Description

Anti-aging composition and application thereof in beauty and skin care
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to an anti-aging composition and application thereof in beauty and skin care.
Background
In the field of new medical beauty and skin care, radio frequency is a beauty technology which is eagerly pursued by market consumers in recent years. Radio frequency utilizes the switching of the polarity of an electric field to produce a thermal effect on the friction of the tissue through which the current passes, and since the conductivity of the dermis is highest in skin tissue, the current is mainly concentrated in the dermis layer, causing bulk volumetric heating of the dermis tissue. When the heating temperature reaches 60 ℃, the collagen is instantly contracted, and when the heating temperature reaches 42 ℃, the conformation of the collagen helical structure is changed, so that the self wound repair mechanism of an organism can be further started, fibroblasts are stimulated to generate new collagen, elasticity and synthesis of other supporting proteins, and the effects of tightening the skin, removing wrinkles, lifting, improving the skin quality and the outline and the like are achieved.
The objective of careful research on parameters such as radio frequency, instrument efficacy and radio frequency depth of action is to apply faster and greater energy to the dermis, but too high energy easily causes thermal damage to healthy cells and surrounding tissue cells, generates oxidative free radicals, and even directly affects the normal metabolism of the skin. In the case of hot marie radio frequency, the customer usually experiences a dry, red, swollen, scab-forming repair period of about 3-10 days after surgery.
In addition, the mechanism of action of the radiofrequency cosmetic device determines that fibroblasts in the dermis synthesize more extracellular matrix, but the newly synthesized extracellular matrix is not uniformly distributed. The reason for this is that fibroblasts are living and can sense and respond to mechanical force, and secreted collagen fibers can form stronger fiber bundles along the direction of the mechanical force to resist the mechanical force. The force non-uniformity is increasingly manifested, and the characters are commonly called Chinese, crow's feet, raised head, etc. As the age increases, wrinkles become more and more obvious, so that the problem of collagen arrangement is not solved only by using the radio frequency beauty instrument, but also the improvement effect on the wrinkles such as expression lines is limited.
Therefore, the anti-aging composition with good moisturizing, anti-oxidation and neurotransmitter conduction inhibiting effects is of great significance for exerting the anti-aging and beautifying effects of the radio frequency beautifying instrument.
Disclosure of Invention
In view of the above-mentioned deficiencies in the prior art, it is an object of the present invention to provide an anti-aging composition. The anti-aging composition provided by the invention adopts safe and non-toxic raw materials, has no side effect and is mild in formula. The anti-wrinkle composition is obtained by compounding the antioxidant, the composite polypeptide, the panthenol and the glycoside compound in a specific ratio, the anti-wrinkle performance of the composition can be remarkably improved, and cosmetics prepared from the anti-aging composition have the effects of resisting oxidation, preserving moisture for a long time, resisting aging, resisting bacteria and the like. The prepared cosmetic is used in combination with a radio frequency beauty instrument, can promote the regeneration of dermal collagen of skin, fill and fade fine lines, and achieve the effects of deep anti-aging, long-acting moisturizing, anti-wrinkle and skin nourishing.
The invention aims to provide an anti-aging composition, which comprises the following raw materials in parts by mass:
Figure BDA0003909721550000021
the antioxidant is at least one selected from arbutin, ergothioneine, grape seed extract, olive leaf extract, glabridin, retinol and its derivatives, and ascorbic acid and its derivatives;
the composite polypeptide is selected from at least one of arginine/lysine polypeptide, acetyl octapeptide-3, acetyl hexapeptide-8, pentapeptide-3, tripeptide-1 and acetyl hexapeptide-1;
the glycoside compound is at least one selected from glycerol glucoside, glucopyranoside, dextran and sodium hyaluronate.
Arbutin is also called arbutin, and is widely existed in plants of multiple families, easy to extract, artificially synthesized and low in toxicity. In cytological experiments, cell morphology observation and biochemical index detection are carried out, and RT-PCR is adopted to verify the relation between the arbutin and Grx-mRNA expression, so that arbutin has certain antioxidation, and meanwhile, the arbutin also has the effects of inhibiting the activity of tyrosinase in vivo, preventing the generation of melanin and sterilizing and diminishing inflammation.
Ergothioneine is a natural antioxidant, can protect cells in human body, is an important active substance in organism, and has multiple physiological functions of scavenging free radicals, whitening skin, resisting aging, preventing ultraviolet radiation damage, maintaining DNA synthesis and normal cell growth, etc.
The grape seed extract is a novel high-efficiency natural antioxidant substance which can not be synthesized in a human body and is extracted from grape seeds. It can effectively remove redundant free radicals in human body, and has the functions of delaying senility and enhancing immunity. Has effects of resisting oxidation, resisting allergy, relieving fatigue, improving body condition, improving sub-health state, delaying aging, improving dysphoria, irascibility, dizziness, debilitation, hypomnesis, etc.
The olive leaves contain triterpenes, flavonoids, iridoids, free alcohol, sterols, alkanes, chalcones and other components, and the olive leaf extract mainly comprises oleuropein, hydroxytyrosol and other effective components, wherein the oleuropein is a nontoxic secoiridoid glycoside compound. The study showed that. Oleuropein has antifungal, antiinflammatory, antiviral, antioxidant, anticancer, and blood glucose reducing effects. Oleuropein is also the component with the strongest biological activity, has strong antioxidant activity in vitro, and the strong antioxidant activity comes from the structure of adjacent dihydroxy substitution, and can be combined with superoxide anion, hydrogen peroxide, nitric oxide free radical and nitro free radical to eliminate free radical. Hydroxytyrosol has a catechol structure, is a typical antioxidant stress substance and free radical scavenger, and has an ORAC antioxidant adsorption free radical energy value which is more than 10 times higher than that of vitamin C and more than 4-6 times higher than that of anthocyanin according to an authoritative data of a Dalelix laboratory in Boston, mass.
Glabridin has antioxidant and antiaging effects, is mainly present at root and stem of Glycyrrhiza glabra Linne, has strong ability of scavenging free radicals, and has skin whitening effect, but easily deteriorates in high temperature environment. In addition, the glabridin also has a certain anti-inflammatory effect, and has anti-inflammatory and whitening effects on skin pigmentation after the glabridin is used persistently.
The retinol can help the skin stratum corneum to restore normal metabolism, thicken the stratum corneum of the skin, enhance the water storage capacity of the skin and promote the generation of collagen in the dermis of the skin; has effects in refining pores, resisting aging, removing wrinkle, removing acne, and whitening skin.
Ascorbic acid and its derivatives are highly effective antioxidants, and because ascorbic acid is easily oxidized to produce dehydroascorbic acid when exposed to air or aqueous solutions, more stable ascorbic acid derivatives such as magnesium ascorbyl phosphate, ascorbyl tetraisopalmitate, ascorbyl ethyl ether, etc. are commonly used in cosmetics to generate ascorbic acid after enzymatic hydrolysis in skin, which plays a role in assisting in synthesizing collagenase, photoprotecting, inhibiting tyrosinase, and resisting photoaging.
Glycerol glucosides are glycoside compounds formed by connecting glycerol molecules and glucose molecules through glycosidic bonds, wherein the structure with important physiological functions is 2-alpha configuration. The glycerol glucoside is an emergency product of plants under extreme conditions, is favorable for protecting the integrity of cell membranes under the extreme conditions, and can promote the expression of skin aquaporin AQP3, enhance the moisture retention capacity of skin, promote the growth of skin cells, accelerate cell repair and metabolism, resist inflammation and the like in skin care products.
Glucopyranoside is a polycyclic structure with the following structural formula:
Figure BDA0003909721550000041
the dextran refers to homotype polysaccharide composed of glucose as monosaccharide, and glucose units are connected by glycosidic bonds.
The sodium hyaluronate belongs to a high molecular polymer and has strong lubricating feeling and film forming property, so that the skin care product containing the sodium hyaluronate has obvious lubricating feeling and good hand feeling when being smeared. The macromolecular sodium hyaluronate forms a layer of breathable film on the surface of the skin, so that the skin is smooth and moist, and the invasion of external bacteria, dust and the like is blocked; sodium hyaluronate with small molecular weight can permeate into dermis, and has effects of slightly dilating capillary, increasing blood circulation, improving metabolism, promoting skin nutrition absorption and waste excretion, preventing skin aging, and caring skin.
Panthenol, also known as panthenol, is a vitamin B5, colorless to yellowish, clear, viscous liquid. Panthenol has effects of preventing pachylosis and aging. Also has obvious anti-inflammatory effect, can delay the progress of inflammatory reaction, and is an ideal auxiliary anti-acne component for sensitive muscles or varicella muscles.

Still another object of the present invention is to provide the use of the anti-aging composition in radio frequency cosmetic and medical cosmetic skin care.
When the skin care product is used singly, the active ingredients of the skin care product are usually blocked by the skin barrier of a human body, most of the active ingredients stay in the horny layer of the epidermis and hardly permeate through the epidermis layer, or the concentration of the active ingredients permeating into the dermis layer of the skin is too low, so that fibroblasts are not stimulated to generate more collagen, elastic fibers and extracellular matrix, the thickness of the dermis layer is not changed obviously, and the anti-oxidation and anti-aging effects are very limited. The anti-aging composition is applied to radio frequency beauty and skin care, energy generated by a radio frequency instrument stimulates a dermis layer to generate more extracellular matrix, and meanwhile, the hydration degree of the epidermis layer can be accelerated, the osmotic pressure of a contact surface is increased, and the transdermal absorption of active ingredients of cosmetics is promoted.
Compared with the prior art, the invention has the following beneficial effects:
(1) The composite polypeptide selected by the invention blocks a muscle fiber sodium ion inflow channel Nav1.4, so that the sodium ion inflow is blocked, the calcium ion inflow is regulated, the muscle operation potential cannot be formed, and further the muscle contraction is weakened, meanwhile, the N-terminal end of the SANP-25 is simulated, and participates in competition of SNAP-25 at the site of a vacuole complex, so that the formation of the complex is influenced, the conduction path of neurotransmitter is finally inhibited/blocked, and further, dynamic expression lines are prevented and removed, and the depth and density of wrinkles are reduced.
(2) According to the invention, the anti-wrinkle composition is obtained by compounding the antioxidant, the composite polypeptide, the panthenol and the glycoside compound in a specific ratio, so that the anti-wrinkle performance of the composition can be obviously improved, and the cosmetics prepared from the anti-wrinkle composition have the effects of resisting oxidation, preserving moisture for a long time, stretching wrinkles and the like.

Determination of DPPH radical scavenging Capacity:
the testing steps are as follows: a certain amount of DPPH is weighed and prepared into 0.1mg/mL DPPH solution by using absolute ethyl alcohol. 2mL of the anti-aging compositions of examples 2 to 11 and comparative examples 1 to 37 were added to 1000g of a sample solution diluted with absolute ethanol and 2mL of a DPPH solution, mixed uniformly, reacted at room temperature for 30min, and then the absorbance Ai was measured at 517 nm. The absorbance A0 of 2mL of DPPH solution +2mL of absolute ethanol and the absorbance Aj of 2mL of sample solution +2mL of absolute ethanol after mixing were measured at the same time. The assay was repeated 2 times. The DPPH radical clearance rate of the sample is calculated by the following formula: the results are shown in Table 1.
DPPH radical scavenging Rate = (A0-Aj + Ai)/A0X 100%
The evaluation method comprises the following steps: the antioxidant capacity can be reflected by the measurement of DPPH free radical scavenging capacity. The higher the DPPH free radical clearance rate, the stronger the antioxidant capacity, and vice versa.

As can be seen from Table 3, by comparing examples 12-21 with comparative examples 56-73, it can be seen that the combination of the antioxidant, the complex polypeptide, the panthenol and the glycoside compound in a specific ratio has a synergistic effect on wrinkle removal of the essence. Meanwhile, the compounding of the antioxidant, the compound polypeptide, the panthenol and the glycoside compound which are outside the specific proportion does not have obvious synergistic effect on the repair and the anti-aging of the essence. The antioxidant, the composite polypeptide, the panthenol and the glycoside compound in a specific ratio are mixed with other moisturizers in the invention, and the obtained anti-aging composition also has a synergistic wrinkle removing effect consistent with the essence prepared in examples 12-21.
The change of skin wrinkles can be known by comparing the use of the facial mask liquid and the radio frequency cosmetic instrument, only using the facial mask liquid and only using the radio frequency cosmetic instrument: when only the radio frequency beauty instrument is used, the skin wrinkles are slightly reduced, which shows that the radio frequency beauty instrument can reduce the wrinkles; when only facial mask liquid is used, skin wrinkles are slightly reduced; when the facial mask liquid is matched with a radio frequency beauty instrument for use, the skin wrinkle reducing effect is greatly increased, so that the cosmetic prepared from the anti-aging composition can promote the regeneration of dermal collagen of skin and fill and fade fine lines by matching the heat effect of the radio frequency beauty instrument, and achieves the effects of deep anti-aging, long-acting moisture retention, wrinkle resistance and skin nourishing.
And (3) testing the irritation:
