O uso aditivos e secagem em meio pressurizado durante o processo de sintese de suportes e biocata... more O uso aditivos e secagem em meio pressurizado durante o processo de sintese de suportes e biocatalisadores imobilizados e uma area de pesquisa ainda recente, pois, uma das principais funcoes dos mesmos e a formacao de uma matriz ordenada na sintese de sol-gel, que alem de poder proteger as enzimas da inativacao por diversos efeitos durante o processo de imobilizacao, pode ainda aumentar a sua estabilidade operacional (BARBOSA et al ., 2014). O objetivo deste estudo foi avaliar diferentes metodos de imobilizacao de lipase em aerogel nas reacoes de hidrolise e transesterificacao. A lipase de Burkholderia cepacia (BC) ou a lipase de Bacillus sp. ITP-001(B-ITP) foi imobilizada no aerogel obtido a partir do precursor tetraetilortossilicato (C 8 H 20 O 4 Si) modificado com o liquido ionico protico (LIP) pentanoato de N-metilmonoetanolamina (C5). Treated silica xerogel with protic ionic liquid (PIL) and bifunctional agents (glutaraldehyde and... more Treated silica xerogel with protic ionic liquid (PIL) and bifunctional agents (glutaraldehyde and epichlorohydrin) is a novel support strategy used in the effective immobilization of lipase from (LBC) by covalent binding. As biocatalysts with the highest activity recovery yields, LBC immobilized by covalent binding with epichlorohydrin without (203%) and with PIL (250%), was assessed by the following the hydrolysis reaction of olive oil and characterized biochemically (Michaelis⁻Menten constant, optimum pH and temperature, and operational stability). Further, the potential transesterification activity for three substrates: sunflower, soybean, and colza oils, was also determined, achieving a conversion of ethyl esters between 70 and 98%. The supports and the immobilized lipase systems were characterized using Fourier transform infrared spectra (FTIR), scanning electron microscopy (SEM), elemental analysis, and thermogravimetric (TG) analysis.
