随着养殖业的快速发展,从上世纪五十年代起,养殖企业为增加动物生产量,在动物饲粮内加入了抗生素,并对整个养殖行业的发展带来了巨大贡献。但随着时间的推移,抗生素使用越来越频繁,危害日益显现,并引起了社会各界人士高度关注。目前饲料安全等同于食品安全的概念在世界范围内己成为共识。研究发现,发酵饲料可抑制消化道内致病菌的生长,对动物疾病起到一定的预防作用,同时,它还具有适口性好、营养吸收利用率高、可替代抗生素使用等优点。
微生物发酵饲料(Fermented feed)是指在人为可控制的条件下,以植物性农副产品为主要原料,经过有益微生物的代谢作用,降解部分多糖、蛋白质和肪等大分子物质,生成有机酸、可溶性多肽等易于消化吸收的小分子物质,形成适口性好、营养丰富、有效益生菌含量高的生物饲料,其有效成分主要包括极易吸收的有益微生物菌体蛋白、各种生物酶类、生物活性寡肤、氨基酸、有机酸、活性益生菌等
目前用于发酵饲料生产的微生物主要包括:芽孢杆菌,酵母菌,曲霉类真菌以及乳酸菌。乳酸菌是在发酵饲料中一方面能产生抗菌素抑制和杀灭有害菌使其不能生长繁殖,减少或杀灭有害物质使内毒素含量降低从而增强使原宿主抗异能力增强在竞争中占优势,另一方面可以提高饲料的营养价值,将糖类分解改善物料风味和适口性。然而,现有的乳酸菌中兼具产酸能力和抑菌特性的菌株较少,加上饲料发酵通常为混菌体系,外源引入的乳酸菌易打破发酵饲料中的混合微生物平衡,使发酵饲料的营养价值提高程度有限、保质期不长。
发明内容
本发明的第一个目的是提供一种植物乳杆菌(Lactobacillus plantarum)JUN-DY-6,已于2017年3月23日保藏于中国典型培养物保藏中心,保藏编号为CCTCC M 2017138,保藏地址为保藏地址为中国,武汉,武汉大学。
本发明的第二个目的是提供所述植物乳杆菌的培养方法,所述方法是接种于MRS培养基中,30~37℃培养24~48h。
本发明的第三个目的是提供一种抑菌方法,所述方法是将所述植物乳杆菌接种至含致病菌的液体、半固体或固体中,所述杂菌包括大肠杆菌、金黄色葡萄球菌或沙门氏菌中的至少一种。
本发明的第四个目的是提供一种饲料,所述饲料含有所述植物乳杆菌和饲料载体。
在本发明的一种实施方式中,所述饲料载体包括豆粕、豆饼、淀粉、糖蜜中的至少一种。
本发明的第五个目的是提供所述饲料的制备方法,所述方法是将所述植物乳杆菌接种至饲料载体中,于30~37℃静置发酵5~7d。
本发明的第六个目的是提供一种延长发酵饲料保质期的方法,是将所述的植物乳杆菌JUN-DY-6添加至饲料载体中,进行发酵。
本发明的第七个目的是提供所述植物乳杆菌的应用。
在本发明的一种实施方式中,所述应用包括制备食品、药品、保健品。
有益效果:1、本发明的植物乳杆菌JUN-DY-6酸谱广,产酸量高,生长迅速可以豆粕为原料生产多种有机酸。且在37℃条件下发酵28h总酸量可达到14.82g/L;2、应用本发明的菌株发酵豆粕,可使发酵豆粕中大分子蛋白基本降解殆尽,可有效减少发酵豆粕中抗营养因子的含量,降低腹泻的发生率,提高营养物质的利用水平;3、应用本发明的菌株制备的饲料利于储存,可使发酵豆粕湿料在常温条件下保存40~60天,比未添加的饲料保质期延长了2~3倍,且没有霉变现象发生。

生物材料保藏
一株植物乳杆菌(Lactobacillus plantarum)JUN-DY-6,已于2017年3月23日保藏于中国典型培养物保藏中心,保藏编号为CCTCC M 2017138,保藏地址为保藏地址为中国,武汉,武汉大学。
具体实施方式
有机酸含量的测定:使用日立高效液相色谱仪,紫外吸收检测器,分析柱用有机酸柱(Aninex Hpx-87H ion exchange column),柱温50℃,流动相5mM硫酸溶液,流速0.6mL/min,进样量10μL,检测时间27min,计算样品中各种有机酸的含量。
实施例1
1:为达到较好的分离效果,按接种量10g/100mL将发酵豆粕接入MRS液体培养基中,30℃,200rpm培养48h,连续增殖培养2~3次。
MRS培养基配方如下:蛋白胨10.0g,牛肉浸取物10.0g,酵母提取液5.0g,葡萄糖5.0g,乙酸钠5.0g,柠檬酸二胺2.0g,吐温-80 1.0g,磷酸氢二钾2.0g,七水硫酸镁0.2g,七水硫酸锰0.05g,碳酸钙20.0g,琼脂20.0g,蒸馏水定容至1.0L,pH 6.8。
2:菌种的分离纯化:以无菌生理盐水逐级稀释样品,涂布于已经制备好的分离培养基(含2%的碳酸钙)平板上,其上再覆盖一层同样的培养基,夹层厌氧培养(30℃,48h),然后挑选溶钙圈大的单菌落,在分离培养基上多次划线分离纯化得到纯培养物并命名为JUN-DY-6。
3:菌株的16SrDNA鉴定
用细菌基因组DNA快速抽提试剂盒(生工生物)提取菌株JUN-DY-6基因组DNA,作为PCR扩增模板,利用165rDNA引物对(引物27F和引物1495),进行PCR扩增。
PCR反应过程如下:95℃变性5min;95℃/30s,55℃/30s,72℃/2min,35个循环,72℃保持5min。将PCR产物经电泳分析得到约1500bp处较亮的条带,用Q-quick GelExtraction Kit(Promeg公司)进行切胶回收纯化,PCR纯化产物送生工生物公司测序。将测序后的菌株与Genbank数据库的序列进行同源性比对,得出菌株JUN-DY-6为植物乳杆菌(Lactobacillus plantarum)。
实施例2
1、指示菌菌悬液的制备:大肠杆菌、沙门氏菌和金黄色葡萄球菌等致病菌甘油管接种于培养基中,在37℃摇床200r/min培养16-18小时,菌浓达1.2×1010CFU/mL。
2、乳酸杆菌菌悬液的制备:在无菌条件将纯种的植物乳杆菌接种于100mL MRS液体培养基中,37℃静置培养24~48h,菌浓达1.5×1010CFU/mL。
3、双层平板的制备取直径约90mm的平板,分别倾注加热融化的营养琼脂培养基18~20mL,使其在平板内均匀摊布,放置水平台面上使凝固,作为底层。另取半固体营养琼脂培养基(琼脂含量为1%)适量加热融化后,放冷至48-50℃,每50~100mL培养基加入指示菌菌悬液0.1~0.2mL,在每1个平板中分别加入5mL,使其在底层上均匀摊布,作为菌层。放置在水平台面上冷却后,在每1平板中以等距离均匀安置牛津杯4个备用。
其中营养琼脂培养基配方如下:胰蛋白胨10g,酵母提取物5g,NaCl 10g用去离子水定容至1L.固体培养基需添加15~20g琼脂,半固体培养基需添加10g琼脂。
4、抑菌性能评价:取步骤2发酵液上清,每个双层平板中的牛津杯中分别滴装200μL,37℃培养18h后,测量各抑菌圈直径(或面积)以作出评价。选取典型平皿拍照片。
抑菌性:抑菌圈<10mm为抑菌效果微弱,10mm<抑菌圈<15mm的为中度抑菌,抑菌圈>15mm的为高度抑菌。
备注:1)抑菌圈越大说明抑菌效果越好;2)抑菌圈是指圆形透明圈的整个直径。

如表1-2所示,菌株JUN-DY-6对大肠杆菌、金黄色葡萄球菌和沙门氏菌明显的抑菌作用,抑菌圈直径达12~22mm,12.77~15.9mm,12.4~16.3mm,平均值为18.0mm、14.8mm、13.9mm,标准差小于0.1。
实施例3
参照《伯杰细菌鉴定手册》和《乳酸细菌分类鉴定及实验方法》,并对菌株JUN-DY-6进行16sRNA测序。其中菌株生理生化实验结果如表4所示,碳源发酵试验结果如表3所示,结合生理生化及碳源发酵实验结果,并将16S信息进行比对。根据细胞显微形态、生理生化数据和16S rRNA基因序列数据,将菌株鉴定为植物乳杆菌(Lactobacillus plantarum)。

实施例4
利用植物乳杆菌(Lactobacillus plantarum)JUN-DY-6生产发酵豆粕,具体步骤如下:
将风干的豆粕100g,水50g,糖蜜3g及3~10mL对数期的植物乳杆菌菌液(OD 7~8)混合均匀后置于经无菌处理的自封袋中,封口置于37℃培养箱中发酵48h。每四小时取一次样。每批次实验做三个平行。每次取样取4g发酵豆粕于50ml离心管中,加入40ml去离子水搅拌混匀静置1h后取上清,经0.22μm滤膜过滤后置于液相瓶中用于高效液相色谱分析,只要分析样品中有机酸的种类以及含量。发酵结束后称取3g豆粕于100ml烧杯中,加入30mL,0.2%NaOH室温下搅拌15min,于6000r/min离心7min,弃脂层和沉淀,小心留取上清液2mL,稀释5倍留样。取30μL经稀释的样品上清液,加10μL 4×buffer,沸水浴10min,13000rpm离心5min用上清上样,进行SDS-PAGE凝胶电泳分析发酵豆粕中大分子物质的降解情况。结果如图3所示,SM为粗豆粕中的蛋白含量,0%为添加植物乳杆菌的发酵豆粕中的蛋白含量。由图可知,植物乳杆菌用于发酵豆粕的生产可明显降解其中的大分子物质。
实施例5
利用植物乳杆菌(Lactobacillus plantarum)JUN-DY-6并添加碱性蛋白酶生产发酵豆粕:
将风干的豆粕100g,水50g,糖蜜3g,碱性蛋白酶0.5g及3~10mL对数期的植物乳杆菌菌液(OD 7~8)混合均匀后置于经无菌处理的自封袋中,封口置于37℃培养箱中发酵48h。每四小时取一次样。每批次实验做三个平行。取样及处理方式同上。
表5为不同发酵工艺生产发酵豆粕的有机酸种类及最高含量(g/L)
注:G1表示只添加植物乳杆菌(Lactobacillus plantarum)JUN-DY-6组;G2表示添加植物乳杆菌(Lactobacillus plantarum)JUN-DY-6及碱性蛋白酶组。均为28h左右时的产酸值。
由表中可知,用植物乳杆菌(Lactobacillus plantarum)JUN-DY-6生产发酵豆粕,48h内该菌即可大量繁殖迅速产酸,且酸谱广产酸量高。当与碱性蛋白酶配合使用时其最高产产酸量可达到14.82g/L。经过SDS-PAGE凝胶电泳(图3)发现,添加植物乳杆菌JUN-DY-6可将豆粕中的蛋白降解约50%,而该菌与少量蛋白酶配合使用时生产发酵豆粕不仅产酸量高,且大分子蛋白基本降解殆尽。
实施例5
利用植物乳杆菌(Lactobacillus plantarum)JUN-DY-6并添加碱性蛋白酶生产发酵豆粕的储藏性实验:
将风干的豆粕100g,水50g,糖蜜3g,碱性蛋白酶0.5g(碱性蛋白酶的酶活单位为15000~20000U/g)及3~10mL对数期的植物乳杆菌菌液(OD 7~8)混合均匀后置于经无菌处理的自封袋中,封口置于37℃培养箱中发酵48h。以未加入植物乳杆菌的发酵豆粕为对照。
发酵完成后将发酵豆粕湿料置于室温中储藏,定期观察其是否发生霉变(即出现霉点)。结果显示经过该植物乳杆菌JUN-DY-6并添加碱性蛋白酶的生产的发酵豆粕湿料可在室温下保存40~60天不发生霉变。而对照组(除不添加植物乳杆菌以外其他配料均相同)湿料常温下只能保存约20天。用商业化的植物乳杆菌进行发酵豆粕的生产,其湿料的储藏期在30天左右。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。
Claims (10)
1.一种植物乳杆菌(Lactobacillus plantarum)JNU-DY-6,已于2017年3月23日保藏于中国典型培养物保藏中心,保藏编号为CCTCC NO:M 2017138,保藏地址为保藏地址为中国,武汉,武汉大学。
2.一种用于非疾病诊断与治疗用途的抑制致病菌的方法,其特征在于,将权利要求1所述的植物乳杆菌JNU-DY-6接种至含致病菌的液体、半固体或固体中。
3.根据权利要求2所述的方法,其特征在于,所述致病菌包括大肠杆菌、金黄色葡萄球菌或沙门氏菌中的至少一种。
4.一种饲料,其特征在于,所述饲料含有权利要求1所述的植物乳杆菌JNU-DY-6和饲料载体。
5.根据权利要求4所述的饲料,其特征在于,所述饲料载体包括豆粕、豆饼、淀粉、糖蜜中的至少一种。
6.一种制备权利要求5所述饲料的方法,其特征在于,将所述植物乳杆菌接种至饲料载体中,于30~37℃静置发酵5~7d。
7.根据权利要求6所述的方法,其特征在于,发酵前还添加蛋白酶;蛋白酶的添加量为75~100U/g饲料载体。
8.一种延长发酵饲料保质期的方法,其特征在于,将权利要求1所述的植物乳杆菌JNU-DY-6添加至饲料载体中,进行发酵。
9.权利要求1所述植物乳杆菌JNU-DY-6在食品领域的应用。
10.含有权利要求1所述植物乳杆菌JNU-DY-6的食品。
