Abstract

The present invention relates to a composition for preventing or improving hair loss comprising an extract of a glutinous millet of the Jizhou island. In particular, the present invention relates to a cosmetic composition comprising a Jizhou island sticky cereal extract as an active ingredient for preventing or improving hair loss by increasing proliferation of dermal papilla cells.

Technical Field
The present invention relates to a composition for preventing or improving hair loss comprising an extract of a glutinous millet of the Jizhou island. In particular, the present invention relates to a cosmetic composition comprising a Jizhou island sticky cereal extract as an active ingredient for preventing or improving hair loss by increasing proliferation of dermal papilla cells.
Background
The dermal papilla cells are key cells responsible for hair development and growth and are located at the very bottom of the hair root, i.e., the root of the subcutaneous hair. In fact, when dermal papilla cells are harvested from the scalp for culturing and then transplanted, new hair grows. However, there are many difficulties in using dermal papilla cells as a therapeutic agent for alopecia. First, it is difficult to separate from the scalp, the culture conditions are severe, and it is difficult to culture a sufficient amount of cells. In addition, there is a limit in that the hair regenerating ability is remarkably reduced in a large amount of culture.
Since human hair has a certain hair growth period (hair growth cycle), a certain number of hairs are always maintained. However, as hair loss progresses, the dermal papilla at the root becomes smaller, and as the dermal papilla becomes smaller, the thickness of hair becomes thinner, and at the same time, the hair growth cycle becomes shorter. Therefore, as hair loss progresses, hair becomes very thin, the growth cycle of hair becomes shorter, and hair grows a little bit, and then falls off.
It is known that the causes of alopecia have not only genetic causes and androgenic effects, but also complicated effects due to endocrine disorders, nutritional deficiencies, severe physical and psychological stress such as medication, labor, fever, surgery, etc. In recent years, not only male pattern alopecia but also pressure increases due to changes in dietary life, social environment, and the like, and the number of female alopecia increases and the age decreases.
Currently, the most commonly used hair loss therapeutic agents include finasteride (trade name: propecia), dutasteride (dutasteride; trade name: avodart), minoxidil (trade name: minoxyl or Rogaine), and the like. Finasteride and dutasteride are 5 alpha-reductase inhibitors (5 alpha-reductase inhibitor) that inhibit the conversion of androgens (testosterone) to 5 a-dihydrotestosterone (5 alpha-dihydrotestosterone, DHT). However, the product has side effects such as decreased sexual desire, erectile dysfunction, and loss of driving and performance. On the other hand, in the case of minoxidil, the mechanism has not been completely elucidated, but it is a potassium channel opener (potassium chaennel opener) that can hyperpolarize cell membranes (hyperpolarization), and it is thought that oxygen, blood, nutrients, etc. supplied to hair follicles are increased to health hair follicles by vasodilation and opening of potassium channels. However, the product also has side effects such as itching at the application site, erythema, skin irritation, eye irritation, etc., and unnecessary hair growth may occur in other parts of the body than the head. Because the side effects of these prior products increase anxiety in patients and in severe cases lead to rejection of the drug by patients, efforts are underway to develop drugs with fewer side effects.
On the other hand, jizhou island sticky grains are native sticky grains in Jizhou areas, are one of millet (SETARIA ITALICA), and are one of grains. Korean patent publication No. 10-2150927 discloses an anti-inflammatory effect, a skin moisturizing effect, a skin barrier strengthening effect and an atopy improving effect of the mixed extract of the Jizhou island sticky cereal, but does not disclose an alopecia improving effect of the extract of the Jizhou island sticky cereal.
Therefore, the present inventors have found that the Jizhou island sticky cereal extract has the effect of proliferating dermal papilla cells in the course of studying substances having few side effects and excellent hair loss prevention and treatment effects, thereby completing the present invention.

The invention may, however, be embodied in various forms and is not limited to the embodiments and examples described herein.
Throughout the specification, when a section "comprises" a structural element, it means that it can also include other structural elements, but does not exclude other structural elements, unless explicitly stated to the contrary.
The present invention relates to a cosmetic composition for preventing or improving hair loss comprising an extract of a glutinous millet of the Jizhou island.
The term "hair loss" in the present invention refers to a state where hair is not present at a portion where hair is normally present for any reason, and may include, but is not limited to, alopecia areata, hereditary androgenetic alopecia, telogen effluvium, traumatic alopecia, hair-plucking nodules, stress alopecia, anagen alopecia, pityriasis alopecia, syphilitic alopecia, seborrheic alopecia, symptomatic alopecia, scarring alopecia, or congenital alopecia.
The Jizhou island sticky cereal (SETARIA ITALICA Beauv.) of the present invention is one kind of millet, is a grain, is a primary sticky cereal grown in Jizhou areas, and has a variety name SAMDACHAL (see Korean variety and seed service center (KSVS) variety). In the present invention, jizhou island sticky valleys grown and harvested in Jizhou islands may be used, but are not limited thereto.
The Jizhou island sticky cereal extract of the present invention can exhibit an effect of preventing or improving hair loss by promoting proliferation of dermal papilla cells to induce the growth phase of hair.
"Preventing" according to the present invention means any act of inhibiting or delaying the occurrence of alopecia by administering the composition according to the present invention, and "ameliorating" means any act of at least reducing a parameter (e.g., severity of symptoms) associated with a condition treated by administering the composition comprising an extract according to the present invention.
The "extract" of the present invention may be obtained using conventional extraction solvents known in the art. As the extraction solvent, a polar solvent or a nonpolar solvent may be used. The polar solvent includes water, alcohols of C1 to C6 (e.g., methanol, ethanol, propanol, butanol, n-propanol, isopropanol, n-butanol, etc.), acetic acid, or a mixture of the above polar solvents. The nonpolar solvent comprises acetone, acetonitrile, ethyl acetate, methyl acetate, butyl acetate, fluoroalkane, hexane, diethyl ether, chloroform, dichloromethane or mixture of the above nonpolar solvents.
In one embodiment, the extract may be extracted using a solvent selected from the group consisting of water, alcohols of C1 to C6, acetic acid, and a mixed solvent thereof. Preferably, the above extract may be 0.01% to 90% ethanol extract or butanol extract, more preferably, 60% to 80% ethanol extract or butanol extract, and most preferably, 70% ethanol extract or butanol extract.
The extract of the present invention may be extracted by hot water extraction, cold dip extraction, reflux cooled extraction, ultrasonic extraction or conventional extraction methods known in the art.
The term "extract" as used in the present invention refers to a crude extract (crude extract) commonly used in the art, but also includes in a broad sense fractions obtained by subjecting the extract to additional fractionation (fractionation). That is, the extract includes not only those obtained using the above-mentioned solvents but also those obtained by additionally applying a purification process. For example, fractions obtained by passing the above extract through an ultrafiltration membrane having a certain molecular weight cut-off, separation by various chromatographs (specifically made for separation according to size, charge, hydrophobicity or affinity), and the like, fractions obtained by performing various additional purification methods are also included in the crude drug extract of the present invention.
The cosmetic composition of the present invention may be provided in the form of a solution, gel, solid or pasty anhydrous product, emulsion, suspension, microemulsion, microcapsule, microgranule, or ionic (liposome) or nonionic foam dispersion, cream, lotion, emulsion, cake, ointment, spray or concealer obtained by dispersing an oil phase in an aqueous phase. And may also be prepared in the form of a foam (foam) or an aerosol composition further containing a compressed propellant.
Further, the above-mentioned cosmetic composition may contain fatty substances, organic solvents, solubilizing agents, concentrating agents and gelling agents, softening agents, antioxidants, suspending agents, stabilizers, foaming agents (foaming agents), fragrances, surfactants, water, ionic or nonionic emulsifiers, fillers, sequestering and chelating agents, preserving agents, vitamins, blocking agents, wetting agents, essential oils, dyes, pigments, hydrophilic or lipophilic activators, lipid vesicles or any other ingredient commonly used in the cosmetic field such as those commonly used in cosmetics.
The cosmetic composition of the present invention may be, for example, a hair cosmetic.
The above-mentioned cosmetic composition for hair is formulated into any one dosage form selected from the group consisting of shampoo, conditioner, hair tonic, hair conditioner, essence, scalp care agent, hair nourishing emulsion, but there is no particular limitation in formulation. In addition, in each dosage form, other ingredients than the above-described essential ingredients may be appropriately selected depending on the type of other external preparations or the purpose of use.

Example 1
Preparation of the extract
Preparing 70% ethanol extract, hot water extract, gu Jiwan extract, gu Yisuan ethyl ester extract, chloroform extract, butanol extract, 70% ethanol extract and hot water extract of Jizhou island glutinous millet.
Example 2
Culture of dermal papilla cells of white rat beard
Use a kit containing 100units/mL penicillin-100 μg/mL streptomycin (100 units/ML PENICILLIN-100 μg/mL streptomycin, gibco Inc, N.Y. U.S.) and 10% heat-inactivated fetal bovine serum (10% heat-INACTIVATED FETAL bone serum, FBS); gibco Inc, N.Y.) medium, immortalized dermis papilla cells isolated from white rat beard (Rat vibrissa immortalized DERMAL PAPILLA CELL: FILSELL ET al., 1994) were cultured in a 5% CO 2 incubator at 37℃and subcultured every three days.
Example 3
Proliferation effect of Jizhou island Mugu 70% ethanol (EtOH) extract on dermal papilla cells
Proliferation of immortalized dermal papilla cells (Rat vibrissa immortalized DERMAL PAPILLA CELL) isolated from white rat beard was determined using the MTT assay (MTT assay). Dermal papilla cells (1.0X10 4 cells/mL) were suspended in DMEM medium containing 1% FBS and incubated in 96-well plates (96 well plates) for 24 hours.
Cells were treated with 70% ethanol extract from Jizhou island sticky valleys at concentrations of 1, 10 and 25 μg/mL. As a positive control, cells were treated with minoxidil (Minoxidil, sigma, misu, U.S.A.) at a concentration of 10. Mu.M. After 48 hours of incubation, 10 μl of WST dye (Daeil Lab Service, korean head) was added and reacted for 3 hours. Absorbance was measured at 450nm using a VERSAmax microplate reader (microplate reader, molecular Devices, california, usa). Average absorbance values of the sample groups were obtained and the extent of proliferation was examined by comparison with absorbance values of the control group, as shown in table 1 and fig. 1 below.

Example 4
Proliferation effect of Jizhou island adhesive Gu Jiwan (hexane) extract on dermal papilla cells
The proliferation effect of Jizhou island adhesive Gu Jiwan extract on dermal papilla cells was tested in the same manner as in example 3, as shown in table 2 and fig. 2 below.

As shown in Table 3 and FIG. 3 above, increased proliferation of dermal papilla cells was confirmed when treated with Jizhou island Muku Valley EtOAc extract at concentrations of 1, 10 and 25 μg/mL. The proliferation effect of chloroform extract of Murray Valley grain in Jizhou island on dermal papilla cells was examined in the same manner as in example 3, as shown in Table 4 and FIG. 4 below.

1. A cosmetic composition for preventing or ameliorating hair loss, comprising a zizhou island sticky cereal extract.
