Hypericum perforatum L. is a pharmaceutically important medicinal plant with antidepressant, anti... more Hypericum perforatum L. is a pharmaceutically important medicinal plant with antidepressant, antiviral, antibacterial and anticancer effects of its metabolites. The hypericin contents and genetic structures of superior H. perforatum clones were identified by spectrophotometric analysis and random amplified polymorphic DNA (RAPD) markers, respectively. At the first harvest the hypericin content of the clones was higher than second harvest and higher in 2007 compared to 2008. For initial screening, 38 random primers were used and 26 of them detected a total of 378 scorable fragments. Effective multiplex ratio (EMR), average heterozygosity (Hav), marker index (MI), resolving power (RP) and polymorphic information content (PIC) of the primers were calculated for RAPD marker system. Genetic similarity value among the clones varied from 0.40 to 0.93 and the average genetic similarity values was determined as 0.60. Duyarli yerli cesit Izmir 85, Lr13 genini tasiyan yakin izogenik hat (12 No’lu hat) ve bunlar arasinda yapilan melezlemenin F1 generasyonunda ayni markorler kullanilmistir. Incelenen dort SSR markorunden bir tanesi (GWM630) Lr13 genini tasiyan hatta ozgun bir bant deseni vermistir. Bu bant deseni duyarli ebeveyn ve F1 generasyonunda da ayni gozlenmistir. Bu sonuc, duyarli ebeveyn Izmir 85 cesidinin Lr13 genini tasiyabilecegine isaret etmektedir. Tolerance level of seven wheat genotypes to drought conditions in post anthesis st age were evaluated based on some tolerance indices in four y ears field experiments differing in rainfall distri bution and quantity. Six selection indices including Relative Decrease in Yield (RDY), Stress Tolerance Index (TOL), Mean Productivity (MP), Geometric Mean Productivity (GMP), Stress Susceptibility Index (SSI) and Stress Tole rance Index (STI) were calculated based on grain yield under dry and wet conditions. Higher grain yield was r ecorded in Basribey-95 during both dry and wet seasons whereas Menemen-88 had considerably lower yield than other genotypes. The trials were arrange d in a Randomized Complete Block Design with four replications. Additive main effects and multiplicat ive interactions analysis (AMMI-biplot) indicated t hat the yield performances of bread wheat cultivars were highly affected by the major environmental factors. The fir st two principal component axes (PCA 1 and PCA 2) were significant and they explained 60.9 % of the total gen otype x environment interaction. A biplot generated using g enotypic and environmental scores of the first two AMMI components demonstrated that cultivars with larger PCA 1 and lower PCA 2 scores were high yielding and stable cultivars and cultivars with lower PCA 1 and larger PCA 2 scores were low yielding and unstable cultivars in tested locations. Electronic shade-matching instruments can be used to determine objectively the color of cotton fiber. The objectives of this study were to investigate brown to white fiber color segregation in the F2 population of ‘Carmen’ (white fiber) x ‘Devetüyü-176’ (brown fiber) using an electronic shade-matching instrument (chromameter) and compare those results to visual color analysis. The results demonstrated that transgressive segregations were identified for all fiber color parameters in the F2 population. The fiber color parameters (L, ∆L, a, b, ∆a, ∆b, and ∆E) and the visual analysis were considered separately. The objective of this study was to establish the inter-relationship and direct and indirect effect of various quality traits on seed cotton yield in colored cotton genotypes. The F3 and F4 generations of the cross between Carmen (white) and Devetüyü-176 (brown fiber) were used for selection process in terms of seed cotton yield per plant and fiber color. Wild tetraploid wheat (Triticum turgidum L. var. dicoccoides) has some genes for high protein content as a promising source. Three winter wheat cultivars of Colorado (Halt, Yuma, Akron) and one experimental line (CO99508) and one accession of T. dicoccoides were crossed to obtain backcrosses. The objective of this study was to determine higher protein content, SDSsedimentation and hardiness values in backcrosses T. aestivum x T. dicoccoides. Analysis of variance showed that highly significant differences were found among parents and backcrosses. The minimum and maximum values of grain protein content of all backcross populations changed from low protein parents (bread wheat cultivars) to high protein parent (T. dicoccoides). The aim of the present study was to detect candidate DNA markers for leaf rust resistance gene Lr... more The aim of the present study was to detect candidate DNA markers for leaf rust resistance gene Lr13. The NIL groups were constructed from a set of near isogenic lines of wheat cv. Thatcher containing in total 41 different Lr resistance genes. A total number of 246 primer combinations in the NIL groups were screened for DNA polymorphism by using the AFLP method. Twenty-eight AFLP primer combinations yielded 33 polymorphic bands in Tc+Lr13 line. To confirm these polymorphic bands, 28 primer combinations were investigated in 41 Thatcher near isogenic lines. Six primer combinations amplified specific band products in Tc+Lr13. It was concluded that these AFLP markers can be linked to Lr13 gene and in further studies on Lr13 resistance gene, these six AFLP markers should be analyzed in near isogenic lines. Also, construction of the NIL groups can be useful in terms of time and cost of the molecular researches including Thatcher near isogenic lines.

Seventeen hybrid maize genotypes were evaluated at four locations (Yeniehir- Marmora Region; Born... more Seventeen hybrid maize genotypes were evaluated at four locations (Yeniehir- Marmora Region; Bornova-Aegean Region; Ceyhan-Mediterranean Region; Seyhan- Mediterranean Region) in 2005 and 2006 cropping seasons under irrigated conditions in Turkey. The analysis of variance for grain yield of the 17 hybrid genotypes tested in eight environments showed mean squares of environments, genotypes and GEI (genotype x environment interaction) were highly significant and accounted for 62.1%, 12.5% and 25.4% of treatment combination sum of squares, respectively. To determine the effects of GEI on yields, the data were subjected to additive main effects and multiplicative interaction (AMMI) and the GGE biplot analysis. Its damage on leaves, stems, spikes and grains reduce the baking quality of flour made from damaged grains. In this study, some wheat genotypes from Turkey and ICARDA were evaluated for the pest resistance. The genotypes were planted in a randomized block design using hill plots in nylon mesh screening cages in wheat growing season of 2011-2012 and 2012-2013. Sunn pest population was collected from Canakkale province, where the pest was intensely found in recent years. The plants of each hill plots were infested with one male and one female Sunn pest adults. The results with 12.5% sucking damage showed that the genotypes from ICARDA had higher resistance than the landraces from Turkey to Sunn pest.
