Description
[0001] 本出願は、2013年12月20日に出願された米国仮特許出願第61/919,026号明細書の出願日の利益を主張し、上記特許出願の開示は参照により本明細書に組み込まれる。
[0002] 本明細書で開示されているのは、1種または複数種の微生物細胞から1種または複数種の多価不飽和脂肪酸(PUFA)を含む微生物油を入手するための方法であって、細胞を溶解して溶解細胞組成物を形成し、次いでこの溶解細胞組成物から油を回収することによる方法である。本明細書でさらに開示されているのは、本明細書に記載した少なくとも1つの方法により微生物細胞から回収される、1種または複数種のPUFAを含む微生物油である。

[実施例1]
[00114] 微生物細胞(シゾキトリウム(Schizochytrium)種)を含む未洗浄の細胞ブロス(500〜700kg)を、60℃で1時間にわたり低温殺菌した。50重量%のNaOH溶液を添加してこのブロスのpHを7.2〜7.5に調整し、ブロスの重量で0.5%のAlcalase(登録商標)2.4 FG(Novozymes(ノースカロライナ州フランクリントン(Franklinton,NC))から入手可能である)を添加し、2時間にわたり55〜60°で撹拌することにより、細胞を溶解した。幅狭ブレードのハイドロホイルインペラにより190〜221RPMで溶解細胞組成物を撹拌し続け、50重量%のNaOHを添加してpHを4に調整し、組成物の重量で2%のNaClを添加し、90℃に加熱することにより、溶解細胞組成物を解乳化した。90℃での数時間後に、解乳化溶解細胞組成物を1750RPMで遠心分離(Seital SR 1010(Seital srl、イタリア)して粗油を生成することにより、この組成物から微生物油を分離し、この粗油から(DHA重量で)76.47%(平均)のDHAを得た。
[00115] 微生物細胞(シゾキトリウム(Schizochytrium)種)を含む未洗浄の細胞ブロス(500〜700kg)を、60℃で1時間にわたり低温殺菌した。ブロスの重量で1.5%の98%硫酸溶液を添加してpHを1.6〜2.1に調整し、幅狭ブレードのハイドロホイルインペラと加熱用の水とにより190〜221RPMで撹拌しつつ90℃に加熱することにより、解乳化溶解細胞組成物を形成した。90℃での数時間後に、50重量%のNaOHを添加してpHを中性(6.5〜8.5)に調整し、解乳化溶解細胞組成物を1750RPMで遠心分離(Seital SR 1010(Seital srl、イタリア)して粗油を生成することにより、この組成物から微生物油を分離し、この粗油から(DHA重量で)84.57%(平均)のDHAを得た。

Claims (31)
- 1種または複数種の微生物細胞から1種または複数種の多価不飽和脂肪酸を含む微生物油を入手するための方法であって、
(a)前記微生物油を含む前記細胞を溶解して溶解細胞組成物を形成する工程;
(b)前記溶解細胞組成物を解乳化して解乳化溶解細胞組成物を形成する工程;
(c)前記解乳化溶解細胞組成物から前記油を分離する工程;および
(d)前記油を回収する工程
を含み、
前記微生物細胞が、トラウストキトリウム(Thraustochytrium)属、シゾキトリウム(Schizochytrium)属またはそれらの混合物に由来し、
(b)が、(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせを含み、
前記(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせが、ハイドロホイルインペラ、ピッチブレードタービンおよびそれらの組み合わせから選択されるインペラによって行われる、方法。 - (a)または(b)の少なくとも一方が、前記細胞または前記溶解細胞組成物を少なくとも60℃に加熱する工程をさらに含む、請求項1に記載の方法。
- (b)が、
(e)前記溶解細胞組成物に塩基を添加する工程;
(f)0.05重量%〜20重量%の量で塩を添加する工程;
(g)少なくとも1種の酵素を添加する工程;および
(h)前記溶解細胞組成物に乳化剤を添加する工程
のうちの少なくとも1の工程をさらに含む、請求項1または2に記載の方法。 - 前記酵素が、β−グルカナーゼ、キシラナーゼ、セルラーゼ、ペクチナーゼ、マンナナーゼ、アミラーゼおよびそれらの組み合わせから選択される、請求項3に記載の方法。
- 前記酵素がβ−グルカナーゼである、請求項3または4に記載の方法。
- 前記酵素が前記溶解細胞組成物の0.05重量%〜10重量%の量で添加される、請求項3〜5のいずれか一項に記載の方法。
- 前記乳化剤がイオン性乳化剤である、請求項3〜6のいずれか一項に記載の方法。
- 前記塩が、アルカリ金属塩、アルカリ土類金属塩、硫酸塩およびそれらの組み合わせからなる群から選択される、請求項3〜7のいずれか一項に記載の方法。
- (b)が、前記溶解細胞組成物のpHを8以上に上昇させる工程をさらに含む、請求項1〜8のいずれか一項に記載の方法。

- 前記油の過酸化物値が5meq/kg未満である、請求項1〜23のいずれか一項に記載の方法。
- (b)が、前記溶解細胞組成物のpHを6以下に低下させる工程をさらに含む、請求項1〜8または10〜24のいずれか一項に記載の方法。
- (b)が、前記溶解細胞組成物の0.5重量%〜20重量%の量で酸を添加する工程をさらに含む、請求項1〜8または10〜25のいずれか一項に記載の方法。
- (a)および(b)が互いに結合されて、1工程の溶解および解乳化する工程を形成する、請求項1〜26のいずれか一項に記載の方法。
- (c)が、前記解乳化溶解細胞組成物のpHを上昇させる工程をさらに含む、請求項1〜27のいずれか一項に記載の方法。
- 1種または複数種の微生物細胞から1種または複数種の多価不飽和脂肪酸を含む微生物油を入手するための方法であって、
(a)前記微生物油を含む前記細胞を溶解して溶解細胞組成物を形成する工程;
(b)前記溶解細胞組成物を解乳化して解乳化溶解細胞組成物を形成する工程;
(c)前記解乳化溶解細胞組成物から前記油を分離する工程;および
(d)前記油を回収する工程
を含み、
前記微生物細胞が、トラウストキトリウム(Thraustochytrium)属、シゾキトリウム(Schizochytrium)属またはそれらの混合物に由来し、
(a)および(b)が互いに結合されて、(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせを含む1工程の溶解および解乳化する工程を形成し、
前記(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせが、ハイドロホイルインペラ、ピッチブレードタービンおよびそれらの組み合わせから選択されるインペラによって行われる、方法。 - 1種または複数種の微生物細胞から1種または複数種の多価不飽和脂肪酸を含む微生物油を入手するための方法であって、
(a)(i)解乳化溶解細胞組成物の形成に適した酵素、(ii)解乳化溶解細胞組成物の形成に適したpHまたは(iii)それらの組み合わせにより、前記微生物油を含む前記細胞を溶解する工程;
(b)前記解乳化溶解細胞組成物から前記油を分離する工程;および
(c)前記油を回収する工程
を含み、
前記微生物細胞が、トラウストキトリウム(Thraustochytrium)属、シゾキトリウム(Schizochytrium)属またはそれらの混合物に由来し、
前記解乳化溶解細胞組成物が、(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせ下での前記溶解中に入手され、
前記(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせが、ハイドロホイルインペラ、ピッチブレードタービンおよびそれらの組み合わせから選択されるインペラによって行われる、方法。 - 前記(i)低剪断撹拌、(ii)軸流撹拌または(iii)それらの組み合わせがハイドロホイルインペラによって行われる、請求項29または30に記載の方法。
