SIMPLE EXTRACTION OF DNA

1. What is the purpose of using different household reagents in simple extraction of DNA?

a. Detergent

The purpose of detergent in DNA extraction is to break down the cell membranes and

nuclear membranes, disrupting the lipid bilayers that surround the cellular and

nuclear contents. Detergents are essential components of purpose of detergent in dna

extraction protocols as they aid in releasing the DNA from the cellular and nuclear

compartments, allowing subsequent purification and isolation of the genetic material.

b. Salt

The purpose of Salt in DNA extraction is to helps in neutralizing DNA molecules. It makes the DNA
molecules less hydrophilic i.e less soluble in water. Salt also helps in detaching protein molecules from
strands of DNA and makes the extraction process easier.

c. Meat Tenderizer papaya extract

The purpose of Meat tenderizer in DNA extraction is a Meat tenderizer is an enzyme that
further breaks down the protein-DNA bonds and dissolves the DNA.

d. Ice-cold ethanol

Using ice-cold water and ice-cold alcohol will increase your yield of DNA. The cold water

protects the DNA by slowing down enzymes that can break it apart. The cold alcohol helps

the DNA precipitate (solidify and appear) more quickly.
