Description
生体の膀胱洗浄を行い、細胞及び混在物が懸濁されている可能性のある洗浄液を収集する。試料の染色液滴の予備的な顕微鏡的分析で、多数の赤血球細胞と少数の標的細胞の可能性のある細胞の存在を確認する。図3参照。残りの試料を本発明の装置の試料流入口に供給し、濾過する。
腹水試料を得る。腹水を染色し、水滴を顕微鏡で検査すると、非常に多くの上皮細胞と少数の標的細胞の可能性のある細胞が観察される。図5参照。残りの試料の10mLを200mLの試料流入口容器に入れ、本発明の装置を介して濾過した。
末梢血液細胞を含む細胞含有試料を用意する。細胞を公知方法により化学的に溶解する。細胞内容物を本発明に関する装置の試料流入口容器に入れる。
Claims (16)
- 試料の精製及び濃縮のための装置であって、
第1の面と第2の面とを有し、攪拌器を有する濾過ユニットと、
前記濾過ユニットの前記第1の面に選択的流体連通している試料流入口と、
前記濾過ユニットの前記第1の面に選択的流体連通している前記第1の液体リザーバと、
前記濾過ユニットの前記第2の面に選択的流体連通している第2の液体リザーバと、
前記濾過ユニットの前記第2の面に選択的流体連通している第2の吸引手段と、を有し、
第1の吸引リザーバを有し、前記濾過ユニットの前記第1の面に選択的流体連通している第1の吸引手段と、
前記濾過ユニットの前記第2の面に流体連通し、流れ停止能を有する圧力計測器と、を有することを特徴とする、試料の精製及び濃縮のための装置。 - 前記第2の吸引手段は第2の吸引リザーバを有する、請求項1に記載の装置。
- 前記第1の吸引手段と前記第2の吸引手段の少なくとも一方は蠕動ポンプである、請求項1又は2に記載の装置。
- 前記圧力計測器は、前記装置の少なくとも1カ所において流れを停止又は開始させることができるクランプ又は圧電膜の少なくとも一方を有する、請求項1〜3のいずれか1項に記載の装置。
- 前記攪拌器は水平方向の機械的振動を与えるものである、請求項1〜4のいずれか1項に記載の装置。
- 第1の面と第2の面とを有し、攪拌器に接続可能なフィルターを有する濾過ユニットと、
前記濾過ユニットの前記第1の面に流体連通している試料流入口管と、
前記濾過ユニットの前記第1の面に流体連通している第1の液体リザーバ管と、
前記濾過ユニットの前記第1の面に流体連通している第1の吸引リザーバ管と、
前記濾過ユニットの前記第2の面に流体連通している第2の液体リザーバ管と、
前記濾過ユニットの前記第2の面に流体連通している第2の吸引管と、を有する請求項1〜5のいずれか1項に記載の装置に使用されるカートリッジであって、
圧力計測器との少なくとも1つのアクセスポイントを有するカートリッジ。 - 前記濾過ユニットは攪拌器を有する、請求項6に記載のカートリッジ。
- 前記攪拌器は、水平方向の機械的振動を与えるものである、請求項7に記載のカートリッジ。
- 試料を精製する方法であって、
フィルターの第1の面に流体連通している試料流入口に試料を供給する工程と、
所定の圧力が測定されるまで前記フィルターを通して前記試料流入口から流体をフィルターの第2の面へ吸引する工程と、
前記フィルターの前記第2の面を、液体を有するリザーバと専用流体連通させる工程と、
前記フィルターを通して前記リザーバから流体を前記フィルターの前記第1の面へ吸引する工程と、
前記フィルターを通して前記リザーバから吸引された前記流体を容器に回収する工程と、を有し、
上記過程の少なくともかなりの部分で前記フィルターを揺動し、
精製された試料を前記容器中に回収する、試料を精製する方法。 - 前記所定の圧力は、水柱を用いて測定した場合、約130〜150mmH2Oの範囲、好ましくは135〜145mmH2Oである、請求項9に記載の方法。
- 前記所定の圧力は、水柱を用いて測定した場合、約140mmH2Oである、請求項10に記載の方法。
- 前記動揺は、水平方向の機械的振動を与えるものである、請求項9〜11のいずれか1項に記載の方法。
- 請求項9〜12のいずれか1項に記載の方法であって、前記回収工程は少なくとも1回の2段階の流体移送後に行われ、前記2段階の流体移送は、
前記フィルターの前記第1の面を、液体を有する第1の流体リザーバと専用流体連通させる工程と、
前記フィルターを通して前記第1の流体リザーバからの流体をフィルターの前記第2の面に吸引する工程と、
前記フィルターの前記第2の面を、液体を有する第2のリザーバと専用流体連通させる工程と、
前記フィルターを通して第2のリザーバからの流体をフィルターの第1の面に吸引する工程と、
前記フィルターを通して第2のリザーバから吸引された前記流体を容器に回収する工程と、を有する流体移送方法。 - 前記試料は生体試料である、請求項9〜13のいずれか1項に記載の方法。
- 前記試料は循環腫瘍細胞を有する、請求項14に記載の方法。
- 前記試料は所定の圧力により標準化される、請求項9〜15のいずれか1項に記載の方法により精製された試料。
