Abstract
The present invention relates to a composition for skin regeneration and hair growth promotion comprising apigenin, more specifically, the composition for skin regeneration and hair growth promotion promotes cell differentiation through Wnt/β catenin (Wnt/beta-catenin) signaling, thereby exhibiting skin regeneration and hair growth promotion effects.
Description
Technical Field
The present application claims the benefit of priority based on korean patent application No. 10-2018-0140423, which was filed on the date of 2018, 11 and 15, and all contents disclosed in the document of this korean patent application are included in the present specification as part of the present specification.
The present invention relates to a composition for skin regeneration and hair growth promotion comprising apigenin.
Background
Skin (skin) exists on the outermost layer of the human body and is directly contacted with the external environment, so that the human body is protected from being damaged by the outside, and the effect of preventing excessive loss of water and heat in the human body is achieved. The skin is composed of three layers of epidermis (epidermis), dermis (dermis), subcutaneous fat (subcutaneous tissue), and most of the epidermis located at the outermost side is composed of keratinocytes, which protect the skin by forming horny layer through proliferation, migration and differentiation processes from the basal layer, and produce various cytokines to participate in inflammatory and immune reactions. In addition, proliferation and migration of keratinocytes not only provide wound healing and skin regeneration, but also induce synthesis of collagen (collagen) to enhance elastic promotion and basal membrane formation of skin, regulate attachment and proliferation of skin cells, enhance barrier function of selectively penetrating harmful substances, provide interaction of epidermis and dermis, and the like. Thus, the movement and proliferation of skin epidermal keratinocytes not only promote skin regeneration and wound healing, but also promote skin elasticity, helping to maintain healthy skin.
With the increasing interest in skin beauty, various attempts for effectively suppressing and alleviating skin aging have been widely performed in order to maintain healthier and beautiful skin, to maintain and enhance the inherent functions of skin and to activate skin cells by using bioactive substances derived from various natural substances, which are obtained from various animals, plants, microorganisms, etc., with high safety.
Apigenin (apigenin) is a nutritional ingredient contained in vegetables as an antioxidant ingredient, and is known to be used as an ingredient for promoting skin whitening, skin regeneration, hair growth, and the like.
Korean laid-open patent No. 2017-019757 discloses the following: as a cosmetic composition using apigenin, germanium and amino acid, skin regeneration and stabilization effects can be exhibited by synergistic effects expressed by the effects of the respective components.
Chinese patent No. 106880693 discloses that the composition comprising apigenin and ginseng extract has anti-alopecia effect.
As such, although technological development is continuously being conducted on products related to skin regeneration or hair loss using apigenin, consumer satisfaction of the efficacy thereof is still at a low level.
Accordingly, there is a continuous demand for research on developing products that will maximize the effects of apigenin having skin regeneration and hair growth promoting effects, and using ingredients that produce synergistic effects with apigenin.

Disclosure of Invention
In order to solve the above problems, the present inventors prepared a composition comprising apigenin together with one or more of caffeine, ginseng extract and green tea extract, and confirmed that the composition has excellent skin regeneration and hair growth promoting effects.
Accordingly, an object of the present invention is to provide a composition for skin regeneration and hair growth promotion comprising apigenin.
In order to achieve the above object, the present invention provides a composition for skin regeneration and hair growth promotion, comprising: apigenin (apigenin); and at least one selected from the group consisting of caffeine, ginseng extract and green tea extract.
The apigenin may be present in an amount of 40 wt% to 60 wt% based on the total weight of the composition.
The one or more selected from the group consisting of caffeine, ginseng extract and green tea extract may be contained in an amount of 40 to 60 wt% based on the total weight of the composition.
The composition may activate a Wnt/beta-catenin signal (Wnt/beta-catenin signal).
The concentration of the composition may be 10ppm to 40ppm based on the solid content concentration.
The composition may comprise apigenin and caffeine.
The composition may comprise apigenin and ginseng extract.
The composition may comprise apigenin and green tea extract.
The composition may be one or more selected from the group consisting of a cosmetic composition, a food composition, and a pharmaceutical composition.
The composition for skin regeneration and hair growth promotion of the present invention contains one or more of caffeine, ginseng extract and green tea extract together with apigenin, so that it exhibits skin regeneration effects due to the synergistic effect of the effects of the components, and in particular, can exhibit excellent skin wound healing and scar reduction effects.
The composition for promoting hair growth has an effect of promoting hair growth and preventing or improving hair loss.
In addition, the composition for skin regeneration and hair growth promotion promotes cell differentiation by transmitting Wnt/β -catenin signal, and thus can further improve skin regeneration and hair growth promotion effects.
The composition for skin regeneration and hair growth promotion can be widely used for cosmetic compositions, food compositions and pharmaceutical compositions having skin regeneration and hair growth promotion effects.

Detailed Description
The present invention is described in detail below so that a person having ordinary skill in the art to which the present invention pertains can easily practice the present invention. The invention may, however, be embodied in many different forms and should not be construed as limited to the embodiments set forth herein.
Composition for skin regeneration and hair growth promotion
The present invention relates to a composition for skin regeneration and hair growth promotion, comprising: apigenin (apigenin); and 1 or more selected from the group consisting of caffeine, ginseng extract and green tea extract.
The apigenin; and 1 or more selected from the group consisting of caffeine, ginseng extract and green tea extract have skin regeneration or hair growth promoting effects, respectively, but when they are combined, the skin regeneration or hair growth promoting effects can be significantly improved due to the synergistic effect of the components thereof.
Preferably, the composition for skin regeneration and hair growth promotion comprises apigenin and caffeine, or apigenin and ginseng extract, or a combination of apigenin and green tea extract. When the composition for skin regeneration and hair growth promotion contains the components of such a combination, the synergistic effects for skin regeneration and hair growth promotion can be optimally varied.
In the present invention, the apigenin is one of flavonoid components contained in fruits, vegetables or herbs, and may be extracted from the fruits, vegetables or herbs.
The apigenin may be contained in an amount of 40 to 60 wt%, specifically, 40 to 42 wt% or 45 wt% or less, or 55 to 58 wt% or 60 wt% or less, based on the total weight of the skin regeneration or hair growth promoting composition. If it is smaller than the range or larger than the range, the synergistic effect with one or more selected from the group consisting of caffeine, ginseng extract and green tea extract may be reduced.
In the present invention, the one or more selected from the group consisting of caffeine, ginseng extract and green tea extract may exhibit synergistic effects for skin regeneration and hair growth promotion by combining with the apigenin.
The one or more selected from the group consisting of caffeine, ginseng extract and green tea extract may be contained in an amount of 40 to 60 wt%, specifically, 40 to 42 wt% or 45 wt% or 55 wt% or less, 58 wt% or 60 wt% or less based on the total weight of the composition for skin regeneration or hair growth promotion. If it is less than the range or more than the range, the synergistic effect with apigenin may be reduced.
The caffeine is not particularly limited, but may be selected from synthetic caffeine; or more than one natural caffeine extract form selected from the group consisting of guarana extract, coffee extract and tea extract.
The concentration of caffeine may be 5ppm to 20ppm, specifically, 5ppm or more or 7ppm or more, and 12ppm or less, 15ppm or less, or 20ppm or less, based on the solid content concentration. If it is less than the above range, the skin regeneration and hair growth promoting effect may be weak, and if it is more than the above range, the optimum concentration for the synergistic effect with apigenin may be exceeded, and the skin regeneration and hair growth promoting effect may be reduced as well. In this case, the concentration of caffeine means the concentration of an aqueous solution of caffeine in the case of synthetic caffeine, and means the concentration of a solid component in the form of a natural caffeine extract.
The ginseng extract may be at least one selected from the group consisting of ginseng root extract, ginseng leaf extract, ginseng seed extract and ginseng seed extract, and preferably is ginseng root extract.
The concentration of the ginseng extract may be 5ppm to 20ppm, specifically, may be 5ppm or more and 7ppm or more, and may be 12ppm or less, 15ppm or less, or 20ppm or less, based on the solid content concentration. If it is less than the above range, the skin regeneration and hair growth promoting effect may be weak, and if it is more than the above range, the optimum concentration for the synergistic effect with apigenin may be exceeded, and the skin regeneration and hair growth promoting effect may be reduced as well.
The ginseng extract may be extracted with an extraction solvent selected from the group consisting of water, an organic solvent, and a mixed solvent of the water and the organic solvent, wherein the organic solvent is at least one selected from the group consisting of ethanol, methanol, butanol, ether, ethyl acetate, and chloroform. Preferably, the extraction solvent may be ethanol.
The method for producing the ginseng extract is not particularly limited as long as it is a conventional extraction method used in the art, and it can be produced, for example, by extracting with the above-mentioned extraction solvent at 10 to 80℃for 3 to 24 hours.
The green tea extract may be at least one selected from the group consisting of green tea leaf extract, green tea flower extract and green tea stem extract, and preferably is a green tea leaf extract.
The concentration of the green tea extract may be 5ppm to 20ppm, specifically, 5ppm or more or 7ppm or more, and 12ppm or less, 15ppm or less or 20ppm or less based on the solid content concentration. If it is less than the above range, the skin regeneration and hair growth promoting effect may be weak, and if it is more than the above range, the optimum concentration for the synergistic effect with apigenin may be exceeded, and the skin regeneration and hair growth promoting effect may be reduced as well.
In addition, the green tea extract may be a hot water extract or C 1 To C 5 Lower alcohols of (2). Preferably, the green tea extract may be an ethanol extract.
In addition, the method of preparing the green tea extract is not particularly limited as long as it is a conventional extraction method used in the art, and for example, when the green tea extract is a hot water extract, it can be obtained through the processes of adding green tea, hot water extraction, filtration, concentration under reduced pressure, and spray drying. Alternatively, when the green tea extract is an ethanol extract, it can be prepared by extracting with ethanol at 10 to 80℃for 3 to 24 hours.
In the present invention, the concentration of the composition for skin regeneration and hair growth promotion may be 10ppm to 40ppm, specifically, 10ppm or more or 15ppm or more, and 25ppm or less, 30ppm or less, or 40ppm or less, based on the solid content concentration. If it is less than the range or more than the range, the skin regeneration and hair growth promoting effect may be reduced. In addition, wnt/β -catenin signaling, as described below, may be activated when the concentration of the composition is within the concentration range.
The composition for promoting skin regeneration and hair growth can activate Wnt/beta-catenin signal (Wnt/beta-catenin signal) to promote skin regeneration and hair growth.
Wnt/β -catenin (Wnt/beta-catenin) signaling is known to be a signaling system associated with the occurrence of living bodies, and acts on skin production and hair follicle generation, and further, if a wound appears on the skin, wnt signaling is expressed essentially during skin tissue repair, and epithelial Cell differentiation is promoted (Fathke et al, "Wnt signaling induces epithelial differentiation during cutaneous wound healing", BMC Cell Biology 2006, 7:4). Therefore, if Wnt signaling is promoted, differentiation of epithelial cells is promoted, which can promote skin regeneration and, in turn, healing of skin wounds. Gardenia jasminoides ellis extract promotes Wnt/beta-catenin (Wnt/beta-catenin) signaling of stem cells. Thus, formulations comprising this ingredient are capable of promoting skin regeneration, in particular, healing of skin wounds (Kateri A. Moore et al., "Stem Cells and Their Niches", science, VOL 311, p 1880-1885).
In the present invention, the composition for skin regeneration and hair growth promotion may be one or more selected from the group consisting of a cosmetic composition, a food composition, and a pharmaceutical composition.
When the composition for skin regeneration and hair growth promotion is a cosmetic composition, it may be formulated to contain a cosmetically or dermatologically allowable medium or mechanism. In addition, the cosmetic composition for skin regeneration and hair growth promotion may be provided in all formulations suitable for topical use, for example, may be provided in the form of an emulsion obtained by dispersing an oil phase in a solution, an aqueous phase, an emulsion obtained by dispersing an aqueous phase in an oil phase, a suspension, a solid, a gel, a powder, a paste, a foam (foam), or an aerosol composition. The composition of such dosage forms may be prepared according to methods conventional in the art.
In addition, the composition for skin regeneration and hair growth promotion may preferably contain other ingredients capable of exerting a synergistic effect on the primary efficacy, in addition to the substance, within a range not impairing the primary efficacy. In addition, the composition for skin regeneration and hair growth promotion may further comprise a moisturizer, an emollient, an ultraviolet absorbent, a preservative, a bactericide, an antioxidant, a pH adjustor, an organic and inorganic pigment, a perfume, a coldness feeling agent, or an antiperspirant. The amount of the above-mentioned components to be blended may be within a range not impairing the object and effect of the present invention, and may be easily selected by those skilled in the art, and may be 0.01% by weight or more and may be 3% by weight or less or 5% by weight or less relative to the total weight of the composition.
When the composition for skin regeneration and hair growth promotion is a food composition, the composition can be applied to various forms of food additives or functional foods. The food composition for skin regeneration and hair growth promotion can be processed into fermented milk, cheese, yogurt, fruit juice, probiotics, health products, etc., and can be used as various food additives.
In addition, the food composition for skin regeneration and hair growth promotion may contain other ingredients that exert a synergistic effect on the primary efficacy within a range that does not impair the primary efficacy for the purpose of the present invention. For example, additives such as perfume, pigment, bactericide, antioxidant, preservative, humectant, thickener, inorganic salt, emulsifier, and synthetic polymer may be further contained for improving physical properties. In addition, the food can further comprise water-soluble vitamins, oil-soluble vitamins, high molecular peptides, high molecular polysaccharides, seaweed essence and other auxiliary components. The components may be appropriately selected and formulated according to the dosage form or purpose of use without any difficulty by those skilled in the art, and the amount to be added may be selected within a range not impairing the purpose and effect of the present invention. For example, the amount of the above-mentioned components may be 0.01 wt% or more and may be 3 wt% or less or 5 wt% or less based on the total weight of the composition.
The food composition for skin regeneration and hair growth promotion according to the present invention can be formulated into various forms such as solutions, emulsions, viscous mixtures, tablets, powders, etc., and can be administered by various methods such as simple drinking, injection administration, spray administration, or extrusion.
When the composition for skin regeneration and hair growth promotion is a pharmaceutical composition, the composition can be applied to pharmaceuticals, and a conventional inorganic or organic carrier can be added to the active ingredient used in the present invention to prepare an oral or non-oral preparation in the form of a solid, semi-solid or liquid phase, and the pharmaceutical composition according to the present invention can be administered orally, non-orally, rectally, topically, transdermally, intravenously, intramuscularly, intraperitoneally, subcutaneously, etc.
The dosage form for the oral administration may be exemplified by tablets, pills, granules, capsules, suppositories, fine granules, powders, opacifiers, syrups, pellets and the like. In addition, the dosage forms for the non-oral administration may be exemplified by injections, drops, ointments, emulsions, sprays, suspensions, emulsions, adjuvants, and the like. The composition according to the present invention can be carried out according to a conventional method so that the active ingredient can be easily formulated, and in this case, surfactants, excipients, colorants, spices, preservatives, stabilizers, buffers, suspending agents, and other commonly used adjuvants can be suitably used.
In addition, the administration dosage of the active ingredient of the pharmaceutical composition of the present invention will vary depending on the age, sex, weight, pathological state of the subject to be administered, and severity thereof, administration route or judgment of the prescribing person. It is within the level of skill in the art to determine an appropriate dosage based on these factors that a 1 day dosage may be, for example, 0.1 mg/kg/day to 100 mg/kg/day, more specifically 5 mg/kg/day to 50 mg/kg/day, but is not limited thereto.
The composition for skin regeneration and hair growth promotion as described above contains one or more of caffeine, ginseng extract and green tea extract together with apigenin, and thus exhibits a skin regeneration effect, specifically an excellent effect of skin wound healing and scar reduction due to the synergistic effect of the effects of the respective components, and exhibits an effect of hair growth promotion, and thus can be widely applied to the fields of cosmetics, foods and pharmaceuticals having such effects.
Skin regeneration and hair growth promoting method
The present invention also relates to a method for regenerating skin and promoting hair growth using the composition for regenerating skin and promoting hair growth.
The composition can be applied to skin such as scalp to regenerate skin or promote hair growth.
Alternatively, oral administration may be performed depending on the dosage form of the composition.
Mode for carrying out the invention
In the following, preferred embodiments are disclosed for facilitating understanding of the present invention, but the following embodiments are merely illustrative of the present invention, and various changes and modifications may be made within the scope and technical spirit of the present invention, which will be apparent to those skilled in the art, and such changes and modifications are intended to fall within the scope of the appended claims as well.
Preparation example 1: preparation of apigenin
Herba seu radix Cirsii Japonici (circium japonicum) is prepared from Han Guozhong in mountain area by drying. Purchased from the Qingdong market (head, korea) and dried for use. Extracting dried herba Cirsii Japonici (Circium japonicum) with 95% ethanol, and concentrating under reduced pressure to obtain crude ethanol extract.
The crude ethanol extract was dissolved in distilled water and ethyl acetate, mixed at a ratio (v/v) of 1:1, repeated 3 times for partitioning between different phases, and the ethyl acetate extract was decompressed, concentrated, and dried.
The dried product was subjected to silica gel flash column chromatography (solvent: chloroform/acetic acid=2:1), and after the active ingredient was decompressed and dried again, it was dissolved in a small amount of methanol in a saturated state, and was filtered using a microporous filter (0.45 nm), and apigenin, an active substance, was separated by a high performance analyzer (HPLC, column chromatography: ODS 20×250nm;80% ethanol).
Preparation example 2: preparation of caffeine
Caffeine was purchased from sigma chemicals company (St.Louis, MO, USA).
Preparation example 3: preparation of Ginseng radix extract
Crushing 1kg of dried white ginseng by using a stirrer, adding 5L of purified water, stirring and extracting with warm water at 38-42 ℃ for 24 hours, and depositing for 1 day at 15 ℃. After that, the residue and the filtrate were separated by filtration through a filter cloth and centrifugal separation, and the separated filtrate was concentrated under reduced pressure to obtain 270g of an extract thereof.
Preparation example 4: preparation of green tea extract
After 1kg of green tea leaves processed for the first time (baking process) were crushed by a stirrer, 5L of purified water was added, and after 24 hours of stirring extraction with warm water at 38 to 42 ℃, the green tea leaves were deposited at 15℃for 1 day. After that, the residue and the filtrate were separated by filtration through a filter cloth and centrifugal separation, and the separated filtrate was concentrated under reduced pressure to obtain 290g of an extract thereof.
Example 1
The apigenin 50 wt% and caffeine 50 wt% prepared in preparation example 1 and preparation example 2 were mixed together to prepare a 20ppm composition.
Example 2
The apigenin 50 wt% and ginseng extract 50 wt% prepared in preparation example 1 and preparation example 3, respectively, were mixed to prepare a 20ppm composition.
Example 3
The apigenin 50 wt% and the green tea extract 50 wt% prepared in preparation example 1 and preparation example 3, respectively, were mixed to prepare a 20ppm composition.
Comparative examples 1 to 8
Comparative examples 1 to 8 were prepared with 20ppm of each of the compositions shown in Table 1.

When Wnt signaling was applied to epidermal stem cells, protein complexes (GSK 3, APC, axin) that inhibit intracellular β -catechin were blocked, and β -catechin was moved into the nucleus, and various genes related to formation and hair growth were expressed, and by using this principle, the Wnt signaling levels of the compositions according to examples and comparative examples were examined. The specific experimental method is as follows.
[ Wnt signaling assay ]
(1) Cell culture
After purchase of the Wnt reporter cell line (Wnt reporter cell line), it was plated in a 96-well plate (well plate) coated with white/clear flat bottom (white/clear flat bottom) of Collagen type I (Collagen type I) (#ENZ-61002-0001) at 1X10 4 Cells/wells) were seeded with Wnt reporter cell lines.
(2) Material handling
The compositions of examples and comparative examples were treated at different concentrations with 100. Mu.L each for 24 hours using serum-free medium (Serum free medium).
The Positive control group (Positive control) used 10mM lithium chloride (10 mM showed 150% to 300% of activity relative to the control group, and 50mM showed 1000% to 4000% of activity relative to the control group, depending on the state of the cells).
(3) Determination of Luciferase (Luciferase) Activity
mu.L of ONE-GloTM luciferase assay reagent (Luciferase Assay reagent) was added to each Well (Well), and the reaction was blocked at room temperature for 3 minutes.
Activity was measured using a Wallac Victor2 multi-label analyzer (multilabel counter).
(4) Data processing method
The value obtained by converting the luminescence (luminescence) intensity of the substance-treated group with respect to the control group into a% value was used as a Response value (Response ratio) based on the absorbance of the control group not subjected to the substance treatment, and the larger the Response value is, the more excellent the Wnt signaling activity is.
FIG. 1 is a graph showing Wnt signaling activities of compositions prepared in examples and comparative examples, respectively.
Referring to fig. 1, it can be seen that the Wnt signaling activity of the composition comprising the combination of apigenin and caffeine of example 1, the combination of apigenin and ginseng extract of example 2, and the combination of apigenin and green tea extract of example 3 was excellent.
In contrast, compared with examples 1, 2 and 3, the Wnt signaling activity of the compositions comprising apigenin, ginseng extract, green tea extract and madecassoside (madecassoside) alone as in comparative examples 3 to 7 was significantly reduced as compared with the compositions comprising apigenin and madecassoside as in comparative example 8.
From this, it was confirmed that the Wnt signaling activity was significantly improved in the composition comprising the combination of apigenin and caffeine of example 1, the combination of apigenin and ginseng extract of example 2, and the combination of apigenin and green tea extract of example 3, due to the synergistic effect of the combination of the components.
While the present invention has been described above with reference to the limited embodiments and the accompanying drawings, the present invention is not limited thereto, and a person having ordinary skill in the art to which the present invention pertains may naturally make various modifications and variations within the technical spirit of the present invention and the scope of equivalents of the claims to be described below.
Claims (3)
1. Use of apigenin and at least one selected from the group consisting of caffeine and ginseng extract for preparing cosmetic for skin regeneration,
the apigenin comprises 40 to 60 wt% based on the total weight of the composition,
one or more selected from the group consisting of caffeine and ginseng extract comprises 40 to 60 wt% based on the total weight of the composition.
2. The use of a composition comprising apigenin and at least one selected from the group consisting of caffeine and ginseng extract according to claim 1, wherein,
the composition comprises apigenin and caffeine.
3. The use of a composition comprising apigenin and at least one selected from the group consisting of caffeine and ginseng extract according to claim 1, wherein,
the composition comprises apigenin and ginseng extract.
