Abstract

Persistent biological indicators of exposure to ionizing radiation, particularly nucleic acid indicators, can be employed in determining whether a subject has been exposed to ionizing radiation. Such biological indicators can be indentified via the technique of differential display.

Claims

AMENDED CLAIMS

1. A method of identifying a biological indicator of exposure to ionizing radiation, comprising the steps of: exposing a population of cells to ionizing radiation; using differential display to compare gene expression in the population of cells three weeks after exposure to the ionizing radiation to gene expression in a control population of cells not exposed to the ionizing radiation; and selecting a gene or gene fragment that has an altered level of gene expression m the exposed population of cells as compared to the control population of cells, which level of expression persists for at least three weeks following exposure to the ionizing radiation.
2. A method according to claim 1, additionally comprising a step of sequencing the gene or gene fragment
3. A method according to claim 1, wherein the step of exposing a population of cells to ionizing radiation is a step of exposing a culture of cells m vi tro .
4. A method according to claim 1, wherein the step of exposing a population of cells to ionizing radiation is a step of exposing cells m vivo .
5. A method according to claim 1, wherein the gene or gene fragment corresponds to a gene that encodes a serum amyloid.
6. A method according to claim 5, wherein the serum amyloid is murine serum amyloid A3.
7. A method according to claim 5, wherein the serum amyloid is human serum amyloid Al or A2. 25. A kit for detecting past exposure to ionizing radiation, comprising: primers specific for a gene or gene fragment which, upon exposure to ioninzing radiation, exhibits an altered level of expression that persists for at least three weeks following exposure to ionizing radiation; reagents for RT-PCR analysis; and instructions for using the primers and the reagents in an assay to determine whether cells removed from a subject contain the gene or gene fragment .
26. A kit as claimed in claim 25, wherein the cells are bone marrow cells.
27. A kit as claimed in claim 26, wherein the primers are specific to an SAA gene or fragment thereof .

In response to the International Search Report, mailed on July 24, 1997, the applicants have amended the claims by replacing present pages 25 and 28 with substitute pages 25 and 28 submitted herewith.
Claim 1 and claim 25 have been replaced with new claims 1 and 25. Claims 2-24 and claims 26-27 are as filed. Accordingly, claims 1-27 are presented for international preliminary examination.
