Abstract
The invention provides a disinfectant containing amines and quaternary ammonium salts and a preparation method thereof. The disinfectant containing amine and quaternary ammonium salt comprises 5-30% of quaternary ammonium salt, 1-10% of amine compound, 1.5-10% of corrosion inhibitor, 0.51-2% of surfactant, 0.5-6% of chelating agent and the balance of pure water. The preparation method comprises the following steps: weighing quaternary ammonium salt, amine compound, corrosion inhibitor, surfactant, chelating agent and pure water according to the formula, dissolving the quaternary ammonium salt and the amine compound in the pure water, stirring, then adding the corrosion inhibitor and the surfactant to dissolve the quaternary ammonium salt and the amine compound uniformly, adding the chelating agent, and stirring. The disinfectant containing amines and quaternary ammonium salt prepared by the invention has good disinfection and sterilization effects, short sterilization time, stable performance and low corrosion to equipment.
Description
Technical Field
The invention relates to the field of disinfectants, in particular to an amine and quaternary ammonium salt containing disinfectant and a preparation method thereof.
Background
The quaternary ammonium salt disinfectant has the advantages of stable performance, small toxicity irritation, quick sterilization effect, low use concentration, no bad smell and the like, has good biodegradability, is an environment-friendly disinfectant, and is deeply concerned by disinfection students. The quaternary ammonium salt disinfectant has been developed in many stages, the bactericidal activity is continuously improved, but a long time is still needed for achieving the ideal disinfection effect. In addition, the quaternary ammonium salt disinfectant is sensitive to metal ions such as calcium and magnesium, the calcium and magnesium ions are easy to form a quaternary ammonium salt compound with lower activity and poorer solubility with quaternary ammonium salt cations, and the bactericidal activity of the quaternary ammonium salt disinfectant is reduced. When hard water is used for preparing quaternary ammonium salt disinfectant or calcium and magnesium ions exist in a disinfection object in the using process, the disinfection effect is greatly influenced.
The sterilization mechanism of the amines and the derivatives thereof is mainly that substances with positive charges are formed in water, the substances are adsorbed on the surfaces of microorganisms with negative charges to form micelles, the micelles enter a lipid layer of a cell membrane through the osmosis effect to change the permeability of the membrane, and when the concentration of the substances reaches a certain degree, the formation of a non-double-layer structure of the cell membrane can cause the rupture of the cell membrane and the exosmosis of cell contents, so that the microorganisms die. In addition, the polyamine compound can change the activity of various enzymes, regulate the change of related gene expression level and obstruct the metabolism of cells, thereby having better sterilization effect, but the polyamine compound has low stability as a disinfectant and has poor effect of inhibiting the corrosion of acid in industrial cleaning.
Disclosure of Invention
The invention aims to provide the disinfectant containing the amines and the quaternary ammonium salt and the preparation method thereof, and the disinfectant has the advantages of good disinfection and sterilization effect, short sterilization time, stable performance, low corrosion to equipment, no residue and no toxicity.
In order to achieve the purpose, the invention provides an amine and quaternary ammonium salt-containing disinfectant, which comprises, by mass, 5-30% of quaternary ammonium salt, 1-10% of an amine compound, 1.5-10% of a corrosion inhibitor, 0.51-2% of a surfactant, 0.5-6% of a chelating agent, and the balance of water.
Preferably, the disinfectant containing amine and quaternary ammonium salt comprises, by mass, 10-25% of quaternary ammonium salt, 2-8% of amine compound, 1.5-5% of corrosion inhibitor, 0.51-1% of surfactant, 1-3% of chelating agent, and the balance of water.
Preferably, the quaternary ammonium salt is selected from one or two of benzalkonium chloride and didecyldimethyl ammonium bromide.
Preferably, the amine compound is dodecyl dipropylene triamine. The dodecyl dipropylene triamine can change the activity of various enzymes, regulate the change of related gene expression level and obstruct the metabolism of cells, so the dodecyl dipropylene triamine has better bactericidal effect, certain hard water resistance and insensitivity to metal ions such as calcium, magnesium and the like.
Preferably, the corrosion inhibitor is one or two of urotropin and sodium nitrite. Urotropine (hexamethylenetetramine) is used as a corrosion inhibitor, is nontoxic, can be well dissolved in quaternary ammonium salt disinfectant, can have synergistic sterilization and bacteriostasis effects with quaternary ammonium salt, and can improve the stability of the product.
Preferably, the surfactant is an amphoteric surfactant.
Preferably, the surfactant is an Amphoteric solubilizer, Tomamine Amphoteric 400 (surfactant a) and a highly efficient acid preservative, Tomamine Inhibitor 60S (surfactant B). The surfactant A can be compatible with anionic, nonionic and cationic systems, is beneficial to dissolving other surfactants into a formula system, and has low foam and high stability. In addition, the amphoteric solubilizer can be adsorbed on the surfaces of liquid and solid, and has the capability of reducing the free energy and surface tension of the surface, thereby having good wetting property. The surfactant B is easy to rinse, can spread a prepared sample on a solid surface, enables an air/solid interface to be replaced by a solid/liquid interface, and simultaneously spreads the air/liquid interface, so that the surfactant B has excellent wetting performance, can be used for metal pretreatment, can enable metal to have excellent anti-corrosion performance when contacting with organic acid and inorganic acid, and has low corrosion to production equipment.
Preferably, the chelating agent is one or more of EDTA, NTA, HEDP, ATMP or sodium salt thereof.
The invention provides a preparation method of a disinfectant containing amine and quaternary ammonium salt, which comprises the steps of weighing quaternary ammonium salt, amine compound, corrosion inhibitor, surfactant, chelating agent and pure water according to the formula, dissolving quaternary ammonium salt and amine compound in pure water, stirring, adding corrosion inhibitor and surfactant to dissolve uniformly, adding chelating agent, and stirring.
The disinfectant containing amine and quaternary ammonium salt comprises 5-30% of quaternary ammonium salt, 1-10% of amine compound, 1.5-10% of corrosion inhibitor, 0.51-2% of surfactant and 0.5-6% of chelating agent in percentage by mass. The quaternary ammonium salt and the amine compound have obvious synergistic bactericidal effect, and a proper amount of the quaternary ammonium salt has good bactericidal and disinfecting effects and higher stability. The addition of the amine compound increases the sterilization effect, enhances the hard water resistance of the disinfectant, and reduces the influence of calcium and magnesium ions possibly existing in a sterilized object on the sterilization effect.
Detailed Description
In order to explain the technical content of the present invention in detail, the following examples are further illustrated, but the present invention is not limited thereto. The reagents and materials used in the examples and comparative examples of the present invention are commercially available products or products that can be produced by a known method.
The first, examples 1-7 and comparative examples 1-2 are given below.

The preparation methods of examples 1 to 7, among others, were:
(1) weighing quaternary ammonium salt, amine compound, corrosion inhibitor, surfactant, chelating agent and pure water according to the formula in Table 1;
(2) dissolving quaternary ammonium salt and amine compound in pure water, and stirring uniformly;
(3) adding the corrosion inhibitor and the surfactant, and stirring to dissolve the corrosion inhibitor and the surfactant uniformly;
(4) adding a chelating agent, and uniformly stirring;
(5) adding pure water, stirring uniformly, and filtering to obtain the disinfectant containing amine and quaternary ammonium salt.
Next, the disinfectant of comparative examples 1-2 was prepared by the following method:
(1) weighing the raw materials according to the formula in the table 1;
(2) dissolving quaternary ammonium salt or/and amine compound in pure water, and stirring uniformly;
(3) adding the corrosion inhibitor and the surfactant, and stirring to dissolve the corrosion inhibitor and the surfactant uniformly;
(4) adding a chelating agent, and uniformly stirring;
(5) adding pure water, stirring uniformly, and filtering to obtain the corresponding disinfectant.
Second, the disinfectant solutions prepared in examples 1 to 7 and comparative examples 1 to 2 and a commercially available quaternary ammonium salt disinfectant solution were further examined.
1. Stability test
1.1 appearance
In examples 1-7, the newly configured amine-containing and quaternary ammonium salt-containing disinfecting solution was stirred for 30min to give a colorless transparent appearance; after 90 days of storage at 37 ℃, the appearance is clear and transparent, and no color change and no sediment and suspended matters exist.
1.2 determination of the content of active ingredients in disinfectant
The stability of the disinfectant is carried out according to the requirements related to the shelf life experiment of products in the technical Specification for disinfection (2002 edition). 5 samples from each group were stored at 37 ℃ for 90 days, and the content of the components was measured to obtain an average value. The stability of the disinfectant of examples 1-7 was reflected by the variation of the contents of quaternary ammonium salt and dodecyldipropylenetriamine, and the results are shown in Table 2.

The experimental result shows that the disinfectant in the examples 1-7 is stored for 90 days at 37 ℃, the content reduction rate of the quaternary ammonium salt and the dodecyl dipropylene triamine is lower than 5%, the requirement that the content reduction rate of the active ingredients is lower than 10% is met, and the shelf life of the product can reach two years; the stability of the amine compound in comparative example 1 is poor; the comparative examples 1-2 and the commercial quaternary ammonium salt disinfectant have lower stability than the disinfectant in examples 1-7. In addition, from the test results of examples 5 to 7, it is understood that the stability of the disinfectant is best when two surfactants, i.e., an Amphoteric solubilizer, Tomamine Amphoteric 400 (surfactant a) and a highly effective acid preservative, Tomamine Inhibitor 60S (surfactant B), are present in the system.
2. Comparison of Metal Corrosion Properties of disinfectant solutions
Metal corrosion tests were performed as required by the Disinfection Specification (2002 edition).
Metal sheet: stainless steel, carbon steel, copper, aluminum.
The corrosion tests of the disinfectant solutions prepared in examples and comparative examples and commercially available disinfectant solutions on stainless steel, carbon steel, copper and aluminum were carried out, metal pieces were immersed in the disinfectant solutions for 72 hours, the metal corrosion rates were calculated and the corrosion was rated. The results of the experiment are shown in table 3.

The experimental results show that the metal corrosion resistance of the disinfectant containing amine and quaternary ammonium salt in the examples 1-7 is obviously better than that of the disinfectant containing quaternary ammonium salt on the market and the disinfectant in the comparative examples 1-2, and the disinfectant has more obvious difference on copper corrosion. From the results of examples 5 to 7, it is clear that the presence of a highly effective acid corrosion inhibitor in the system contributes to the increase in the corrosion preventing performance of the metal.
3. Detection of sterilization effect of disinfectant
The disinfectant solutions prepared in the examples and comparative examples and the commercially available disinfectant solutions were used according to the requirements of the experimental methods in the technical Specification for Disinfection (2002 edition), and a quantitative sterilization experiment using a suspension was carried out. The indicator bacteria are: fourth generation of E.coli (ATCC 25922); fourth generation of staphylococcus aureus (ATCC 6538); pseudomonas aeruginosa (ATCC 15442) fifth generation; candida albicans (ATCC 10231) fifth generation. Neutralizing agent: 10g/L glycine +5g/L lecithin +80g/L Tween 80 in phosphate buffer. The results of the experiments are shown in tables 4-7.

As can be seen from tables 4-7, the disinfecting and killing effects of the disinfectant containing amine and quaternary ammonium salt in examples 1-7 are better than those of the disinfectant containing quaternary ammonium salt on the market and the disinfectant in comparative examples 1-2, and the sterilizing time can be shortened to 1 min. From examples 2 and 4, it is clear that the presence of benzalkonium chloride and didecyldimethyl ammonium bromide in the system provides better disinfecting and faster disinfecting action than the presence of didecyldimethyl ammonium bromide alone. From examples 5 to 7, it is understood that the disinfecting effect of the disinfecting liquid is better when both surfactants are present in the system.

Claims (2)
1. An amine-containing and quaternary ammonium salt disinfectant comprises, by mass, 5-30% of quaternary ammonium salt, 1-10% of dodecyl dipropylene triamine, 1.5-10% of a corrosion Inhibitor, 0.51-2% of an Amphoteric surfactant, 0.5-6% of a chelating agent and the balance of water, wherein the quaternary ammonium salt is selected from one or two of benzalkonium chloride and didecyl dimethyl ammonium bromide, the corrosion Inhibitor is one or two of urotropine and sodium nitrite, the Amphoteric surfactant is Tomamine Amphoeric 400 and Tomamine Inhibitor 60S, and the chelating agent is one or more of EDTA, NTA, HEDP, ATMP or sodium salts thereof.
2. The method for preparing the disinfectant containing amine and quaternary ammonium salt according to claim 1, wherein the quaternary ammonium salt, dodecyl dipropylene triamine, the corrosion inhibitor, the surfactant, the chelating agent and the pure water are weighed according to the formula; dissolving quaternary ammonium salt and dodecyl dipropylene triamine in pure water, and stirring; adding a corrosion inhibitor and an amphoteric surfactant to uniformly dissolve the components; adding chelating agent and stirring.
