Abstract
1,184,422. Antibiotic granaticin B; L-rhodinose. CIBA Ltd. 20 Feb., 1967 [22 Feb., 1966; 26 Jan., 1967], No. 7974/67. Headings C2A and C2C. An antibacterial antibiotic granaticin B is produced by the cultivation of Streptomyces violaceoruber strain 11,382 (NRRL 3262) or a mutant thereof. The antibiotic is isolated from the clarified broth by extraction with a waterimmiscible solvent, e.g. ethyl acetate at pH 4 to 5. The crude is subjected to countercurrent distribution using the solvent system CHCl 3 /CCl 4 / CH 3 OH/H 2 O (55: 55: 75: 25). After dilution with water, active fractions are extracted with chloroform. Further purification is obtained by precipitation from ethyl acetate and petroleum ether. Granaticin B may be converted to an alkanoyl ester by acylation, the alkanoyl ester having not more than 5 carbon atoms in the or each alkanoyl radical e.g. tetra-acetyl granaticin B. Granaticin B is a red powder, empirical formula C 28 H 30 O 12 ; molecular weight 558; infra-red spectrum as in Fig. 2; shows maxima in the ultra-violet at 223 (log # = 4À42); 285 (log # = 3À68); 498 (log # = 3À71); 527 (log # = 3À76) and 566 (log # = 3À57) mÁ (Fig. 1, not shown). On acid hydrolysis, granaticin B produces granaticin and L-rhodinose of formula A pharmaceutical composition comprises granaticin B or an ester thereof, and a carrier, e.g. in the form of a tablet, powder, suppository, solution, suspension or emulsion. The antibiotic may be added to foodstuffs or to animal feeds.
