ROS measurement is a chal subject for researchers. Due to their high reactivity, ROS are claimed to be involved in many overproduced or when the levels of antioxidants become severely depleted, these reactive species become highly harmful, causi oxidative stress through the oxidation of biomolecules, leading to cellular damage that may become irreversibile and c death. Fluorescent probes are excellent sensors of ROS a method that may be used in future biomedical investigation to measure concentration at a particular moment in time. MWES-1 is designed for the measurement of the cell membrane fluorescence anisotropies (MFA) simultaneously with MW exposure. MWES-2 is designed for the cells culture exploration under an inverted microscope before, during and after MW exposure. MWES-1 consists mainly of a 2.45 GHz microwave generator (MWG-2.45GHz-SAIREM) of 0-25 W, equipped with forward power and reflected power displaying, and an adjustable coaxial antenna immersed directly into the cuvette with the cells-suspension of a Spex type spectrofluorometer. The MW effect on membrane fluidity of B16F10 malignant melanoma (B16F10-MM) cells in suspension were investigated with MWES-1, by MFA measurements. We used Digital Holographic Microscopy (DHM) to compare RI and dry mass density of two B16 murine melanoma sublines of different metastatic potential. Using statistical methods, the phase shifts distribution within the reconstructed quantitative phase images (QPIs) was analyzed by the method of bimodality coefficients. The observed correlation of RI and bimodality profile with the cells metastatic potential was validated by real time impedance based-assay and clonogenic tests. We suggest RI and QPIs histograms bimodality analysis to be developed as optical biomarkers useful in label-free detection and quantitative evaluation of cell metastatic potential.

The chemical profile of hydroalcoholic polyphenolic extracts, prepared either by conventional extraction or microwave-assisted method, was analyzed by reverse-phase high-performance liquid chromatography with photodiode array detector (HPLC-PDA) analysis, while their radical scavenger activity (RSA) was evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl radical) and ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) assays. There are few published data referring to the biochemical alterations of platelet membrane in patients with MPN. The aim of our study was to determine whether changes of platelet membrane fluidity may be correlated with the modified expression of platelet receptors and altered platelet function in MPN. Materials and methods. This retrospective study included 71 patients with MPN: 21 chronic myeloid leukemia (CML), 13 primary myelofibrosis (PM), 24 essential thrombocytemia (ET) and 13 primary polycytemia (PP), as well as 10 healthy volunteers. Even though there are many strategies for controlling the drug release kinetics, these must be adapted through trial and error on a case-by-case basis. Here we explore the possibility of tailoring the release kinetics of hydrophilic, water soluble therapeutic agents from mesoporous silica through addition of a hydrophobic excipient, 1-tetradecanol. In vitro drug release experiments performed at 37 °C, in phosphate buffer solution (pH 7.4) show that the addition of tetradecanol yields slower drug release kinetics, which was correlated with the presence of a liquid fatty alcohol interfacial layer. The layer mass is 11-23 wt.% of the metoprolol-loaded silica sample, and it causes up to 1.6 times decrease of initial release rate with respect to materials without the fatty alcohol. Changes in optical and shape-related characteristics of B16F10 cells after electroporation were i... more Changes in optical and shape-related characteristics of B16F10 cells after electroporation were investigated using digital holographic microscopy (DHM). Bipolar rectangular pulses specific for electrochemotherapy were used. Electroporation was performed in an "off-axis" DHM set-up without using exogenous markers. Two types of cell parameters were monitored seconds and minutes after pulse train application: parameters addressing a specifically defined area of the cell (refractive index and cell height) and global cell parameters (projected area, optical phase shift profile and dry mass). The biphasic behavior of cellular parameters was explained by water and mannitol dynamics through the electropermeabilized cell membrane.

The paper presents two radiation exposure facilities (REFs) which permit separate and simultaneou... more The paper presents two radiation exposure facilities (REFs) which permit separate and simultaneous irradiation with microwaves (MW) of 2.45 GHz and electron beams (EB) of 6.23 MeV for malignant melanoma (MM) cell investigations, in vitro (MW+EB-REF-vitro) and in vivo (MW+EB-REF-vivo). The REFs are specifically designed for the following medical studies: 1) The effects of separate and combined (successive and simultaneous) MW and EB irradiation on the B16F10 mouse--MM cell cultures without/with drugs incubation, 2) The effects of separate and combined MW and EB irradiation on human blood components irradiated in samples of integral blood from healthy donors and from donors with MM; 3) The effects of separate and combined MW and EB whole body irradiation on the C57 BL/6 mice bearing MM without/with drugs administration. Several representative results obtained by experiments with REFs in vitro and in vivo are discussed. The most important conclusion of the experimental results is that low dose-total body MW+EB irradiation combined with drugs administration could present a valuable potential for an advanced study in malignant melanoma therapy.

It also modifies the aggregation/agglutination of freshly prepared platelets from volunteers (Vlasic et al, Life Sci. 1993). Reactive oxygen species (ROS) are involved in integrin αIIbβ3 activation (Begonja et al, Blood Cells, Mol. and Dis. 2006; Clutton et al, Arterioscler. Thromb. Vasc Biol. 2004). Detecting reactive oxygen species (ROS) in living cells is a difficult task because of their shor... more Detecting reactive oxygen species (ROS) in living cells is a difficult task because of their short lifetime, presence of cellular antioxidants, risk of cell death and unwanted transformations of the detector substance. Fluorescence-based techniques are currently most widely used, with high sensitivity and easy processing of samples but the classical instruments need a high amount of sample and consumables. We optimized and validated a microvolumetric fluorescence-based method for ROS detection in living cells, using 2’,7’-dichlorodihydrofluorescein acetate (H2DCFDA) and a microvolumetric fluorospectrometer. In our experimental conditions we determined that a measurement volume of 3 µL containing between 125 and 500 cells was sufficient for a reliable measurement of oxidized 2’,7’-dichlorofluorescein fluorescence.
