本発明に係る洗浄ならびに除菌用組成物は、上記したごとく、洗浄作用、グラム陰性菌に対して除菌作用を示すpH11以上の強アルカリ電解水と柑橘類の果実の種子抽出成分、代表的には、食品添加物名簿に記載され、主に食品の日持向上剤として使用されるグレープフルーツ種子抽出物からなっており、一液で優れた洗浄力と各種細菌に対して除菌作用を有していることが極めて大きな特長である。
つまり、この発明に係る洗浄および除菌用組成物は、強アルカリ電解水が有する効果的な洗浄作用およびグラム陰性菌に対する除菌効果と、柑橘類の果実の種子成分が有する効果的なグラム陽性菌に対する除菌効果の相乗効果を有するものであり、本来柑橘類果実の種子抽出成分が有効ではないアルカリ域で特に除菌効果が発揮される点に特長がある。
この発明の洗浄および除菌用組成物は、例えば、大腸菌、黄色ブドウ球菌ばかりではなく、その他のグラム陽性、グラム陰性を含む各種細菌などにも有効である。この発明を実施例により更に詳細に説明する。

The cleaning and disinfecting composition of the present invention is effective not only for Escherichia coli and Staphylococcus aureus, but also for various bacteria including gram positive and gram negative. The present invention will be described in more detail with reference to examples.
下記成分を所定量秤量し、それぞれを混合して洗浄ならびに除菌用組成物を得た。
成 分:
三槽式の電解槽を用い、食塩水の電気分解により生成した強アルカリ電解水pH12.5にグレープフルーツ種子抽出物を濃縮エキス換算で、0.005%、0.01%、0.05%、0.1%、0.5%添加して洗浄ならびに除菌用組成物を得た。

1.除菌試験
上記組成からなる洗浄ならびに除菌用組成物について、大腸菌、黄色ブドウ球菌に対する除菌作用を試験した。
使用した菌は、大腸菌としてEscherichia coli O−157ATCC 35150、黄色ブドウ球菌としてStaphylococcus aureus ATCC 25923を使用した。
各菌をBHIブロスに接種し、35℃で24時間培養して菌液とした。
上記洗浄ならびに除菌用組成物9mlに菌液1mlを加えて25℃を保持し、1分、5分放置した。
同様にコントロールとして滅菌水9mlに菌液1mlを加え、25℃で5分放置した。
細菌数を測定するため、各試験液を滅菌リン酸緩衝生理食塩水で希釈し、シャーレーに1ml添加後、標準寒天培地を20ml加え、混釈培養した。
35℃で48時間培養後、菌数を測定した。
その結果を表1に示す。

Similarly, 1 ml of the bacterial solution was added to 9 ml of sterilized water as a control and left at 25 ° C. for 5 minutes.
In order to measure the number of bacteria, each test solution was diluted with sterile phosphate buffered saline, 1 ml was added to the petri dish, 20 ml of a standard agar medium was added, and the mixture was cultured in pour.
After culturing at 35 ° C. for 48 hours, the number of bacteria was measured.
The results are shown in Table 1.
2.洗浄試験
上記組成からなる洗浄ならびに除菌用組成物について、人工汚垢を用いた洗浄力試験を行った。
人工汚垢は牛脂と大豆油を体積比1:1で混合した油脂20g、モノオレイン 0.25g及びオイルレッド0.1gを同時にクロロホルム60ccに溶解し、その後加熱酸化させたものを使用した。
人工汚垢をスライドガラスに塗布し、80℃で約30分乾燥させて試験板とした。
上記洗浄ならびに除菌用組成物をビーカーに入れ、試験板を浸漬し、超音波洗浄器を用いて15秒間及び30秒間洗浄した。
洗浄前後の試験板の人工汚垢重量を測定し、洗浄時間30秒のpH12.5の強アルカリ電解水の洗浄力を100%として相対評価により洗浄力を求めた。
その結果を表1に示す。

Artificial dirt was applied to a slide glass and dried at 80 ° C. for about 30 minutes to obtain a test plate.
The cleaning and sterilizing composition was placed in a beaker, the test plate was immersed, and washed for 15 seconds and 30 seconds using an ultrasonic cleaner.
The artificial dirt weight of the test plate before and after washing was measured, and the washing power was determined by relative evaluation with the washing power of pH 12.5 strong alkaline electrolyzed water having a washing time of 30 seconds as 100%.
The results are shown in Table 1.
比較例として表2に示す成分割合で調整した比較例1〜4について実施例と同様の試験を行った結果を表2に示す。比較例1は強アルカリ電解水pH12.5のみのもの、比較例2はグレープフルーツ種子抽出物濃縮エキス換算で0.05%のみのもの、比較例3はpH10.9の強アルカリ電解水にグレープフルーツ種子抽出物の濃縮エキス換算で0.05%添加したもの、比較例4は滅菌水である。 Table 2 shows the results of tests similar to the examples for Comparative Examples 1 to 4 adjusted at the component ratios shown in Table 2 as comparative examples. これら実施例と比較例から明らかなように、強アルカリ電解水のpHはpH11以上なければ洗浄力、除菌力ともに不充分である。
pH12.5の強アルカリ電解水は洗浄力、大腸菌に対する除菌力はあるが、黄色ブドウ球菌に対する除菌力が不足している。
またpH11以下のアルカリ電解水にグレープフルーツ種子抽出物を添加すると、アルカリ電解水が酸性分で中和されるため、除菌力は向上するが洗浄力が低下することがわかる。
pH11以上の強アルカリ電解水にグレープフルーツ種子抽出物を添加すると添加量に応じて黄色ブドウ球菌の除菌力が向上し、しかも同時に洗浄力も向上することがわかる。
さらにpH11以上の強アルカリ電解水にグレープフルーツ種子抽出物を濃縮エキス換算で0.005%以上添加すると、洗浄力が強アルカリ電解水単独と比較して6%〜22%上昇し、大腸菌、黄色ブドウ球菌とも5分以内で1×107以上の菌を除菌することが可能となる。
したがって、本発明で規定する範囲内で柑橘類種子抽出物質を添加することで、一液での洗浄、除菌効果が発揮されることが明らかである。

It can be seen that when grapefruit seed extract is added to strong alkaline electrolyzed water having a pH of 11 or more, the sterilizing power of Staphylococcus aureus is improved according to the amount added, and at the same time, the cleaning power is also improved.
Furthermore, when grapefruit seed extract is added to strong alkaline electrolyzed water having a pH of 11 or more in an amount of 0.005% or more in terms of concentrated extract, the detergency is increased by 6% to 22% compared to strong alkaline electrolyzed water alone, and Escherichia coli, yellow grapes It becomes possible to disinfect 1 × 10 7 or more bacteria within 5 minutes with both cocci.
Therefore, it is clear that the addition of the citrus seed extract within the range defined in the present invention exhibits the effect of washing and disinfecting with a single solution.
