Abstract
本发明提供同时进行生化检查和血液检查的多重系统,其特征在于:在多从圆盘(1)上以旋转中心为准呈扇形划分形成生物化学检查部(100)、血液检查部(200)及免疫检查部(300);通过使用本发明的多重圆盘(1),具有可同时在一个装置上进行通过测量血清的生物化学成分的浓度诊断人体功能的生物化学检查、通过分离血液样本测量检查红血球或白血球的数量及变形的血液检查及通过测量血清的抗原‑抗体反应诊断癌症或病毒并进行检查的免疫检查的效果。

技术领域
本发明涉及同时进行生化检查和血液检查的多重系统,尤其涉及利用基于离心力的圆盘,快速简便的评价反应物质对血液成分的反应与否,可在一个平台测量及检查利用血液的血清内的酶促反应的生物化学测量检查和利用毛细管法和染色法等计算红血球或白血球的数量的血液检查及通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查的免疫检查的同时进行生化检查和血液检查的多重系统及用于此的多重圆盘。
背景技术
血液不仅起到向体内的所有脏器的细胞供应氧气、营养成分、激素及营养物质的重要作用,而且是对抗感染的防御机构,还起到搬运毒素和代谢物的功能,因此,体内发生的变化将对血液成分产生影响。因此,为了准确判断疾病的诊断、治疗及预后,需要检查血液的各种成分。
为了进行上述血液检查,对采集的血液进行形态学、生物化学、免疫血清学检查和特殊化学检查。
在此,血液的形态学检查使用全血实施测量血色素、血球容积、红血球、白血球、血小板数、白血球的细胞百分比等的血球检查及血涂片检查(将血液薄薄地涂抹于载玻片进行染色之后,直接利用显微镜观察血球的形状、数量等),检查出血时间、凝固时间、毛细血管脆性等,进行出血倾向血液疾病感染症等的诊断和各种疾病的经过和预后判断,此时,利用电化学方法和染色法进行测量。
另外,生物化学方法或免疫学方法及特殊化学方法,首先利用离心分离等去除血液内的雪球之后,利用上清液(血清或血浆)进行检查。
一般而言,生物化学方法主要采用在各试剂中反应血清(或血浆)之后,对其测量特定波长的吸光度换算成浓度的方法,根据所使用的试剂的种类分为湿式和干式。
湿式是大部分选择的方法,试剂呈液体形式,与血浆(或血清)进行反应。为更简便地进行测量,干式方法在薄片上固化试剂进行涂布之后,将其与血清(或血浆)进行反应并测量吸光度。
另外,免疫血清检查是检查对各种细菌及病毒的抗原、抗体,诊断乙肝、丙肝、艾滋病、梅毒等,另外,从血液中分离免疫细胞实施风湿病的诊断及检查各种免疫细胞的功能,通过测量对成为其他过敏的各种原因的物质的血液内的特定免疫球蛋白,查明过敏的原因。
在大部分免疫血清检查的情况下,利用抗原-抗体的免疫反应,一般采用在需要检查的抗体(抗原)上标记标识测量该抗体(抗原)的量的方法。
为了同时进行利用血液的上述大量的试料检查,近来积极开展相关的技术开发,而DNA chip或lab-on-a-chip等方法就是其一例。上述方法主要用于同时进行免疫血清检查和基因检查。
同样,亟需开发出一种能够同时在一个装置上完成用于计算血球的数量的形态学检查、生物化学检查、免疫血清检查和基因检查的检查装置及试剂盒。

技术课题
本发明的目的在于克服现有技术之不足而提供一种利用基于离心力的圆盘,快速简便的评价反应物质对血液成分的反应与否,可同时在一个平台上进行生物化学检查和血液检查及免疫检查等全部检查的多重系统。
课题解决方案
为解决上述问题,本发明的同时进行生化检查和血液检查的多重系统,包括以旋转中心为准呈扇形划分形成生物化学检查部100及血液检查部200的多重圆盘1。
另外,在本发明的多重系统中,上述多重圆盘1还具备免疫检查部300,从而还同时进行免疫检查。

反应槽板20,与上述上部盖10拆卸式结合或一体形成。

另外,在本发明的多重系统中,上述血液接收槽110在与旋转中心相邻的位置以旋转轴为中心呈弧形形成,以接收所供应的血液样本;
上述血液过滤槽120以旋转中心的旋转轴为中心呈弧形形成,在内部具备过滤器,以过滤移送到的血液样本;
上述多个血清分配槽130,因各血清分配槽130的入口向上述弧形的分配通道开放,按一定量分配得到经过滤的血液成分,而各血清分配槽130的出口与对应的各试剂反应槽140连通;
上述多个试剂反应槽140,在边缘一侧分布形成多个槽,以供从上述血清分配槽130供应的血液成分和试剂进行反应。
另外,在本发明的多重系统中,连接上述血液接收槽110和上述血液过滤槽120的通道形成于离各槽的旋转中心最远的线上,以通过旋转离心力使血液的移送变得容易。
另外,在本发明的多重系统中,上述血清分配槽130和上述试剂反应槽140的连接通道相对于旋转方向形成一定的角度,以通过旋转离心力使血液的移送变得容易。

染色槽220,对血液样本进行染色试剂处理,对五种白血球进行染色,以对白血球进行鉴别,计算数量。

发明效果
使用本发明的同时进行生化检查和血液检查的多重系统及用于此的多重圆盘,具有可同时在一个装置上进行通过测量血清的生物化学成分的浓度诊断人体功能的生物化学检查、通过分离血液样本测量检查红血球或白血球的数量及变形的血液检查及通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查的免疫检查的效果。

下面,结合附图对本发明的同时进行生化检查和血液检查的多重系统的较佳实施例进行详细说明。

如图1至图5所示,本发明的同时进行生化检查和血液检查及/或免疫检查的多重系统,包括多重圆盘1、照明部2、摄像头部3、显示单元4及DB部5。
在此,多重圆盘1以旋转中心为准呈扇形分割形成生物化学检查部100和血液检查部200或免疫检查部300,通过旋转控制装置(未图示)以旋转中心C为中心进行旋转,由上部盖10和反应槽板20构成。
上部盖10为具备旋转中心C的圆盘形板,可在反应槽板20色上部拆卸式结合或与反应槽板20一体形成。在上部盖10上,在与旋转中心相邻的一定位置以贯通盖子的形式具备第一血液投入口11和第二血液投入口12,而在边缘一侧呈弧形以贯通盖子的形式分布形成多个试剂投入口13。
第一血液投入口11是向反应槽板20的血液接收槽110供应投入血液样本(全血)的孔,形成于在与反应槽板20结合时与血液接收槽110对应的位置。
另外,第二血液投入口12是向反应槽板20的染色槽220供应投入血液样本(全血)的孔,形成于在与反应槽板20结合时与染色槽220对应的位置。
另外,多个试剂投入口13是各向反应槽板20的多个试剂反应槽140供应投入血液样本(全血)的孔,形成于在与反应槽板20结合时与试剂反应槽140对应的位置。
反应槽板20是具备旋转中心C并具备固定尺寸的深度的上部开放的圆盘容器型板,以旋转中心C为准呈扇形分割形成通过测量血清的生物化学成分的浓度诊断人体功能的生物化学检查部100、通过分离血液样本测量检查红血球或白血球的数量及变形的血液检查部200及通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查的免疫检查部300,拆卸式结合于上部盖10的下部或与上部盖10一体形成。
此时,如图4所示,反应槽板20也只以旋转中心C为准呈扇形分割形成生物化学检查部100和血液检查部200。
生物化学检查部100是用于进行向内部投入血液样本(全血),只过滤血液中的血清成分之后,使其与各试剂进行反应,之后通过测量吸光度计算出浓度值的生物化学检查的构成,可由血清分配槽130、试剂反应槽140构成。
生物化学检查使血清内化学成分和检查试剂进行反应(10分钟左右),则发生显色反应,此时测量吸光度,利用Beer-Lambert定律将其换算为浓度,与正常值比较人体功能(肝功能、心脏功能、肾脏功能等)进行诊断。
血液接收槽110形成于与旋转中心C相邻的位置,且形成于与上部盖10的第一血液投入口11对应的位置,接收通过投入口供应的续页样本。在此,血液接收槽110以旋转中心的旋转轴为中心呈弧形形成为宜,与血液过滤槽120通过通道连通。
血液过滤槽120是用于过滤移动到的血液样本分离需要的血液成分的构成,以旋转中心的旋转轴为中心呈弧形形成,在槽内部具备过滤器以在血液中只分离血清。血液过滤槽120的入口侧与血液接收槽110通过通道连通,而出口侧与向旋转中心外侧呈弧形形成的分配通道连接。
此时,连接血液接收槽110和血液过滤槽120的通道形成于离各槽的旋转中心最远的线上为宜,以通过旋转离心力使血液的移送变得容易。
血清分配槽130是从血液过滤槽120按一定量分配得到作为经过滤的血液成分的血清的多个槽,各血清分配槽130的入口向与血液过滤槽120的出口侧连通的弧形的分配通道开放,而各血清分配槽130的出口与对应的各试剂反应槽140通过通道连通。
试剂反应槽140是用于使所供应的血液成分和试剂进行反应的槽,多个槽呈弧形分布形成,且形成于与上部盖10的试剂投入口13对应的边缘一侧位置,以供应获得从血清分配槽130移送的作为经过滤的血液成分的血清,并供应得到通过试剂投入口13投入的试剂。此时,可在各试剂反应槽140中分注适合于要测量的检查项目的干式试剂之后,干燥固定。
此时,血清分配槽130和试剂反应槽140的连接通道相对于旋转方向形成一定的角度为宜,以通过旋转离心力使血液的移送变得容易。通过上述结构,在多重圆盘1旋转时,血清分配槽130的血液成分容易移送至试剂反应槽140。
此时,若在规定时间内以固定rpm旋转多重圆盘1,则供应至血液接收槽110的血液样本通过离心力移送至血液过滤槽120,而在血液过滤槽120过滤的血液成分通过离心力分配至血清分配槽130,从各血清分配槽130移送至各试剂反应槽140。
血液检查部200是用于通过分离血液样本测量检查红血球或白血球的数量及变形的构成,可由毛细管搁置槽210和染色槽220构成。
血液检查是以获得对患者的全面健康状态的信息为目的,在诊断检查中最广泛使用的检查,当随血流循环的白血球、红血球、血小板等个细胞的数量异常高或低,则表示存在疾病。
毛细管搁置槽210用于搁置装有血液样本的毛细管(未图示),搁置的装有血液样本的毛细管通过旋转的多重圆盘1的离心力被离心分离,从管的下部划分为红血球(RBC)、白血球(WBC)和血小板(Platelet)、血浆。
此时,若将多重圆盘1以4,000rpm旋转10分钟,或以10,000rpm旋转5分钟,则血液样本内的各成分通过离心力被划分区分。
采集分离的图像测量不同区间的长度,以容积量计算出整体百分比,从而计算红血球、白血球和血小板等血球数量。通常而言,若以正常的血液样本100%为准,则红血球约为45%,白血球和血小板约小于1%,而血浆约为55%。
在本发明中,将毛细管搁置槽210分为五个但非限制,可根据搁置的毛细管的尺寸改变槽的数量。
染色槽220是用于对血液样本进行染色试剂处理,对五种白血球进行染色,以对白血球进行鉴别,计算数量的槽,形成于与上部盖10的第二血液投入口12对应的位置,以供应得到通过第二血液投入口12投入的血液样本(全血)。染色液可预先具备于槽内或通过第二血液投入口12获得供应。
通常而言,五种白血球(WBC)为淋巴球(Lymphocyte)、单核细胞(Monocyte)、嗜中性白细胞(Neutrophil)、嗜酸性白细胞(Eosinophil)、嗜碱性白细胞(Basophil),而染色主要是利用吉姆沙染法(Giemsa stain)进行染色。当然,染色不限于吉姆沙染法.
吉姆沙溶液作为色素包含AzureⅡ-Eosin azureⅡ-eosin和AzureⅡ,利用固定甲醇之后用磷酸缓冲液稀释的吉姆沙溶液进行染色,或利用连甲醇固定的不需要的赖特(Wright)-吉姆沙溶液进行染色。
在利用吉姆沙溶液进行染色时,通常红血球呈粉红色,血小板呈淡粉色,淋巴球细胞质呈天蓝色,单核细胞细胞质呈浅蓝色,白血球核染质呈紫红色。
免疫检查部300是用于通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查的槽,进行在底面对要检查的DNA切面或抗体(抗原)上标记标识并去污之后进行固定,测量该抗体(抗原)的量的免疫检查。
免疫检查部300可具备离心分离通过另外的投入口投入的血液样本,只提供经分离的血清的结构(未图示),或可具备与具备于生物化学检查部100的分配通道连通,以只提供作为经过滤的血液成分的血清的结构。
本发明的多重圆盘1将上部盖10和反应槽板20都用透明材质形成,从而在反应结束之后,容易利用摄像头部3采集图像或测量吸光度。
照明部2可由光源2a和滤光器2b构成,通过单色仪(monochrometer)等滤光器从光源向多重圆盘1照射所需要的单色光。
摄像头部3是高分辨率的CCD摄像头或CMOS摄像头,设置于多重圆盘1的上部,在多重圆盘1结束反应之后,测量吸光度或采集图像传送至显示单元4。
此时,为测量吸光度,还可具备分光光度计等吸光度测量单元。
可利用发生显色反应的测得的吸光度计算出血清成分的浓度值,可从采集的图像资料进行血球的含量、白血球鉴别及计算数量。
此时,在摄像头部还可具备光学及/或荧光镜头,以进行对具有超微细结构的对象或与荧光色素结合的对象的精密的测量。
显示单元4是用于显示从摄像头部3传送得到的吸光度及采集的图像资料的装置,而DB部5是与摄像头部3及显示单元4联动,保存测得的资料或保存与保存的标准资料进行比较的资料,并将其提供至显示单元4的装置。
在上述构成的同时进行生化检查和血液检查的多重系统中,可选择性地同时进行生物化学检查和血液检查、生物化学检查和血液检查及免疫检查。
参照附图并以实施例为参考说明了本发明,但这只不过是例示性的,本领域的普通技术人员应当理解,由此可以进行多种变形及均等的实施例。本发明的真正的技术保护范围应由所附的权利要求书的技术思想来确定。

1.一种同时进行生化检查和血液检查的多重系统,包括以旋转中心为准呈扇形划分形成生物化学检查部(100)及血液检查部(200)的多重圆盘(1)。
2.根据权利要求1所述的同时进行生化检查和血液检查的多重系统,其特征在于:上述多重圆盘(1)还具备免疫检查部(300),从而还同时进行免疫检查。
3.根据权利要求1所述的同时进行生化检查和血液检查的多重系统,其特征在于:

在上述反应槽板(20)上,包括:生物化学检查部(100),通过测量血清的生物化学成分的浓度诊断人体功能;血液检查部(200),通过分离血液样本测量检查红血球或白血球的数量及变形;及免疫检查部(300),通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查。
5.根据权利要求1所述的同时进行生化检查和血液检查的多重系统,其特征在于:

血液接收槽(110);
血液过滤槽(120),与上述血液接收槽(110)连通;
多个血清分配槽(130),通过弧形的分配通道与上述血液过滤槽(120)连通;及

6.根据权利要求5所述的同时进行生化检查和血液检查的多重系统,其特征在于:
上述血液接收槽(110)在与旋转中心相邻的位置以旋转轴为中心呈弧形形成,以接收所供应的血液样本;
上述血液过滤槽(120)以旋转中心的旋转轴为中心呈弧形形成,在内部具备过滤器,以过滤移送到的血液样本;
上述多个血清分配槽(130),因各血清分配槽(130)的入口向上述弧形的分配通道开放,按一定量分配得到经过滤的血液成分,而各血清分配槽(130)的出口与对应的各试剂反应槽(140)连通;
上述多个试剂反应槽(140),在边缘一侧分布形成多个槽,以供从上述血清分配槽(130)供应的血液成分和试剂进行反应。
7.根据权利要求5所述的同时进行生化检查和血液检查的多重系统,其特征在于:
连接上述血液接收槽(110)和上述血液过滤槽(120)的通道形成于离各槽的旋转中心最远的线上,以通过旋转离心力使血液的移送变得容易。
8.根据权利要求5所述的同时进行生化检查和血液检查的多重系统,其特征在于:
上述血清分配槽(130)和上述试剂反应槽(140)的连接通道相对于旋转方向形成一定的角度,以通过旋转离心力使血液的移送变得容易。
9.根据权利要求1所述的同时进行生化检查和血液检查的多重系统,其特征在于:
上述血液检查部(200),包括:
毛细管搁置槽(210),搁置装有血液样本的管;及
染色槽(220),对血液样本进行染色试剂处理,对五种白血球进行染色,以对白血球进行鉴别,计算数量。
10.根据权利要求2所述的同时进行生化检查和血液检查的多重系统,其特征在于:
上述免疫检查部(300)具备离心分离通过另外的投入口投入的血液样本,只提供经分离的血清的结构,或具备与具备于上述生物化学检查部(100)的分配通道连通,以只提供作为经过滤的血液成分的血清的结构。
11.根据权利要求1所述的同时进行生化检查和血液检查的多重系统,其特征在于:
上述多重系统,还包括:
照明部(2),向上述多重圆盘(1)照射光;
摄像头部(3),拍摄上述多重圆盘(1);
显示单元(4),显示从上述摄像头部(3)传送得到的资料;及
DB部(5)。
12.根据权利要求11所述的同时进行生化检查和血液检查的多重系统,其特征在于:
上述照明部(2)包括光源(2a)和滤光器(2b),向上述多重圆盘(1)照射单色光;
上述摄像头部(3)设置于上述多重圆盘(1)的上部,在上述多重圆盘(1)结束反应之后,测量吸光度或采集图像传送至上述显示单元(4);
上述显示单元(4)显示从上述摄像头部(3)传送得到的吸光度及图像资料;
上述DB部(5)与上述摄像头部(3)及上述显示单元(4)联动,保存测得的资料或保存与保存的标准资料进行比较的资料。
13.一种多重圆盘,包括:
生物化学检查部(100),通过测量血清的生物化学成分的浓度诊断人体功能;
血液检查部(200),通过分离血液样本测量检查红血球或白血球的数量及变形;及
免疫检查部(300),通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查。
14.根据权利要求13所述的多重圆盘,其特征在于:
上述多重圆盘,包括:
上部盖(10),在与旋转中心相邻的位置具备第一血液投入口(11)和第二血液投入口(12),在边缘一侧呈弧形分布形成多个试剂投入口(13);及
反应槽板(20),与上述上部盖(10)结合,包括:生物化学检查部(100),通过测量血清的生物化学成分的浓度诊断人体功能;血液检查部(200),通过分离血液样本测量检查红血球或白血球的数量及变形;及免疫检查部(300),通过测量血清的抗原-抗体反应诊断癌症或病毒并进行检查。
15.根据权利要求14所述的多重圆盘,其特征在于:
上述多重圆盘的生物化学检查部(100),包括:
血液接收槽(110),具备于与上述第一血液投入口(11)对应的位置;血液过滤槽(120),与上述血液接收槽(110)连通;多个血清分配槽(130),通过弧形的分配通道与上述血液过滤槽(120)连通;及多个试剂反应槽(140),与上述多个血清分配槽(130)连通并具备于与上述试剂投入口(13)对应的位置;
上述多重圆盘的血液检查部(200),包括:
毛细管搁置槽(210),搁置装有血液样本的管;及染色槽(220),对血液样本进行染色试剂处理,对五种白血球进行染色,以对白血球进行鉴别,计算数量;
上述多重圆盘的免疫检查部(300),包括:
具备离心分离通过另外的投入口投入的血液样本,只提供经分离的血清的结构,或具备与具备于上述生物化学检查部(100)的分配通道连通,以只提供作为经过滤的血液成分的血清的结构。
