Scope This method is applicable for Active ingredient in the range of 0.1--30%m/m.
Method IS 4956 : 2020
Principle A solution of anionic detergent containing added methylene blue is shaken in chloroform. The
mixture is titrated with a cationic detergent. The end point is taken when sufficient methylene
blue has been displaced into the aqueous layer to produce phases of equal colour intensity

1. Weigh around 0.9--1 g of sample & make up to 100ml with distilled water.
2. Take 10 ml sample solution in 100 ml cylinder.
3. Add 15 ml chloroform & 25 ml methylene blue reagent (0.005 %) to cylinder
4. Shake well , the chloroform layer (lower) shall be coloured blue or greenish blue
5. Add from burette Benzalkonium Chloride (0.004 M) slowly
6. After each addition stopper the cylinder and shake well and allow to phases separates
7. Initially chloroform layer will be blue or greenish blue, towards the end the colour would
start migrating to the aqueous layer.
