Insulin or insulin-like growth factor 1 (IGF-1) promotes the activation of phosphoinositide 3 kin... more Insulin or insulin-like growth factor 1 (IGF-1) promotes the activation of phosphoinositide 3 kinase (PI3K)/Akt signaling in immune cells including dendritic cells (DCs), the most potent professional antigen-presenting cells for naive T cells. Klotho, an anti-aging protein, participates in the regulation of the PI3K/Akt signaling, thus the Ca(2+)-dependent migration is reduced in klotho-deficient DCs. The present study explored the effects of insulin/IGF-1 on DC function through klotho expression. To this end, the mouse bone marrow cells were isolated and cultured with GM-CSF to attain bone marrow-derived DCs (BMDCs). Cells were treated with insulin or IGF-1 and followed by stimulating with lipopolysaccharides (LPS). Tumor necrosis factor (TNF)-α formation was examined by enzyme-linked immunosorbent assay (ELISA). Phagocytosis was analyzed by FITC-dextran uptake assay. The expression of klotho was determined by quantitative PCR, immunoprecipitation and western blotting. As a result, treatment of the cells with insulin/IGF-1 resulted in reducing the klotho expression as well as LPS-stimulated TNF-α release and increasing the FITC-dextran uptake but unaltering reactive oxygen species (ROS) production in BMDCs. The effects were abolished by using pharmacological inhibition of PI3K/Akt with LY294002 and paralleled by transfecting DCs with klotho siRNA. In conclusion, the regulation of klotho sensitive DC function by IGF-1 or insulin is mediated through PI3K/Akt signaling pathway in BMDCs.

ABSTRACT High Level Architecture (HLA)-based simulations employing optimistic synchronization all... more ABSTRACT High Level Architecture (HLA)-based simulations employing optimistic synchronization allows federates to process event and to advance simulation time freely at the risk of over-optimistic execution and execution rollbacks. In this paper, an adaptive resource provisioning system is proposed to accelerate optimistic HLA-based simulations in Virtual Execution Environment (VEE). A performance monitor is introduced using a middleware approach to measure the performance of individual federates transparently to the simulation application. Based on the performance measurements, a resource manager distributes the available computational resources to the federates, making them advance simulation time with comparable speeds. Our proposed approach is evaluated using a real-world simulation model with various workload inputs and different parameter settings. The experimental results show that, compared with distributing resources evenly among federates, our proposed approach can accelerate the simulation execution significantly using the same amount of computational resources.

ABSTRACT The purpose of this paper is to examine the impact on Vietnam of sub-regional integratio... more ABSTRACT The purpose of this paper is to examine the impact on Vietnam of sub-regional integration initiatives which have been proposed in the East Asian region. To investigate this issue, we attempt to find out the determinants of Vietnam’s bilateral trade for the period 1992-2006 by individually applying gravity models for exports and imports. Our main results indicate that Vietnam’s bilateral trade flows were positively associated with economic size while the impact of geographical distance appeared to be rather modest. This suggests that other kinds of trade barriers may determine transaction costs in the specific case of Vietnam. Interestingly, our empirical results show that the effective contribution of FDI inflows may be overestimated. Also, the real exchange rate did not play an important role in promoting Vietnam’s exports, while the positive sign associated with its coefficient on the import side is counter-intuitive. More importantly, our empirical results show that Vietnam was constrained by the external balance as its exports and imports reacted differently to the process of trade liberalization within the region. The value of coefficients on the partners’ GDP suggests that Vietnam was more influenced by external demand shocks than supply shocks: hence, any global crisis will affect more demand for Vietnam’s exports than supply of imported commodities. Finally, the intra-regional factor played a significant role in Vietnam’s trade expansion. The deepening of sub-regional integration in East Asia contributed substantially to drive Vietnam’s export growth, at the expense of third markets like EU.

In the search for novel scaffolds leading to potential antihypertensive agents, a series of 2-aryl- and 2-amido-benzothiazoles (HUP) were assessed for their vasorelaxing property on rat aorta rings and for their L-type Ba2+ currents [IBa(L)] blocking activity on single myocytes isolated from the rat tail artery. HUP5 and HUP30, the most potent of the series, inhibited phenylephrine-induced contraction with IC50 values in the range 3–6 mM. The presence of endothelium did not modify their spasmolytic activity. Both HUP5 and HUP30 increased tissue levels of cGMP and shifted to the left the concentration–response curve to sodium nitroprusside.
