Pharmacological properties
PBX vet classification code QM01A – non-steroidal anti-inflammatory and anti-rheumatic veterinary drugs. QM01AE03 – Ketoprofen.
Ketoprofen is a non-steroidal anti-inflammatory drug of the carboxylic acid group, propionic acid derivatives, which has analgesic and antipyretic effects. The inflammatory process damages the cell, activates phospholipase, releases arachidonic acid, which is the substrate of the enzyme systems of cyclooxygenase and lipoxygenase, the activity of which leads to the production of important inflammatory mediators: prostaglandins, thromboxanes and leukotrienes. Ketoprofen acts as a dual inhibitor of inflammation, inhibiting the activity of cyclooxygenase and lipoxygenase, preventing the formation of prostaglandins and leukotrienes. The analgesic effect of ketoprofen is associated with the inhibition of bradykinin, which stimulates the nerve endings of nociceptors (pain receptors) and, thus, provokes pain. In addition to antibradykinin activity, ketoprofen affects the central nervous system, suppressing the perception of pain. However, in horses, ketoprofen neutralizes the effect of endotoxins and counteracts intestinal cramps caused by bradykinin.
Ketoprofen is rapidly absorbed. The peak concentration in blood plasma occurs less than an hour after parenteral administration, bioavailability is almost complete. Ketoprofen binds strongly to plasma proteins, accumulates in exudate and sites of inflammation. Its duration is from one to four hours. Ketoprofen penetrates the synovial fluid and remains there at a higher level than in blood plasma, and with a half-life two to three times higher.
Ketoprofen is metabolized in the liver, 90% is excreted in the urine.
Application
- Cattle: treatment of animals with acute and chronic inflammatory processes of the musculoskeletal system (traumatic edema, arthritis, osteoarthritis, etc.) and respiratory organs, with acute mastitis in cows, to reduce pain during breast edema.
- Pigs: anti-inflammatory, analgesic and antipyretic therapy for metritis-mastitis-agalactia syndrome, treatment of inflammatory and painful conditions in diseases of the musculoskeletal system and respiratory system.
- Horses: treatment of animals with acute and chronic inflammatory processes of the musculoskeletal system, namely: lameness of traumatic origin, arthritis, osteoarthritis, joint injuries (sprains, synovitis), tendinitis, tendovaginitis, pododermatitis, laminitis, reduction of postoperative pain and inflammation . Symptomatic treatment of colic.
Dosage
- Cattle: intravenously or intramuscularly at a dose of 3 ml of the drug per 100 kg of body weight (3 mg of ketoprofen per 1 kg of body weight) daily for 1-3 days.
- Pigs: intramuscularly at a dose of 0.3 ml of the drug per 10 kg of body weight (3 mg of ketoprofen per 1 kg of body weight) daily for 1-3 days.
- Horses: intravenously at a dose of 1 ml of the drug per 45 kg of body weight (2.2 mg of ketoprofen per 1 kg of body weight) daily for 3-5 days.
One injection is enough for symptomatic treatment of colic. If necessary, re-injection is possible only with additional examination of the animal.
