Abstract

"POLIPEPTìDEOS IMPLICADOS NA BIOSSìNTESE DAS ESPIRAMICINAS, SEQuêNCIAS NUCLEOTìDICAS CODIFICANDO ESSES POLIPEPTìDEOS E SUAS APLICAçõES". A presente invenção refere-se ao isolamento e à identificação de novos genes da via de biossíntese das espiramicinas e de novos polipeptídeos implicados nessa biossíntese. A invenção é também relativa a um processo de produção desses polipeptídeos. Ela é também relativa à utilização desses genes com a finalidade de aumentar as taxas de produção e a pureza da espiramicina produzida. A invenção refere-se notadamente a um microorganismo que produz a espiramicina I, mas que não produz a espiramicina II e III e a utilização desse microorganismo. A invenção refere-se também à utilização dos genes da via de biossíntese das espiramicinas para a construção de mutantes que podem levar à síntese de novos antibióticos ou a formas derivadas de espiramicinas. A invenção refere-se ainda às moléculas produzidas graças à expressão desses genes e das composições farmacologicamente ativas de uma molécula produzida graças à expressão desses genes."Polypeptides implicated in the BIOSYNTHESIS OF SPIRAMICINS, NUCLEOTIDIC SEQUENCES CODING THESE POLYPEPTIDES AND THEIR APPLICATIONS". The present invention relates to the isolation and identification of novel spiramycin biosynthesis pathway genes and novel polypeptides implicated in such biosynthesis. The invention also relates to a process for producing such polypeptides. It is also related to the use of these genes for the purpose of increasing production rates and purity of the produced spiramycin. The invention notably relates to a microorganism that produces spiramycin I but does not produce spiramycin II and III and the use of such microorganism. The invention also relates to the use of spiramycin biosynthesis pathway genes for the construction of mutants that may lead to the synthesis of new antibiotics or to spiramycin-derived forms. The invention further relates to molecules produced by the expression of these genes and the pharmacologically active compositions of a molecule produced by the expression of these genes.
