加入乳糖酶酶解乳糖,酶解2~3小时,90℃,30分钟灭酶,得到液体-Ⅲ;
(7)采用200分子量的纳滤膜进行纳滤,收集回流段液体-Ⅳ,所述的液体-Ⅳ中富集有所述的具有天然生物活性的初乳纳米肽。
其中,优选的,所述方法还包括步骤(8),即将获得的回流段液体-Ⅳ进一步采用0.2um的有机滤器进行除菌,得到符合商业无菌要求的液体-Ⅴ,所述的液体-Ⅴ中富集有所述的具有天然生物活性的初乳纳米肽。
其中,优选的,所述的健康奶畜包括牛、羊以及骆驼。
其中,优选的,步骤(2)中所述的离心除去脂肪是指在40℃~45℃下,5000rpm离心除去脂肪。
其中,优选的,步骤(4)中所述的离心是指采用碟片式离心机5000rpm离心去除沉淀。
其中,优选的,步骤(6)中的酶解条件为40℃,pH6~7,酶浓度500~1000ppm。
进一步的,本发明还提出了按照以上任一项所述的方法制备得到的富集具有天然生物活性的初乳纳米肽的产品。以及
所述的产品在制备食品以及日化用品中的应用。
其中,优选的,所述的产品在制备食品以及日化用品中的应用包括:1)将液体-Ⅳ与果汁、维生素调配后,采用0.2um的有机滤器进行除菌后灌装制成口服液产品,起到调节免疫力,促进骨骼生长,改善胃肠道,促进认知发育等作用;或将液体-Ⅳ采用冷冻干燥或低温喷雾干燥制成粉末产品。2)将液体-Ⅴ加入到化妆品基质中制成护肤品,起到改善表皮营养,延缓皮肤衰老,防皱增白等作用;或将液体-Ⅴ加入到牙膏基质中制成牙膏,起到可以快速促进细胞增殖,修复口腔微损伤等作用;或将液体-Ⅴ加入到含表面活性剂的洗发基质中制成防脱、固发的洗发水。
相较于现有技术,本发明的有益效果是:
①常规的工艺是采用酸法或者酶法沉淀酪蛋白,收集乳清后在进行下一步分离。这种工艺会造成初乳资源的浪费,降低经济效益。本发明采用超滤法获得初乳液,不影响主体初乳产品的下游生产工艺,而且免疫球蛋白IgG、乳铁蛋白等特征成分的含量会有所提升。
②通过调整液体pH、离心可以去除α-乳白蛋白,β-乳球蛋白(可单独开发成产品)。
③采用200D聚丙烯腈纳滤膜固定化乳糖酶的方法,水解乳糖变单糖,再纳滤除糖、除无机盐,增加了纳滤的效率,而且保留了200D分子量以上的功效物质含量。主要成分为富脯氨酸多肽(PRP)、胰岛素样生长因子-1(IGF-1)、表皮细胞生长因子(EGF)、转化生长因子(TGF)。
④终端产品形式多样,可制成无菌液体口服液,调配成饮料,制备成粉末状产品,起到调节免疫力,促进骨骼生长,改善胃肠道,促进认知发育等作用,或是加入到化妆品基质中制成护肤品,起到改善表皮营养,延缓皮肤衰老,防皱增白等作用;或加入到牙膏基质中制成牙膏,起到可以快速促进细胞增殖,修复口腔微损伤等作用;或加入到含表面活性剂的洗发基质中制成防脱、固发的洗发水。
⑤工艺全程采用低温操作,避免了生物功能性物质的活性损失。

具体实施方式
下面结合具体实施例来进一步描述本发明,本发明的优点和特点将会随着描述而更为清楚。但实施例仅是范例性的,并不对本发明的范围构成任何限制。本领域技术人员应该理解的是,在不偏离本发明的精神和范围下可以对本发明技术方案的细节和形式进行修改或替换,但这些修改和替换均落入本发明的保护范围内。
实施例1具有天然生物活性的初乳纳米肽及其相关产品的制备
本发明实施例1的方法流程图如图1所示,具体包括如下步骤:
(1)收集健康奶牛产犊后48小时内的初乳,冷藏贮运;
(2)初乳采用连续碟片式离心机,40℃~45℃下,5000rpm离心除去脂肪,获得脱脂初乳;
(3)脱脂初乳采用膜孔径为100KD的滤膜进行超滤,获得初乳液;
(4)用乳酸调节初乳液pH值至5.4~5.6,静置15分钟,再调节初乳液pH至3.9,40℃水浴150分钟;然后采用碟片式离心机5000rpm离心去除沉淀,获得液体-Ⅰ;
(5)再将液体-Ⅰ用40%w/w的NaOH溶液调节pH至4.5,静置15分钟后用高速离心机15000rpm离心30分钟,去除沉淀,得到液体-Ⅱ;
(6)将液体-Ⅱ加入到已在200D聚丙烯腈纳滤膜内表面固定化乳糖酶的纳滤系统的循环罐中,关闭透过液端,循环回流进行酶解反应2.5小时,酶解条件为40℃,pH6.5,酶浓度800ppm,得到液体-Ⅲ;
(7)开启纳滤系统滤出端,滤出液为单糖组分,回流段为液体-Ⅳ,收集回流段液体-Ⅳ,所述的液体-Ⅳ中富集有所述的具有天然生物活性的初乳纳米肽。
将液体-Ⅳ采用0.2um过滤器除菌后,再采用无菌灌装机灌装制成满足商业无菌要求的产品-Ⅰ;或将液体-Ⅳ与果汁、维生素调配后,采用0.2um的有机滤器进行除菌后灌装制成口服液产品-II;或将液体-Ⅳ采用冷冻干燥或低温喷雾干燥制成粉末产品-III。制备得到的产品具备调节免疫力,促进骨骼生长,改善胃肠道,促进认知发育等作用。
对原初乳、产品-I以及产品-III中的小分子功效物质(PRP、IGF-1、EGF以及TGF)含量进行检测,结果如表1所示。

实施例2具有天然生物活性的初乳纳米肽及其相关产品的制备
本发明实施例2的方法流程图如图2所示,具体包括如下步骤:
(1)收集健康奶牛产犊后72小时内的初乳,冷藏贮运;
(2)初乳采用连续碟片式离心机,40℃~45℃下,5000rpm离心除去脂肪,获得脱脂初乳;
(3)脱脂初乳采用膜孔径为80KD的滤膜进行超滤,获得初乳液;
(4)用乳酸调节初乳液pH值至5.4~5.6,静置15分钟,再调节初乳液pH至3.9,40℃水浴150分钟;然后采用碟片式离心机5000rpm离心去除沉淀,获得液体-Ⅰ;
(5)再将液体-Ⅰ用40%w/w的NaOH溶液调节pH至4.5,静置15分钟后用高速离心机15000rpm离心30分钟,去除沉淀,得到液体-Ⅱ;
(6)将液体-Ⅱ加入到已在200D聚丙烯腈纳滤膜内表面固定化乳糖酶的纳滤系统的循环罐中,关闭透过液端,循环回流进行酶解反应2小时,酶解条件为40℃,pH7,酶浓度1000ppm,得到液体-Ⅲ;
(7)开启纳滤系统滤出端,滤出液为单糖组分,回流段为液体-Ⅳ,收集回流段液体-Ⅳ,所述的液体-Ⅳ中富集有所述的具有天然生物活性的初乳纳米肽;
(8)将获得的回流段液体-Ⅳ进一步采用0.2um的有机滤器进行除菌,得到符合商业无菌要求的液体-Ⅴ,所述的液体-Ⅴ中富集有所述的具有天然生物活性的初乳纳米肽。
对原初乳以及产品-V中的小分子功效物质(IGF-1、EGF以及TGF)含量进行检测,结果如表2所示。

将液体-Ⅴ加入到化妆品基质中制成护肤品(例如膏霜等),起到改善表皮营养,延缓皮肤衰老,防皱增白等作用;或将液体-Ⅴ加入到牙膏基质中制成牙膏,起到快速促进细胞增殖,修复口腔微损伤等作用;或将液体-Ⅴ加入到含表面活性剂的洗发基质中可制成防脱、固发的洗发水。
Claims (10)
1.一种具有天然生物活性的初乳纳米肽的制备方法,其特征在于,包括以下步骤:
(1)收集健康奶畜产犊后72小时内的初乳,冷藏贮运;
(2)初乳采用连续碟片式离心机,离心除去脂肪,获得脱脂初乳;
(3)脱脂初乳采用膜孔径为50KD~100KD的滤膜进行超滤,获得初乳液;
(4)用乳酸调节初乳液pH值至5.4~5.6,静置15分钟,再调节初乳液pH至3.9,40℃水浴150分钟;然后离心去除沉淀,获得液体-Ⅰ;
(5)再将液体-Ⅰ用40%w/w的NaOH溶液调节pH至4.5,静置15分钟后用高速离心机15000rpm离心30分钟,去除沉淀,得到液体-Ⅱ;
(6)将液体-Ⅱ加入到已在200D聚丙烯腈纳滤膜内表面固定化乳糖酶的纳滤系统的循环罐中,关闭透过液端,循环回流进行酶解反应2~3小时,得到液体-Ⅲ;
(7)开启纳滤系统滤出端,滤出液为单糖组分,回流段为液体-Ⅳ,收集回流段液体-Ⅳ,所述的液体-Ⅳ中富集有所述的具有天然生物活性的初乳纳米肽。
2.如权利要求1所述的制备方法,其特征在于,还包括步骤(8),即将获得的回流段液体-Ⅳ进一步采用0.2um的有机滤器进行除菌,得到符合商业无菌要求的液体-Ⅴ,所述的液体-Ⅴ中富集有所述的具有天然生物活性的初乳纳米肽。
3.如权利要求1所述的制备方法,其特征在于,所述的健康奶畜包括牛、羊以及骆驼。
4.如权利要求1所述的制备方法,其特征在于,步骤(2)中所述的离心除去脂肪是指在40℃~45℃下,5000rpm离心除去脂肪。
5.如权利要求1所述的制备方法,其特征在于,步骤(4)中所述的离心是指采用碟片式离心机5000rpm离心去除沉淀。
6.如权利要求1所述的制备方法,其特征在于,步骤(6)中的酶解条件为40℃,pH6~7,酶浓度500~1000ppm。
7.按照权利要求1-6任一项所述的方法制备得到的富集具有天然生物活性的初乳纳米肽的产品。
8.权利要求7所述的产品在制备食品以及日化用品中的应用。
9.如权利要求8所述的应用,其特征在于,将液体-Ⅳ与果汁、维生素调配后,采用0.2um的有机滤器进行除菌后灌装制成口服液产品;或将液体-Ⅳ采用冷冻干燥或低温喷雾干燥制成粉末产品。
10.如权利要求8所述的应用,其特征在于,将液体-Ⅴ加入到化妆品基质中制成护肤品;或将液体-Ⅴ加入到牙膏基质中制成牙膏;或将液体-Ⅴ加入到含表面活性剂的洗发基质中制成洗发水。
