yes they look like ES, they also look very similar to mouse ES cells.

Thank you very much for your reply. The cells are human cells. I am trying to reprogram the cells into stem cells. The colones I obtained do not look like typic ES cells or iPS cells (see the picture below). I mean I could hardly see individual cells in my clones although they pile up very high.
What are morphological differences between hESC and mESC?

ah.. ES morphology.. big mistery :-) the only way to tell if you got stem cells or iPS or ES-like is to use markers: RT-PCR and immunostaining, and if these things are positive then try to differentiate them in as many cell type as possible. Morphology can give you a 1st indication but nobody will believe you only based on morphology.
about differences between mouse and human ES cells, I think that there is a lot of variation between different lines. but for what i've seen directly generally to me mouse ESc colonies look more 3d with nice rounded borders, human look a little more flat, and often they have a "polygonal" border. Keep in mind though that this might not mean anything..

I totally agree. I am waiting for an ES cell kit to do some staining.
I picked one clone today and found it is very hard to disperse, there is jelly-like matrix with cells trapped inside. Very strange to me. I was wondering whether it is a EB.
