Abstract
The invention discloses a composition for improving or promoting whey protein absorption, which comprises the following raw materials in parts by weight: whey protein, vitamin D3 and black pepper extract, wherein the black pepper extract is obtained using a specific extraction process, as well as formulations and sports nutritional foods for promoting muscle synthesis comprising the composition for improving or promoting whey protein absorption. According to the composition for improving or promoting absorption of whey protein of the present invention, whey protein, vitamin D3 and black pepper extract are compounded in a specific ratio. The test results prove that: the three extracts of the whey protein, the vitamin D3 and the black pepper extract are compounded according to a certain proportion, so that the muscle increasing effect can be obviously improved, and the composition has a synergistic effect, so that the composition has an excellent muscle increasing effect.

Technical Field
The invention belongs to the field of sports nutrition food, and relates to a composition for improving or promoting whey protein absorption, in particular to a composition for improving or promoting whey protein absorption, which comprises whey protein, vitamin D3 and black pepper extract, a preparation method thereof, a preparation containing the composition for improving or promoting whey protein absorption, and application of the composition for improving or promoting whey protein absorption and the preparation containing the composition for improving or promoting whey protein absorption in preparing a muscle-increasing sports nutrition product.
Background
Muscles are critical to the health of the body, and there is a need to promote muscle mass, increase and maintain muscle mass, both for sports people and the general public. Exercise in combination with rational nutritional supplementation is an effective way to promote muscle growth. The exercise is followed by a golden period of protein supplementation, which has a great impact on muscle production, metabolism and repair of injured muscles. At this time, the supplementation of high-quality protein can provide a large amount of essential amino acids, especially leucine, for the body, which not only can provide raw materials for muscle protein synthesis, but also can promote insulin secretion and stimulate muscle protein synthesis. Leucine also directly stimulates mTOR (a key factor in protein synthesis) to initiate muscle protein synthesis.
Whey protein is a protein extracted from milk by using a modern production process technology, has the characteristics of being rich in essential amino acids and leucine, low in fat content and high in digestibility, and is a protein supplement commonly used by sports people. The strenuous exercise may lead to a decrease in digestive function of the human body, which may be related to the reasons such as dehydration caused by exercise, insufficient blood flow in the digestive system caused by a large concentration of blood in skeletal muscle. Although whey protein is an easily digestible protein, there are still cases where people eat whey protein after exercise to suffer from dyspepsia, abdominal distension, etc.
At present, the components of the muscle-increasing nutritional food for the sports crowd are complex, but the muscle-increasing effect of the whey protein with a single component is not ideal. Therefore, how to develop a sports nutritional food which is used for the crowd of the muscle building exercise to bring new choices of sports nutritional food, has relatively simple ingredients and obvious muscle building effect, and is a technical problem to be solved urgently.
Disclosure of Invention
The invention aims to provide a composition which can promote digestion and absorption of whey protein in intestinal tracts, has the functions of rapidly improving the concentration of leucine and essential amino acids in blood, can stimulate and promote muscle synthesis of human bodies, and does not cause damage to the health of the human bodies. Compositions that improve or promote whey protein absorption are specifically designed for the sports population.
In order to achieve the object of the present invention, the novel composition for improving or promoting whey protein absorption according to the present invention is composed of the following raw materials in parts by weight: 30-98 parts of whey protein, 30.001-0.06 part of vitamin D and 0.002-2 parts of black pepper extract.
The preparation method of the black pepper extract comprises the following steps:
pulverizing fructus Piperis, reflux-extracting with 95% ethanol of 6 times weight of fructus Piperis for 2 times each for 1.5 hr, filtering, mixing filtrates, vacuum concentrating the filtrate to paste to obtain paste crude extract, freeze-drying at-45deg.C, and pulverizing with ball mill to obtain paste crude extract powder;
washing the pasty crude extract powder with n-hexane washing solution at 25-60deg.C for 2 times, dissolving with 95% ethanol 4 times of the weight of fructus Piperis extract, filtering with active carbon, and vacuum concentrating to obtain concentrated filtrate;
freeze-drying the concentrated filtrate obtained by vacuum concentration at-15deg.C to obtain freeze-dried product, and grinding the freeze-dried product at-15deg.C to obtain fructus Piperis extract granule with average particle diameter of 15-30 μm.
More preferably, the novel composition for improving or promoting whey protein absorption according to the invention consists of the following raw materials in parts by weight: 60 to 96 portions of whey protein, 0.004 to 0.02 portion of vitamin D and 0.006 to 1 portion of black pepper extract.
Most preferably, the novel composition for improving or promoting whey protein absorption according to the invention consists of the following raw materials in parts by weight: 80 to 96 portions of whey protein, 0.002 to 0.013 portion of vitamin D and 0.01 to 0.5 portion of black pepper extract.
The starting whey protein in the composition for improving or promoting whey protein absorption according to the invention comprises concentrated whey protein, isolated whey protein and/or hydrolysed whey protein, all commercially available as vitamin D3.
According to the preparation method of the black pepper extract, the dosage of the extraction solvent is calculated according to the mass-volume ratio of the extraction solvent to the black pepper, and the unit can be g/mL. The percentages of the ethanol solution are expressed in the invention as volume percentages.
It is another object of the present invention to provide a method for preparing the composition for improving or promoting whey protein absorption, the method comprising: weighing the raw materials, sieving, and mixing.
It is a further object of the present invention to provide a formulation comprising the composition for improving or promoting whey protein absorption, the formulation comprising the composition for improving or promoting whey protein absorption and a pharmaceutically acceptable carrier or excipient.
Further, the formulation comprises 100 parts by weight of the composition for improving or promoting whey protein absorption and 0.01-3500 parts by weight of a pharmaceutically acceptable carrier or excipient.
The pharmaceutically acceptable carrier or excipient is preferably selected from one or more of xylitol, erythritol, sorbitol, isomalt, isomaltulose, white granulated sugar, ethanol, microcrystalline cellulose, lactose, maltodextrin, starch, hydroxypropyl methylcellulose, sodium carboxymethyl cellulose, magnesium stearate, sweetener, flavoring essence, etc.
The preparation according to the invention may be in the form of tablets, capsules, powders or granules.
The parts by weight in the present invention may be those known in the pharmaceutical field such as mu g, mg, g, kg.
It is a further object of the present invention to provide the use of said composition for improving or promoting whey protein absorption or a formulation comprising said composition for improving or promoting whey protein absorption in the preparation of a myotonic exercise nutritional food.
Advantageous effects
According to the novel composition for improving or promoting the absorption of whey protein of the present invention, whey protein, vitamin D3 and black pepper extract are compounded in a specific ratio. The whey protein is rich in essential amino acids and leucine, can provide a large amount of essential amino acids, especially leucine for organisms, can provide raw materials for muscle protein synthesis, and can promote insulin secretion and stimulate muscle protein synthesis. Leucine also directly stimulates mTOR (a key factor in protein synthesis) to initiate muscle protein synthesis. Vitamin D3 has a variety of biological effects on muscle and its synergistic effect with leucine in enhancing protein anabolism, potentially beneficial for muscle function.
Vitamin D3, when used alone, has little effect on muscle protein synthesis and muscle strength, but is effective in improving muscle status when taken in synergistically enriched amounts of essential amino acid proteins.
The black pepper extract not only can improve the bioavailability of various nutrients, but also can promote the absorption and utilization of substances such as proteins in three aspects of improving the activity of digestive enzymes, promoting the absorption of intestinal epithelial cells and enhancing metabolism through thermogenesis.
The test of the invention proves that: the three substances of whey protein, vitamin D3 and black pepper extract are compounded according to a certain proportion, so that the composition has a synergistic effect, and can promote muscle synthesis together, so that the composition has an excellent muscle increasing effect.

Detailed Description
Hereinafter, the present invention will be described in detail. Before the description, it is to be understood that the terms used in this specification and the appended claims should not be construed as limited to general and dictionary meanings, but interpreted based on the meanings and concepts corresponding to technical aspects of the present invention on the basis of the principle that the inventor is allowed to define terms appropriately for the best explanation. Accordingly, the description set forth herein is merely a preferred example for the purpose of illustration and is not intended to limit the scope of the invention, so that it should be understood that other equivalents or modifications may be made thereto without departing from the spirit and scope of the invention.
The composition for improving or promoting the absorption of whey protein according to the present invention employs three active ingredients of whey protein, vitamin D3 and black pepper extract, wherein the black pepper extract is treated with a specific process to most effectively and maximally retain key ingredients piperine in black pepper.
In the preparation method of the black pepper extract, 95% ethanol is firstly used for ethanol extraction treatment so as to extract key components piperine as far as possible, then the obtained pasty crude extract is freeze-dried at the temperature of minus 45 ℃, and then the obtained pasty crude extract is crushed by a ball mill to obtain crude extract powder. The freeze drying step can effectively protect piperine from being damaged in the middle of the later steps such as crushing and the like. Then dissolving with ethanol again, filtering with active carbon, and vacuum concentrating the filtrate to ensure the purity of piperine. Then freeze-drying again at-15 deg.C, and grinding at the same temperature to obtain black pepper extract particles with average particle diameter of 15-30 μm. The temperature in the second freeze-drying is not too low, because the piperine is subjected to the ethanol extraction treatment of the previous two ethanol solubilizations, the piperine purity is high and the integrity of the molecular structure is kept as much as possible, so the second freeze-drying temperature is not too low, and the proper lifting temperature is also beneficial to saving energy consumption.

The vitamin D3 content used in the invention is 100,000IU/g.
EXAMPLE 1 preparation of composition powders for improving or enhancing whey protein absorption
The preparation method of the composition powder for improving or promoting whey protein absorption of the embodiment comprises the following steps:
1. formulation of a composition for improving or promoting whey protein absorption: 98g of whey protein, 30.01g of vitamin D, 1g of black pepper extract, 0.9g of isomaltulose and 0.1g of edible essence.
Wherein, the black pepper extract is prepared according to the following method: pulverizing fructus Piperis, reflux-extracting with 95% ethanol for 2 times each for 1.5 hr, filtering, mixing filtrates, vacuum concentrating to obtain paste, lyophilizing at-45deg.C, pulverizing with ball mill to obtain coarse extract powder, dissolving with 95% ethanol at 25deg.C to 60deg.C, filtering with activated carbon, vacuum concentrating filtrate, lyophilizing again at-15deg.C, and grinding at-15deg.C to obtain fructus Piperis extract with average particle diameter of 15-30 μm.
2. Process for preparing compositions for improving or promoting whey protein absorption
1) The raw materials are respectively weighed according to the formula proportion in the step 1: whey protein, vitamin D3 and black pepper extract, and sieving with 40 mesh sieve respectively for use.
2) The auxiliary materials are respectively weighed according to the formula proportion: and uniformly mixing the raw materials and the auxiliary materials according to an equivalent progressive method and bagging.

EXAMPLE 2 preparation of composition granules for improving or enhancing whey protein absorption
The preparation method of the composition granule for improving or promoting whey protein absorption of the embodiment comprises the following steps:
1. formulation of a composition for improving or promoting whey protein absorption: 30g of whey protein, 30.06g of vitamin D, 0.01g of black pepper extract, 30g of maltodextrin and 40g of isomaltooligosaccharide.

1) The raw materials are respectively weighed according to the formula proportion in the step 1: whey protein, vitamin D3 and black pepper extract, and sieving with 40 mesh sieve respectively for use.
2) Weighing auxiliary materials maltodextrin and isomaltooligosaccharide according to a formula proportion, uniformly mixing the raw materials and the auxiliary materials according to an equivalent progressive method, using 50% ethanol as an adhesive to prepare a soft material, granulating the soft material in a swing granulator with a 14-mesh screen, placing the granules into a multifunctional boiling dryer, boiling and drying at 50 ℃ for about 40min, controlling the moisture of the granules to be 5%, and then finishing the granules, subpackaging and bagging the granules.

2) Respectively weighing isomaltulose and magnesium stearate according to the formula proportion, mixing the raw materials and the auxiliary materials (except magnesium stearate) uniformly by an equal progressive method, using ethanol with the volume percentage concentration of 50% as an adhesive to prepare a soft material, granulating the soft material in a swing granulator with a 14-mesh screen, putting the granules into a multifunctional boiling dryer, boiling and drying at 50 ℃ for about 40min, controlling the moisture of the granules to be 5%, finishing the granules, adding magnesium stearate into the granules, mixing uniformly, tabletting, coating a film coat and subpackaging.

42 male university students of Beijing sports university with exercise habits are selected as study objects. All subjects had no other sports nutritional supplements taken one month prior to the experiment. 1.2 dietary control and nutritional supplements
During the experiment, all subjects were asked to eat the same food at the same place and at the same time, and for 1 day 3 meals, caloric intake was calculated as 45 kcal per kg body weight per day, with caloric ratios of three major energy substances of 65% carbohydrate, 15% protein, 20% fat. The experimental diet is started one week before the experiment starts, and no drinking, caffeine intake, night cooking and other actions occur.
Nutritional products were taken daily after the start of the experiment, group a: whey protein, group B: whey protein + vitamin D3, protein content 75%, group C: whey protein + black pepper extraction, group D: powder of example 1, group E: powder of comparative example 1, group F: the powder of comparative example 2. The protein content of each group of nutrition is 75%, 40g of each nutrition is taken each time, and the nutrition is brewed by warm water, wherein the nutrition is taken 30min after the training is finished.
1.3 study methods:
1.3.1 Experimental design
The experiment is divided into two parts:
(1) study of the Effect of a single intake of whey protein composition on whey protein digestion and absorption and skeletal muscle protein synthesis rate
There was no vigorous exercise three days before the start of the test, and a fasted diet of 22:00 was started the day before the start of the test. Venous blood is then drawn every 30 minutes within 120 minutes, and every 60 minutes within 120 to 240 minutes. Muscle biopsy samples were taken at 120min and 240. Three muscle biopsy samples were taken from the mid-thigh area of the lateral muscle.
(2) Whether the whey protein composition provided by the invention can improve the body components of college students with exercise habits by combining with anti-exercise after 4 weeks of study and improve the muscle strength.
The subjects had a rest for one day after completion of experiment (1). The test measures height, weight and body composition. GYMAWARE is adopted for deep squat and horizontal pushing 1RM test, and the original underground squat and vertical jump (CMJ) height test of the force measuring platform is adopted.
Training plans are formulated according to the 1RM test result: squatting 70%1RM,4 groups, 6-8 times each, resting for 60 seconds between groups, and resting for 90 seconds after the 4 th group is completed; then 4 groups of 50%1RM squat exercises are performed, each group is 6-8 times, rest is performed for 60 seconds between groups, and rest is performed for 150 seconds after the 4 th group is completed; pushing 70% of 1RM horizontally, 6-8 times for each of 4 groups, resting for 60 seconds between groups, and resting for 90 seconds after the 4 th group is completed; finally, 4 groups of 50%1RM lying and pushing exercises are completed, each group has 6-8 times, rest is performed for 60 seconds between groups, and the total movement time of the acute resistance training is about 30 minutes. The subject is required to complete the range of motion joint activity each time.
Nutrient intake and training was continued for 4 weeks, with two days of rest after the end of the last training, with body composition, squat and push 1RM test and CMJ test.
1.3.2 test index
Serum insulin, EAA and leucine content were tested.
Determination of L- [ Cyclo-Rebaume ] in muscle protein and non-tissue amino acid pools (non-synthetic muscle protein amino acids) 13 C 6 ]Phenylalanine enrichment, the partial synthesis rate of muscle protein was calculated.
Testing the physical form index of the subject before and after the experimental intervention: height, weight, body composition.
Muscle strength index: squat and push 1RM test and CMJ test.
1.4 statistical analysis
All measured data are expressed as mean ± standard deviation (x±sd) and are counted and analyzed using SPS25.0 software. Each index between and within the group was analyzed by one-way variance analysis, with P <0.05 indicating significant level differences.
2 experimental results
2.1 Effect of intake of whey protein composition on serum leucine, essential amino acids and insulin
Figures 1-3, tables 2-4 show experimental results of the effect of intake of a composition for improving or promoting whey protein absorption according to the invention and the diet of a control group on serum leucine, essential amino acids and insulin levels, from which it can be seen that each subject's serum leucine, essential amino acids and insulin levels rise rapidly after intake of the nutritional product, whereas group D rises faster than the other groups. Serum leucine, essential amino acids and insulin levels were significantly higher in groups D, 30min and 60min than in groups a, B, C, E and F (P < 0.05).

Note that: tables 2 to 4 show significant differences (P < 0.05) compared to group a; # represents a significant difference (P < 0.05) from group B, a significant difference (P < 0.05) from group C, a significant difference (P < 0.05) from group E, and a significant difference (P < 0.05) from group F.
2.2 Effect of intake of whey protein composition on the partial Synthesis Rate of muscle proteins
The muscle protein Fraction Synthesis Rate (FSR) may represent the ability of a protein to stimulate postprandial muscle protein synthesis. As shown in table 5 and fig. 4, group D FSR was significantly higher than groups a, B, C, E and F (P < 0.05).

2.3 Effects of 4 week trial intervention on subject body composition
As shown in table 6, both body weight and lean body mass increased in the 4-group subjects after 4 weeks of experimental intervention. Group D subjects had significantly higher body weight and lean body mass gain than group a, group B, group C, group E and group F (P < 0.05).

2.4 Effects of 4 week trial intervention on subject squat and crouch pushing 1RM and CMJ longitudinal jump
As shown in tables 7 and 8, 4 week experimental intervention increased subjects squat and crouch pushing 1RM and longitudinal jump height. Group D subjects 1RM and the increase in elevation in longitudinal jump were significantly higher than those in groups a, B, C, E and F (P < 0.05).

the composition comprises the following raw materials in parts by weight: 30 to 98 parts of whey protein, 0.001 to 0.06 part of vitamin D3 and 0.002 to 2 parts of black pepper extract.

the composition comprises the following raw materials in parts by weight: 60 to 96 portions of whey protein, 30.004 to 0.02 portion of vitamin D and 0.006 to 1 portion of black pepper extract.
3. The composition for improving or promoting absorption of whey protein according to claim 1,
the composition comprises the following raw materials in parts by weight: 80 to 96 parts of whey protein, 30.002 to 0.013 part of vitamin D and 0.01 to 0.5 part of black pepper extract.
4. A composition for improving or promoting the absorption of whey protein as defined in any of claims 1 to 3,
the preparation method of the black pepper extract comprises the following steps:
pulverizing fructus Piperis, reflux-extracting with 95% ethanol of 6 times weight of fructus Piperis for 2 times each for 1.5 hr, filtering, mixing filtrates, vacuum concentrating the filtrate to paste to obtain paste crude extract, freeze-drying at-45deg.C, and pulverizing with ball mill to obtain paste crude extract powder;
washing the pasty crude extract powder with n-hexane washing solution at 25-60deg.C for 2 times, dissolving with 95% ethanol 4 times of the weight of fructus Piperis extract, filtering with active carbon, and vacuum concentrating to obtain concentrated filtrate;
freeze-drying the concentrated filtrate obtained by vacuum concentration at-15deg.C to obtain freeze-dried product, and grinding the freeze-dried product at-15deg.C to obtain fructus Piperis extract granule with average particle diameter of 15-30 μm.
5. A composition for improving or promoting the absorption of whey protein according to any of claims 1 to 3,
the whey proteins comprise concentrated whey proteins, isolated whey proteins and/or hydrolysed whey proteins.
6. A formulation comprising the composition for improving or promoting absorption of whey protein according to any of claims 1 to 3,
the formulation comprises the composition for improving or promoting whey protein absorption and a pharmaceutically acceptable carrier or excipient.
7. The formulation of claim 6, wherein,
the formulation comprising 100 parts by weight of the composition for improving or promoting whey protein absorption according to any one of claims 1 to 3 and 0.01 to 3500 parts by weight of a pharmaceutically acceptable carrier or excipient.
8. The formulation of claim 6, wherein,
the pharmaceutically acceptable carrier or excipient is selected from one or more of xylitol, erythritol, sorbitol, isomalt, isomaltulose, white granulated sugar, ethanol, microcrystalline cellulose, lactose, maltodextrin, starch, hydroxypropyl methylcellulose, sodium carboxymethyl cellulose, magnesium stearate, sweetener and edible essence.
9. The formulation of claim 6, wherein,
the preparation is in the form of tablet, capsule, powder or granule.
10. Use of a composition for improving or promoting the absorption of whey protein according to any of claims 1 to 3 or a formulation comprising a protein according to any of claims 6 to 9 for the preparation of a myotonic exercise nutritional product.
