Loop-mediated isothermal amplification (LAMP) is known as a rapid and reliable alternative to conventional single-step or nested PCR for detection of genomic DNA of various pathogens in clinical samples. In this study, LAMP assay was developed for canine parvovirus (CPV) and compared with single-step and nested PCR assays. Out of 50 fecal samples from dogs clinically suspected for CPV infections, 19 were found positive by single-step PCR, 22 by nested PCR and 26 by LAMP. LAMP products were subjected to restriction analysis and sequencing to check their specificity. LAMP assay turned out to be a rapid and fairly reproducible method, did not amplify other common canine pathogens and was more sensitive than nested PCR assay. Therefore, it can be regarded as a highly reliable method for routine field diagnosis of CPV infection. canine parvovirus; nested polymerase chain reaction; loop-mediated isothermal amplification; sensitivity; specificity.

The antimicrobial photodynamic therapy is an alternative method for killing bacterial cells in view of the rising problem of antibiotic resistance microorganisms. The present study examined the effect of a water soluble photosensitizer, Rhodamine 6G (R6G) in stealth liposomes on multidrug resistant Pseudomonas aeruginosa in the presence of visible light. Liposomes were prepared with cholesterol and phospholipids that extracted from hen eggs in a cost effective way and characterized by light microscopy, particle size analyzer, electron microscopy, steady state spectrophotometry and spectrofluorometry. The photoefficacies of R6G in polymer encapsulated liposomes and positively charged liposomes are much higher compared to the free R6G (R6G in water) in terms of singlet oxygen quantum yield. Methicillin Resistant staphylococcus aureus is a common problem at the community level worldwide. India like countries which are not having standard quality control measurements and poor hygiene, are the major sufferer. Hence the protocols should be applied to monitor antibiotic resistance in all media like clinics and waste water for better control. Here in this study samples from our hospital clinical, milk of suspected from bovine mastitis cases and wastewater bodies were surveyed for MRSA by PCR and sequencing. Phylogenetic analysis of mecA gene revealed the strong relationship between clinical, milk and waste water isolates through mobile genetic transfer. So the results exhibiting high proportions of MRSA in all the three categories, urging better city and town planning.
