Description
Aは、式:−(CH2)n−R4の基を表し、
R4は、水素原子、アルキル基、アリール基、複素環又は式:−NR5R6の基を表し、
R5及びR6は、同一であるか又は異なっており、水素原子又はアルキル基を表し、
nは、0〜24の整数である]により表されるビスホスホナート誘導体(ただし、R1、R2及びR3が、同時に水素原子を表す、式(I)により表される化合物、並びにR′及びR″が、同時に水素原子を表す、式(I′)により表される化合物を除く)、その光学及び幾何異性体、そのラセミ体、その塩、その水和物並びにその混合物を含むことを特徴とする。
−式(II):ACOXにより表される、少なくとも1つの酸ハロゲン化物、又は式(III):
Aは、上に定義されるものであり、
Xは、ハロゲン原子、好ましくは塩素を表し、
alkは、C1-6アルキル基であり、
xは、2又は3である]により表される、少なくとも1つの亜リン酸シリルとを接触(又は反応)させる工程、
−前工程で得られる化合物を加水分解する工程
を特徴とする。
−式(II):ACOXにより表される、少なくとも1つの酸ハロゲン化物と、式(V):P(OR1)(OR2)(OR)[式中、R1、R2及びRは、同一であるか又は異なっており、アルキル、アリール、アシルオキシアルキル、又は複素環基を表す]により表される、少なくとも1つのホスファイトとを接触させて、式(III)により表される、α−ケトホスホナートを生成させる工程、
−前工程で得られるα−ケトホスホナートと、前記と同義の式(IV)により表される、少なくとも1つの亜リン酸シリルとを接触させる工程、
−前工程で得られる化合物を加水分解する工程
を特徴とする。

1−ヒドロキシ−1−フェニルエチリデン−1,1−ビスホスホン酸の調製
クロロホルム5ml中のトリメチルホスファイト(0.01モル)とジメチルホスファイト(0.01モル)の混合物に、乾燥窒素下で反応混合物を撹拌しながら、0℃でクロロホルム5ml中の塩化フェニルアセチルの溶液を滴下により加えた。次にこの混合物を80℃に約8時間加熱した。
メカニカルスターラー、冷却剤、導入アンプル、温度計及び窒素注入弁を取り付けた、4つ口フラスコに、必要ならば前もって蒸留した、ジメチルホスホナート83g中の濃アンモニア溶液100mlを滴下により加えた。得られた溶液から減圧下で溶媒を留去した。ピリジン(100ml)及びベンゼン(2×100ml)との反復同時蒸発後、得られた白色の固体をヘキサメチルジシラザン140mlで処理した。生じた混合物を6時間還流した。次にこの溶液を蒸留することにより、目的化合物を得た。
酸塩化物1当量を、磁石を取り付けた3つ口フラスコに不活性雰囲気(Ar)下で入れた。トリス(トリメチルシリル)ホスファイト2当量を室温で撹拌しながら加えた。次にこの溶液を室温で数分間静置した。加水分解は、メタノール中で25℃で1時間行った。揮発性画分を真空下で蒸発させた。生成物は、エーテル洗浄により精製した。
メカニカルスターラー、冷却剤、導入アンプル、温度計及び窒素注入弁を取り付けた、4つ口フラスコに、酸塩化物1当量中のトリメチルホスファイト1当量を0℃で滴下により加えた。室温で2時間の反応時間後、α−ケトホスホン酸ジメチルが完全に生成した(反応は、31P NMRにより追跡した)。次にトリス(トリメチルシリル)ホスファイト1当量を滴下により加えた。次いでこの溶液を15分間撹拌した。次にメタノールを加え、この溶液を室温で2時間撹拌した。次いで溶媒及び揮発性画分を蒸発させた。生じた生成物は、白色の粉末であったが、これをエーテルでの連続洗浄により精製した。
メカニカルスターラー、冷却剤、導入アンプル、温度計及び窒素注入弁を取り付けた、4つ口フラスコに、塩化物1当量中のメチル−ビス(トリメチルシリル)ホスファイト2当量を滴下により加えた。室温で10分の反応時間後、溶液を2時間メタノール分解した。次に溶媒及び揮発性画分を蒸発させた。生じた生成物は、白色の粉末であったが、次にこれをエーテルでの連続洗浄により精製した。
メカニカルスターラー、冷却剤、導入アンプル、温度計及び窒素注入弁を取り付けた、4つ口フラスコに、0℃で酸塩化物1当量中のトリメチルホスファイト1当量を滴下により加えた。室温で2時間の反応時間後、α−ケトホスホン酸ジメチルが完全に生成した(反応は、31P NMRにより追跡した)。次にメチル−ビス(トリメチルシリル)ホスファイト1当量を滴下により加えた。次いでこの溶液を15分間撹拌した。次にメタノールを加え、溶液を室温で2時間撹拌した。次いで溶媒及び揮発性画分を蒸発させた。生じた生成物は、無色の油状物であった。これをエーテルでの連続洗浄により精製した。
式が以下に示される、BP1及びBP2と称される2つのビスホスホナートを、細胞及び動物モデルで試験した。BP1は、部分エステル化されておらず、比較例の役割を担う。
材料と方法:
ビスホスホナートBP1及びBP2は、様々な細胞株で試験した:
−FRCjun MRA、腫瘍遺伝子jun−4によるFR3T3細胞のトランスフェクションによって産生された、マウス線維肉腫細胞株、
−A431ヒト癌腫細胞。これらの細胞は、VEGF(血管内皮増殖因子)オートクリンループを収容し、免疫抑制マウスに移植後、脈管形成性の高い腫瘍の急速な発生を誘導する。
−MDA MB435ヒト乳癌細胞、
−HUV−EC−C、形質転換ヒト臍帯内皮細胞(HUVEC)、
−BBC、ウシ脳毛細血管内皮細胞。
ビスホスホナート(0.1〜2mM)は、FRCjunMRA、MDA MB435及びA431細胞の増殖を、時間−及び用量−依存的に阻害した(図2及び3)。例えば、BP2(2mM)は、培養の48時間後、3つの細胞株の増殖を、それぞれ40%、60%及び30%阻害した。BP2(2mM)の存在下で72時間後、A431細胞の増殖は、80%阻害された(P<0.05)(図3)。
腫瘍のない免疫抑制マウスを、注射1回当たり及びマウス1匹当たり0.06〜6mgの範囲の用量でBP1及びBP2で処置した。マウスには、1日に1回の注射を1週間投与した。マウスは、処置の前、間及び後に秤量した。
材料と方法:
A431細胞を、免疫抑制マウスに皮下注射した。マウスには1週間以内に腫瘍が発生した。腫瘍担持マウスは、対照群、又は様々な濃度のBP1及びBP2を投与される処置群にランダムに割り当てた。
V=(4/3)πR12R2[ここで、R1は、半径1であり、そしてR2は、半径2である(ただし、R1<R2)]。
ビスホスホナートでの処置は、A431腫瘍の増殖を阻害し、そして最低用量でさえ最大効果があった(図4)。5週間の処置後、BP1及びBP2(0.6mg/注射/マウス)は、A431腫瘍の増殖をそれぞれ40%及び56%阻害した(P<0.05)。
細胞増殖及び細胞生存度に及ぼすフェニル酢酸ビスホスホン酸ナトリウムの作用の試験は、後述されるように実施した。
フェニル酢酸ビスホスホン酸ナトリウムの毒性の性質を決定するために、MCF7及びMCF7−ras細胞を培養し、次いで24時間後に洗浄し、10%ウシ胎仔血清を含むDMEM培地で3時間培養した。フェニル酢酸ビスホスホナート(10mM)を一方の群に加えたが、もう一方の群には加えなかった。アポトーシスは、FITC抗体と結合させたアネキシンV(Annexine V)の試験により判断した。初期アポトーシス(アネキシンVには陽性染色、かつヨウ化プロピジウムには陰性染色)及び後期アポトーシス(アネキシンV及びヨウ化プロピジウムに陽性染色)を判断するために、ヨウ化プロピジウムを使用した。
フェニル酢酸ビスホスホン酸ナトリウムの抗腫瘍作用をマウスにおいて検証した。
更に、フェニル酢酸ビスホスホナートのアポトーシス促進作用は、抗体染色の方法により確認した。アポトーシス細胞の染色は、上皮細胞により産生されるサイトケラチン18フィラメント上のカスパーゼの特異部位を認識する、M30抗体により行った。このようなフィラメントは、サイトケラチンの高リン酸化により、初期アポトーシスの細胞内で急速に凝集する。この細胞質染色により、顆粒状構造が細胞質内部に観測されたが、これは、後期アポトーシスの細胞に相当する。
抗脈管形成性は、GSL1レクチン法により検証した。
本試験は、急性肝炎を有するヌードマウスで実施した。マウスを2群に分割し、一方は、フェニル酢酸ビスホスホン酸ナトリウムで処置し、2番目の群は処置しなかった。フェニル酢酸ビスホスホナートは、1週間に2回で5週間、処置マウスに皮下投与(80mg/kg又は160mg/kg)した。マウスを屠殺して、HES(ヘマトキシリン・エオシン染色法)後に肝臓を組織化学的に分析した。
Claims (10)
- 下記式(I):
−式(II):ACOXにより表される、少なくとも1つの酸ハロゲン化物、又は式(III):
Aは、上記に定義されるものであり、
Xは、ハロゲン原子を表し、
alkは、C1-6アルキル基であり、
xは、2又は3である]により表される、少なくとも1つの亜リン酸シリルとを接触させる工程、
−前工程で得られる化合物を加水分解する工程
を含むことを特徴とする方法。 - Xが塩素原子であることを特徴とする、請求項1記載の方法。
- アルキル、アリール、複素環又はアシルオキシアルキル基が、アリール基、複素環基、ヘテロシクロアルキル基、アルキル基、アルケニル基、アルキニル基、アルキルチオ基、ハロゲン原子、ヒドロキシル基、NO 2 基、アルコキシ基、式:−COORのエステル基、式:−NRR″′のアミノ基、酸残基、及び式:−CONHR又は−NHCORのアミド基(式中、R及びR″′は、相互に独立に、水素原子、アルキル、アリール、ヘテロアリール又はアシルオキシアルキル基を表す)からなる群から選択される、少なくとも1個の基により置換されていることを特徴とする、請求項1又は2記載の方法。
- 以下の工程:
−式(II):ACOXにより表される、少なくとも1つの酸ハロゲン化物と、式(V):P(OR1)(OR2)(OR)[式中、R1、R2及びRは、同一であるか又は異なっており、アルキル、アリール、アシルオキシアルキル、又は複素環基を表す]により表される、少なくとも1つのホスファイトとを接触させて、式(III)により表される、α−ケトホスホナートを生成させる工程、
−前工程で得られるα−ケトホスホナートと、請求項1に定義される式(IV)により表される、少なくとも1つの亜リン酸シリルとを接触させる工程、
−前工程で得られる化合物を加水分解する工程
を含むことを特徴とする、請求項1〜3のいずれか1項記載の方法。 - 亜リン酸シリルが、式(IV)(ここで、xは、2である)により表される化合物である、請求項4記載の方法。
- 亜リン酸シリルが、式(IV)(ここで、xは、3である)により表される化合物である、請求項4記載の方法。
- 式( II ):ACOXにより表される、少なくとも1つの酸ハロゲン化物を、式( IV )(ここで、xは、2である)により表される、少なくとも1つの亜リン酸シリルと接触させる、請求項1記載の方法。
- 式(II):ACOXにより表される、少なくとも1つの酸ハロゲン化物を、式(IV)(ここで、xは、2である)により表される、少なくとも1つの亜リン酸シリルと接触させ、次にこうして得られる生成物を、式(IV)(ここで、xは、3である)により表される、少なくとも1つの亜リン酸シリルと接触させる、請求項1又は7記載の方法。
- Aが、式:−(CH2)n−R4の基を表し、ここでR4が、C1−C6アルキル基、複素環基、フェニル基、又は式:−NR5R6の基であり、ここでR5及びR6が、同一であるか又は異なっており、水素原子又はC1-6アルキル基を表し、ここでnが、1〜6の整数である、請求項1〜8のいずれか1項記載の方法。
