Hepatocellular carcinoma (HCC) is a wan prognosis and is highly resistant to cancer therapy, push... more Hepatocellular carcinoma (HCC) is a wan prognosis and is highly resistant to cancer therapy, pushing the search for safe drugs as an essential issue. Plant-based bioproducts may reduce the negative side effects of cancer treatment. By the present study, we evaluated the ability of a defatted methanol and hexane extracts from Portulaca oleracea to regulate HCC in an animal model. The anticancer activity of Portulaca oleraceae L. (PO) extract was evaluated against HepG2 cell line where IC 50 was 241.79 mg/ml for PO MeOH extract and 122.19 mg/ml for PO Hex extract. Overall, treatment with DEN+ PO Hex 200 mg/kg shows 100% survival rate. Furthermore, it improved the preservation of hepatocyte architecture in animals with small hepatocyte alterations but interspersed with foci of hepatocyte apoptosis as well as liver enzymes ALP (11.77 §4.02 IU/L), AST (114.15 §11.79 IU/L), ALT (22.11 §2.66 IU/L) and TBILR (0.4 §0.20 IU/L). The relative expression of serum TNFa was significantly up-regulated in DEN induced HCC mice to reach (322.44 §11.2) compared to normal mice (0.07 §0.02). Besides the TNF-a in DEN/ PO groups were significantly down-regulated to be (73.98 §2.7), so the relative expression level of tumor markers BCl-2 and AFP. In addition, apoptotic markers BAX and Cas-8 were significantly increased in DEN/ P. oleracea treated mice. Chemical profiling of methanol and hexan extracts using GCÀMS and HPLC analysis revealed that 9,12,15 octadecatrienoic acid, (2-phenyl-1,3-dioxolan-4-yl) methyl ester, mesitylene, 2-Hydroxy-3-[(9E)-9-octadec-enoylox] propyl(9E) octadecenoate, and 2-(phenymethylene) octanal are the major compounds that were identified by GC_MS analysis of PO Hex extract. While, HPLC clarified the presence of pyrogallol, Syringenic acid, Caffeic acid, P-coumaric acid and Cinnamic acid as major phenolic compounds besides Catechin, Luteolin, and rutin as major flavonoid compounds in PO MeOH extract. Taken together, these results provide the first visualization of the action effects of several compounds in PO and represent potential anti HCC therapy.

(family Apiaceae) is widely used as an essential spice due to its health benefits. This study aimed to investigate the constituents of P.crispum aerial parts and evaluate its hepatoprotective activity. Ethyl acetate and n-butanol fractions of P. crispum subjected to different chromatographic separation techniques. The compounds were identified on the basis of spectral data and compared with previously reported data. Also, the methanolic extract and n-butanol fraction were evaluated for in-vivo hepatoprotective activity against carbon tetrachloride (CCl 4) induced hepatotoxicity in mice (10 groups; 8 mice/group). All groups intraperitonially injected by CCl 4 (0.5mg/kg b.w., i.p.) in single dose at 11th day, the treated groups received (100, 200, 300, 400 mg/kg b.w.) of MeOH extract and BuOH fraction separately for 14 days. The results revealed that seven compounds were isolated from EtOAc and n-BuOH fractions which were identified as β-sitosterol (1), quercetin (2), isorhmentin (3), apigenin (4), apigenin-7-O-β-D-glucopyranoside (cosmosiin) (5), diosmetin-7-O-β-Dglucopyranoside (6), apigenin 7-O-β-D-apiofuranosyl (1→2)-β-D glucopyranoside (apiin) (7). On other hand, CCl 4induced hepatotoxicity cleared in severe liver damage, showed elevation of liver enzymes. The MeOH extract and n-BuOH fraction exhibited significant hepatoprotection in improvement liver enzymes, with a propensity to return to near-normal levels. So, P. crispum extracts are promising in-vivo hepatoprotective activity.

In the present study, extracts of two plants-Olea europaea and Syzygium aromaticum-were tested for their phytochemical compositions, cytotoxic effect on hepatocellular carcinoma cells, and chemical profile of the most active fractions. Both plants were extracted with 85% methanol and then fractionated with various solvents. Hemolysis and cytotoxicity effects on normal (Vero) and hepatocellular carcinoma (HepG2) cells were examined. Chemical profiles were studied using GC-MS and chromatographic isolation on silica gel. Phytochemical screening revealed that both plants contain carbohydrates, saponins and flavonoids, while steroids were found only in O. europaea and alkaloids were found only in S. aromaticum. Hemolysis assay showed complete safety of O. europaea, while S. aromaticum exhibited low toxicity. Furthermore, the EtOAc fraction of O. europaea and BuOH fraction of S. aromaticum showed the best IC 50 against HepG2. The GC-MS analysis of EtOAc fraction of O. europaea revealed 15 compounds, the major one being a-linoleic acid methyl ester, and the chromatographic separation of BuOH fraction of S. aromaticum resulted in a pure compound of 3-O-methyl-butyrate-6,7,3¢,4¢-tetrahydroxy-5-methoxy-8-methylflavan. Thus, the study proved that the two title plants have components possessing cytotoxic activity against HepG2 cells.

This study aimed to evaluate the antimicrobial activity of the Phyllanthus emblica methanol extract and its chemical profile. The plant was extracted with 85% methanol and the total phenolic and flavonoid contents as well as antimicrobial activity on some Gram- positive, Gram-negative bacteria and some fungi species were evaluated. Also, GC-MS analysis of plant extract was administered. Phyllanthus emblica methanolic extract showed high total phenolic content (216.41± 2.1 mg GAE/g of dry extract) and total flavonoid content (32.31± 1.2 mg RE/g of dry extract). The antimicrobial activity of extract against some pathogenic microbes proved that the plant extract is effective against all species except Escherichia coli. The highest activity was showed against Aspergillus fumigatus & Syncephalastrum racemosum (fungi) as well as Staphylococcus aureus, Syncephalastrum racemosum & Streptococcus pneumonia (bacteria). GC-MS analysis of Phyllanthus emblica exhibited the presence of thirty-one bioactive compound. The major compounds are 1, 3-Dimethylindole (24.11%), Isopropyl palmitate (23.94%), Gamolenic acid (7.73%) 1, 5-Hexanediol (7.58%), 8, 11-Octadecadienoic acid methyl ester (6.66%), 2-Methylhexadecanoic acid (5.62%) and Linolenic acid (3.49%). Thus, Phyllanthus emblica may be used as a potential oral antibiotic against some pathogenic microbes.

In this study, Total phenolic and flavonoid contents and the antioxidant activity of the methanol extract of Mentha spicata were determined as well as evaluation of the cytotoxic activity of it. Also, the identification of some bioactive compounds in the plant was analysed. Place and Duration of Study: Extraction and antioxidant at Medicinal Chemistry Department, Theodor Bilharz Research Institute, cytotoxic evaluation and GC-Ms analysis at Al-Azhar University, Egypt. Methodology: The antioxidant activity was determined by two methods, DPPH (2, 2-diphenyl-1-picrylhydrazyl) and ABTS (2-2՝azinobis (3-ethylbenzthiazoline-6-sulphonic acid) while, the cytotoxic assay was determined by MTT assay on HepG-2 (Human hepatocellular carcinoma) and MTC-116 (Human colon carcinoma). Regarding to the chemical identification of methanol extract was carried out by GC-MS analysis. Results: The study proved that Mentha spicata has high Total phenolic and flavonoid contents (388.20±2.38mg GAE/gm of extract &204.01±17.93mg RE/gm of extract) respectively. Also, exhibited promising antioxidant activity by DPPH & ABTS (IC 50 =65.13±1.29 µg/ml & 52.31±0.81 Original Research Article Abdel-Hady et al.; EJMP, 26(1): 1-12, 2018; Article no.EJMP.45751 2 µg/ml) respectively. The methanol extract of the plant showed a good cytotoxic effect on HepG2 and HTC-116 (IC 50 =25.2±3.6µg/ml & 62.1±4.9µg/ml) respectively. GC-MS analysis of the methanol extract of Mentha spicata showed 43 oxygenated hydrocarbon compounds. The major ones are Hexadecanoic acid, methylEster (palmitic acid ester) (31.51%) followed by 9,12,15-Octadecatrienoic acid,methyl ester (CAS)(methyl linolenate) (22.10%), 2-Pentadecanone,6,10,14-trimethyl-(CAS)(6.82%), Phytol (6.20%), 9,12-Octadecadienoic acid(Z,Z)-, methyl ester (6.18%), Hexadecanoic acid (palmitic acid) (5.95%) and Methyl stearate (4.49%). Conclusion: Mentha spicata is a potential antioxidant and anticancer agents.

Cyperus rotundus L. commonly called nut grass is a highly potent traditional herbal drug used for... more Cyperus rotundus L. commonly called nut grass is a highly potent traditional herbal drug used for the treatment of a number of ailments. The present study was aimed to evaluate the antimicrobial activity as well as determine the possible bioactive components of Cyperus rotundus rhizomes using GC-MS analysis. In this study, the plant rhizomes were extracted with 85% methanol and quantitative phytochemical screening was determined. Also, antimicrobial activity of the plant was estimated against fungi and bacteria (Gram positive and Gram negative) by using the agar well diffusion method. The chemical compositions of the methanol extract of Cyperus rotundus rhizomes were investigated using Gas chromatography The results exhibited that the plant has antimicrobial activity against the tested species except Proteus vulgaris. Forty four compounds were identified by GC-MS analysis; linolenic acid (21.9%) was found to be major component followed by hexadecanoic acid, 2-methyl-ester(9.1%), γ-curjunene (5.72%), oleic acid (5.45%), 1-(+)-ascorbic acid 2,6 dihexadecanoate (5.39%), larixol (3.76%), limonen-6-ol,pivalate (3.63%), longipinocarvone (3.11%), methyl stearate (3.11%), aristolone (2.84%), sclareol (2.44%), dodecanoic acid (2.29%) and platambin (2.18%). These results have shown that the plant has a significant biological effect, which can be used as an antimicrobial drug against bacteria and fungi.

Also, it has many pharmacological properties. This study aimed to evaluate cytotoxic and antimicrobial activities of the plant extract on murine spleen cells and hepatocellular carcinoma (HepG2 cell line) as well as GC-MS analysis. The seeds were extracted with 85% methanol and tested on murine spleen cells and HepG2 cell line with different concentrations. Also, antimicrobial activity of the extract was studied by mean inhibition zone. The outcomes proved that the cytotoxic activity of the plant on murine spleen cells is very weak (380.94µg/ml) as well as on HepG2, it inhibits the cell viability% to 8.5% after 24 hrs. (IC 50 = 42.5µg/ml). Also, the plant showed good antimicrobial activity in a concentration-dependent manner on fungi and bacteria (Gram positive and Gram negative). GC-MS analysis of bioactive compounds in the plant extract revealed the presence of 11 bioactive compounds. The major bioactive compounds are 9,12-octadecadienoic acid (Z,Z)-, methyl ester (41.24 %), hexadecanoic acid, methyl Ester (27.39 %), 9 octadecenoic acid (Z)-,methyl ester (16.58%), methyl stearate (4.71%), ethyl linoleate (3.92%) and n-hexadecanoic acid (2.34%). According to these finding, Glycine max methanol extract is potential as anticancer and antimicrobial agents that is may be due to the identified bioactive compoundsby GC-MS analysis. Therefore, the phytochemicals of the plant may be responsible for the various pharmacological actions.

Allium kurrat is an edible cultivated vegetable in Egypt. The aim of this survey is to determine total phenolic and flavonoid contents of the methanol extract and certainly obtained fractions from it. Antioxidant activity of these extracts was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl radical), ABTS (2,2-azino-bis-3-ethylbenzothialozine-sulphonic acid) and total antioxidant capacity assays. Also, cytotoxic activity of these extracts was determined against human hepatocellular carcinoma (HepG2) and human colon carcinoma (Caco-2) using Neutral red assay. LC-ESI-MS analysis was carried out for the most active extracts. The results revealed that ethyl acetate fraction has the highest phenolic and flavonoid contents (127.45 ± 0.71 (mg GAE eq./g extract) and 98.55 ± 1.87 (mg RE eq./g extract)) as well as showed the highest antioxidant activity. On the other hand, the butanolic fraction had the highest antitumor activity on HepG2 and Caco-2 cell lines (36.2 µg/ mL and 44.6 µg/mL) respectively. These results proved that the antitumor activity of this plant is correlated with many active ingredients in it. The LC-ESI-MS analysis identified a mixture of phenolic compounds (flavonoid glycosides and phenolic acids). Thus, Allium kurrat contains active ingredients which inhibit the risk of many chronic diseases.

Aims: In the current research, the total phenolic and flavonoid contents as well as the antioxidant activities of P. crispum methanolic extract and its fractions were evaluated. Methodology: The total phenolic content was estimated by Folin-Ciocalteu method and total flavonoid content was tested by aluminum chloride assay. The antioxidant activity was evaluated by 1, 1-diphenyl-2-picryl hydrazyl radical (DPPH) assay, 2, 2ʹ-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) assay (ABTS), and total antioxidant capacity assay. Results: The ethyl acetate fraction derived from the methanolic extract exhibited the highest total phenolic content (121.95±2.15, mg GAE/ g extract) and total flavonoids content (106.45±2.18 mg rutin equivalent / g extract). Conclusion: The results of the current study showed that P. crispum contains an important amount of phenolic compounds and higher antioxidant activities, which serve as a good source of food ingredients and dietary supplements.

This study aimed to determine total phenolic and flavonoid contents as well as evaluate antioxida... more This study aimed to determine total phenolic and flavonoid contents as well as evaluate antioxidant activity of the defatted methanolic extracts of flowers, leaves, stems and certain fractions (dichloromethane, ethyl acetate, n-butanol and water residue) derived from the defatted methanolic extract of Lantana camara and Cucurbita pepo (Squash) leaves besides, the essential oils of L. camara leaves and flowers by using (DPPH, ABTS and TAC assay). The chemical composition of L. camara essential oils was identified by GC-MS analysis. The results exhibited that the defatted methanolic extracts of L. camara (flowers, leaves and stems) have higher phenolic and flavonoid contents as well as antioxidant activity than C. Pepo (Squash) parts. Also, the ethyl acetate fraction of L. camara has the highest phenolic and flavonoid contents as well as the highest antioxidant activity (DPPH; SC 50 = 11.82±0.21 µg/ mL, TAC; 290.96±6.71mg eq. ascorbic acid /g extract, ABTS; 94.03±0.25mm Trolox ® eq. / 100g extract). There is a positive correlation between the total phenolic and flavonoid contents and the antioxidant activity of the tested extracts or fractions. GC-MS analysis of essential oil of L. camara leaves and flowers revealed the presence of 47 compounds in leaves and 40 compounds in flower. The major compounds in the leaf oil were 7(11)-selinen-4α-ol (14.50%) and cedrenol
