Abstract

本发明涉及微生物技术领域,具体地说是涉及一新种放线菌。本发明所述的云南链霉菌,其命名为云南链霉菌(streptomyces yunnanensis),其保藏登记号为CGMCCNO.0509。本发明所述的云南链霉菌为一新种放线菌,它能够通过微生物发酵途径、液相色谱制备方法获得具有新旋光类型的环己酰亚胺类化合物,所获得的该类化合物为新化合物;具有强烈的抗真菌活性,利用本发明所述微生物获得的化合物产率高可达到0.04‰。

Description

云南链霉菌
本发明涉及微生物技术领域,具体地说是涉及一新种放线菌。
环己酰亚胺类化合物,具有抗真菌活性,该类化合物主要是通过微生物发酵途径获得,并广泛应用于农用抗生素、实验生化以及医药制造等多个领域,现有的环己酰亚胺类化合物(如放线酮)主要是通过微生物发酵途径获得。随着人类对真菌的认识,需要更多的抗各种真菌的该类化合物。
本发明的目的是提供一种能够产生新的具有抗真菌活性的环己酰亚胺类化合物的新种放线菌——云南链霉菌。
本发明所述的云南链霉菌,其命名为云南链霉菌(streptomycesyunnanensis),其保藏登记号为CGMCC NO.0509。
本发明所述的云南链霉菌系由云南昆明郊区的红土中采集,在实验室分离培养获得,该菌株已在国家知识产权局专利局指定的保藏单位保藏,保藏日期为2000年11月19日,保藏登记号为CGMCC NO.0509。菌株的形态特征为气丝和基丝发达,基丝不断裂;孢子链长、呈螺旋形;孢子(直径0.5-10μm)表面多皱并带短刺。细胞化学:细胞壁含有LL-DAP和甘氨酸:全细胞水解液含半乳糖。
培养条件:
(一)斜面培养:培养7天。培养基:葡萄糖1-5g、酵母膏1-5g、麦芽膏1-5g、复合维生素3.75mg/L、琼脂15-20g、蒸馏水1000ml,PH7.2;
(二)种子培养:从斜面挑取部分菌丝体接入种子液中,摇瓶培养36小时。种子培养基:糊精50-160g、大豆粉10-50g、酵母膏0.5-5g、色氨酸0.1-2g、β-丙氨酸1-6g、硫酸镁0.1-2g、磷酸铵0.1-2g、蒸馏水1000ml,PH7.2:
(三)发酵:按10%的接种量接入种子培养基中,摇瓶培养6天。培养基:大豆粉1-20g、葡萄糖1-25g、蛋白胨1-10、氯化钠0.1-5g、碳酸钙2g、蒸馏水1000ml,PH7.2。

图1显示了本发明所述云南链霉菌菌株孢子链的状态,图2所显示的是菌株孢子链上单个孢子的细致外观,如有皱皮并且具有短刺。
本发明所述的云南链霉菌为一新种放线菌,它能够通过微生物发酵途径、液相色谱制备方法获得具有新旋光类型的环己酰亚胺类化合物,所获得的该类化合物为新化合物;具有强烈的抗真菌活性,利用本发明所述微生物获得的化合物产率高可达到0.04‰。
实施例:
从云南昆明郊区的红土中采集土样,在实验室中按下述条件进行培养,即可获得本发明所述的放线菌菌株云南链霉菌YIM41004。
培养条件:
(一)斜面培养条件:培养基:葡萄糖4g、酵母膏4g、麦芽膏5g、复合维生素3.75mg/L、琼脂15g、蒸馏水1000ml,PH7.2;培养7天;
(二)种子培养条件:从斜面挑取部分菌丝体接入种子液中,摇瓶培养36小时。种子培养基:糊精120g、大豆粉40g、酵母膏2g、色氨酸0.5g、β-丙氨酸5g、硫酸镁0.5g、磷酸铵0.2g、蒸馏水1000ml,PH7.2;
(三)发酵培养条件:按10%的接种量接到种子培养基中,摇瓶培养6天。培养基:大豆粉10g、葡萄糖10g、蛋白胨3g、氯化钠2.5g、碳酸钙2g、蒸馏水1000ml,PH7.2。

Claims (3)

1、一种云南链霉菌,其特征在于命名为云南链霉菌(streptomycesyunnanensis),其保藏登记号为CGMCC NO.0509。
2、按照权利要求1所述的云南链霉菌,其特征在于培养条件为:
(一)斜面培养:培养7天。培养基:葡萄糖1-5g、酵母膏1-5g、麦芽膏1-5g、复合维生素3.75mg/L、琼脂15-20g、蒸馏水1000ml,PH7.2;
(二)种子培养:从斜面挑取部分菌丝体接入种子液中,摇瓶培养36小时。种子培养基:糊精50-160g、大豆粉10-50g、酵母膏0.5-5g、色氨酸0.1-2g、β-丙氨酸1-6g、硫酸镁0.1-2g、磷酸铵0.1-2g、蒸馏水1000ml,PH7.2;
